Book excerpt: In the first 20 years that followed the purinergic signalling hypothesis in 1972, most scientists were sceptical about its validity, largely because ATP was so well established as an intracellular molecule involved in cell biochemistry and it seemed unlikely that such a ubiquitous molecule would act as an extracellular signalling molecule. However, after the receptors for ATP and adenosine were cloned and characterized in the early 1990s and ATP was established as a synaptic transmitter in the brain and sympathetic ganglia, the tide turned. More recently it has become clear that ATP is involved in long-term (trophic) signalling in cell proliferation, differentiation and death, in development and regeneration, as well as in short-term signalling in neurotransmission and secretion. Also, important papers have been published showing the molecular structure of P2X receptors in primitive animals like Amoeba and Schistosoma, as well as green algae. This has led to the recognition of the widespread nature of the purinergic signalling system in most cell types and to a rapid expansion of the field, including studies of the pathophysiology as well as physiology and exploration of the therapeutic potential of purinergic agents. In two books, Geoffrey Burnstock and Alexej Verkhratsky have aimed at drawing together the massive and diverse body of literature on purinergic signalling. The topic of this first book is purinergic signalling in the peripheral and central nervous systems and in the individual senses. In a second book the authors focus on purinergic signalling in non-excitable cells, including those of the airways, kidney, pancreas, endocrine glands and blood vessels. Diseases related to these systems are also considered.

Book excerpt: These volumes record the presentations made at the VIII International Symposium on Purine and Pyrimidine Metabolism in Manheld at Indiana University, Bloomington, USA from May 22- May 27, 1994. This was a continuation of meetings held every three years with the idea of bringing clinicians and basic scientists together, which we hope results in cross-fertilization of ideas. Some of the papers presented in this volume represent oral contributions and others are from posters, but we emphasize that both are considered of equal merit. As is obvious from a perusal of the titles of the papers there has been a shift in the focus of this meeting, which reflects a general shift in the area of purine and pyrimidine metabolism. The emphasis has definitely shifted to gene structure and molecular genetics, with the beginnings we hope of gene therapy as an important branch of this area of science. Although many of the inherited diseases discussed in this text can be treated with drugs, the major thrust in the futurewill be in gene therapy, where the gene (or cDNA) will be used to treat the patient with enzyme deficiency, particularly if the patient is young. As can be seen from the Iist of authors there is a remarkable degree of international cooperation in this area across countries and continents. We thank the many participants who have attended these symposia many times, and we welcome the large group of scientists from Eastern Europe who are attending this meeting for the first time.

Since the first of this series of meetings was held in Israel in 1973, conventions were organized every three years in different parts of the world including the USA, Japan, and Europe. The different aspects of purine and pyrimidine metabolism bring together re searchers working in molecular genetics, biochemical pharmacology, biochemistry, devel opmental biology, immunology, epidemiology and the clinics. Oriented research in the field has been seminal for the development of potent anticancer and antiviral drugs. As the number of genes which are cloned, grows, the understanding of metabolism is increas ingly enlarged and might provide leads to further improve therapeutic concepts and to bet ter understand mechanisms responsible for the development of resistance against these drugs. In certain diseases purine and pyrimidine analogs represent not only the drugs of choice but in fact are the sole therapeutic alternative at present. The field has also taken an early lead in attempting to correct inborn errors of purine and pyrimidine metabolism by gene therapy. The organization of this meeting involved a large number of people who dedicated their time in an effort to make this symposium a success. We thank the Abstract Review Committee, the International Advisory Board and in particular the Symposium Secretariat for doing a wonderful job.

Previous meetings in this series were held in Tel Aviv (Israel), Baden (Austria), Madrid (Spain) and Maastricht (The Netherlands). The proceedings of each of these meetings were published by Plenum. The next meeting will be in Japan. This Symposium differed from those that went before in that it permitted us to honor Dr. J.E. Seegmiller, Professor of Medicine at the University of California San Diego, for his many contributions to our understanding of purine metabolism in man. This publication is dedicated as a Festschrift to Jay. Dr. Richard W.E. Watts delivered the keynote address outlining in scholarly fashion the history of Dr. Seegmiller's accomplishments in research on purine metabolism and the great number of currently active scientists in this field who have worked with him. This address is published as the first contribution to Volume I. Dr. Dewitt Stetten, Jr., was scheduled to be the speaker at our banquet. Unfortunately, he could not be with us. Dr. Seegmiller has written an appreciation of Dr. Stetten and his contributions to our field, and this has been published following Dr. Watts' paper. The growth of knowledge in purine and pyrimidine metabolism continues to be exponential. The variety of subjects included in these volumes is impressive. New or previously unrecognized disorders of purine metabolism continue to be uncovered. An entire section on disorders of purine and pyrimidine metabolism other than deficiency of HPRT is led off by two papers on adenylosuccinase deficiency. Among the disorders of pyrimidine metabolism there are papers on orotic aciduria and dihydrothymine dehydrogenase deficiency. Clinical and biochemical studies of gout and urolithiasis continue to be actively pursued. At the same time the study of purine metabolism has become an integral feature of immunology. The importance of purines in clinical oncology was first demonstrated with the synthesis by George Hitchings of 6-mercaptopurine. Its continuing impact on hematology and oncology is seen throughout these volumes, particularly in the effects of inhibition of adenosine deaminase on T cells and on T cell leukemia. This publication has implications for internal medicine, pediatrics, urology, biochemistry, immunology, genetics, hematology, and oncology. Modern molecular biology and techniques involving recombinant DNA were evident in papers on HPRT and on adenosine deaminase, as well as in studies on APRT and UMP synthase. The genes for HPRT, adenosine deaminase and puine nucleoside phosphorylase have been cloned. The background for ultimate approaches to gene therapy in man was provided in papers from Dr. Seegmiller's laboratory on the insertion of HPRT cDNA into human bone marrow cells and on metabolic cooperation. Purine receptors have been discovered in the central nervous system, in lymphocytes and in a variety of other tissues. There are also adenosine receptors in Leishmania and a number of purine riboside analogs are under study as potential therapeutic agents in leishmaniasis.
