Book excerpt: One of the central questions in immunology is the understanding in molecular terms of antigen-antibody interactions and of the cellu lar recognition of antigens. It is hoped that this understanding will extend eventually to the immunobiological basis of host defense to infectious agents and of tissue damage or deranged cell functions which stem from these reactions. A variety of natural and artificial substances have been used as models for these studies. Emphasis was placed upon substances of known and relatively uncomplicated chemical structures. These included polysaccharides, amino acid polymers, nu cleic acids and haptens. On the other hand. until recently there has been very little information on protein antigens. The complexity of these molecules posed an immense chemical obstacle to precise immuno chemical analysis. Indeed, it is this difficulty with proteins that spurred the synthesis and immunological studies of amino acid poly mers. The control and normal regulation of the immune system at the cellular-molecular interface and the great majority of antigens asso ciated with immune disorders are attributed to protein molecules. In the last few years great advances have been made in the analysis and synthesis of the antigenic sites of some proteins. The entire antigenic structures of myoglobin and lysozyme and the partial anti genic structures of several other proteins have been determined. Moreover, in the past seven years several biological responses resulting from the reactions of proteins and their peptides with cells of the immune system were described.

Book excerpt: The discovery of specifically acquired immunity which followed the major contributions of Louis Pasteur completely over-shadowed the first studies of the host's natural resistance. Later, the exquisite sensitivity and precision of antigen-antibody reactions made the study of immunochemistry much more attractive than the rather primitive and ambiguous field of non-specific immunity. Neverthe less, during the last three decades, a considerable body of informa tion was developed and also means by which natural resistance could be enhanced or depressed by exogenous agents such as lipopolysaccharides or BCG. An important advance was the chemical recognition of the biologically active components of these agents which in turn allowed the synthesis or" analogues. More recently, endogenous host products which can play a role in nonspecific immunity, such as thymic hormones, have also been identified, produced and used both experimentally and clinically. It therefore seemed worthwile to Drs. Miescher and Mueller-Eberhard to devote two volumes of Seminars in Immunopathology to the topic of Immunostimuhltion. Because of the good response obtai ned from readers, Springer Verlag decided to issue a hard cover book and asked their guest editor to make a preface. Prefaces, although they are found in the opening pages, are always written after the first issue has been completed.

It is obvious that exaggerated expectations will lead to disappointments, and complete neglect of a once-exciting field is to be followed by renewed attention, if the scientific basis of earlier interest was solid and remained so throughout. The healing ef fect of fever was praised as nature's blessed cure, until it was discov ered that it is caused by bacteria. When it became evident that by careful application one can arrive at therapeutic doses without intro ducing shock and hypotension, the era of fever therapy began. During this time, indiscriminate use of pyrogens was so widespread that the high expectations had to be followed by disillusionment. Since then, in terest in endotoxins experienced a few more oscillations, with varying wavelengths and amplitudes, the latest being triggered by the mitotic effect of endotoxin. This led to justified excitement among researchers who study the activation of the cells of the lymphatic system and the consequences thereof, including the release of important mediator mol ecules. Fever therapy is passe, but the healing that often followed the ap plication of endotoxins remains a well-documented fact.
