Available in PDF, EPUB and Kindle. Book excerpt: In his thesis, Kiyoun Lee describes his studies into tandem and organocatalytic oxa-conjugate addition reactions for the synthesis of complex tetrahydropyrans (THP). Readers gain insight into the new methods Lee employs for the synthesis of biologically interesting natural products including (+)-leucascandrolide A, (+)-dactylolide, and (±)diospongin A. The reactions Lee investigates are applicable to a broad range of substrates and proceed with excellent stereoselectivity. Moreover, the methodologies allow the synthesis of a wide range of THP-containing compounds. The development of reactions, such as those discussed by Lee, has the potential to impact natural product synthesis, pharmaceutical development and chemical biology.

Book excerpt: Described in this dissertation are several methods developed for the stereoselective synthesis of tetrahydropyrans rings. Initially, an effort to expand the Mukaiyama Aldol-Prins cyclization reaction to disubstituted (E)-enol ethers was explored. The Mukaiyama-Michael cascade reaction was then discovered by a fortuitous result. Investigating this method provided an unexpected tetrahydropyran product containing 2,6-trans geometry, and is rationalized considering both steric and electronic effects. Finally, a two-step [4+2] inverse demand HDA annulation reaction and rearrangement was developed for the diastereoselective synthesis of 2,4,5,6- tetra-substituted tetrahydropyran rings.

Book excerpt: Brings together the best tested and proven stereoselective synthetic methods Both the chemical and pharmaceutical industries are increasingly dependent on stereoselective synthetic methods and strategies for the generation of new chiral drugs and natural products that offer specific 3-D structures. With the publication of Stereoselective Synthesis of Drugs and Natural Products, researchers can turn to this comprehensive two-volume work to guide them through all the core methods for the synthesis of chiral drugs and natural products. Stereoselective Synthesis of Drugs and Natural Products features contributions from an international team of synthetic chemists and pharmaceutical and natural product researchers. These authors have reviewed the tremendous body of literature in the field in order to compile a set of reliable, tested, and proven methods alongside step-by-step guidance. This practical resource not only explores synthetic methodology, but also reaction mechanisms and applications in medicinal chemistry and drug discovery. The publication begins with an introductory chapter covering general principles and methodologies, nomenclature, and strategies of stereoselective synthesis. Next, it is divided into three parts: Part One: General Methods and Strategies Part Two: Stereoselective Synthesis by Bond Formation including C-C bond formation C-H bond formation C-O bond formation C-N bond formation Other C-heteroatom formation and other bond formation Part Three: Methods of Analysis and Chiral Separation References in every chapter serve as a gateway to the literature in the field. With this publication as their guide, chemists involved in the stereoselective synthesis of drugs and natural products now have a single, expertly edited source for all the methods they need.

Book excerpt: Design of new processes that avoid the use of toxic reagents has been the focus of intense research of late. Catalysis by metals and non-metals offers diverse opportunities for the development of new organic reactions with promising range of selectivities—chemoselectivity, regioselectivity, diastereoselectivity, and enantioselectivity. Furthermore, these transformations frequently occur under mild conditions, tolerate a broad array of functional groups, and proceed with high stereoselectivity. The area of catalysis is sometimes referred to as a ‘foundational pillar’ of green chemistry. Catalytic reactions often reduce energy requirements and decrease separations because of increased selectivity; they are also capable of permitting the use of renewable feedstocks of less toxic reagents or minimizing the quantities of reagents needed. New catalytic organic synthesis methodologies have, thus, offered several possibilities for considerable improvement in the eco-compatibility of fine chemical production. Hence, these catalytic methodologies have emerged as powerful tools for the efficient and chemoselective synthesis of heterocyclic molecules. Key Features: Presents the synthesis of different five-membered heterocycles. Contains the most up-to-date information in this fast-moving field. Covers novel catalytic approaches used in the study and application of catalysts in synthetic organic reactions. Presents new methodologies for the synthesis of heterocycles.

The interdisciplinary author team with an excellent reputation within the community discusses experimental and theoretical studies, along with examples of improved catalysts, and their application in organic synthesis, biocatalysis, and supported organometallic catalysis. As a result, readers will gain a deeper understanding of the catalytic transformations, allowing them to adapt the knowledge to their own investigations. With its ideal combination of fundamental and applied research, this is an essential reference for researchers and graduate students both in academic institutions and in the chemical industry. With a foreword by Nobel laureate Roald Hoffmann.
