I recommend making expectations explicit in order to design controls against unconscious influence, formulate alternative outcomes more clearly-and to add zest. Both the immediately upcoming experiment and the expected result of many long years of work by many people after one is gone are proper subjects for betting or the most informed and serious guessing. The working title for this collection of new and old papers was for some time "Betting on how brains work" and then "Betting on brains. " It goes without saying that the book will not answer the title question but will speak to it, in particular making a series of propositions that I think are more likely to be confirmed by future research than the alternatives we can presently recognize. It follows that a significant message, implied in many chapters of the book is this.

Book excerpt: To bring physiology and pathology of the human brain into better micro-anatomical and histological context, studies with different methodologies are required. Established techniques such as electron microscopy or histology show limitations in view of invasiveness, labor-intense and artifact-prone sample preparation, as well as an adequate ratio between resolution and volume throughput. For this reason, X-ray phase-contrast tomography (PC-CT) has been proposed as a three-dimensional non-destructive imaging technique, which requires less effort in sample preparation and can assess larger volumes. Furthermore, it offers quantitative electron density based contrast even for unstained tissue. Up to now, however, PC-CT studies fell short in number of samples, so that structural alterations caused by neurodegenerative diseases cannot be distinguished from physiological inter-subject variations. In this thesis, the scalability of PC-CT with respect to the required number of samples and resolution-to-volume-throughput is demonstrated, and the methodology is advanced with respect to data acquisition, processing and segmentation. In addition to the human cerebellum, cortex and hippocampus are studied. Concerning quantification and analysis of PC-CT data, this work introduces optimal transport analysis to obtain quantitative metrics of the cyto-architecture and to identify changes due to neurodegenerative diseases. For the case of Alzheimer’s disease, this workflow reveals a yet undescribed compactification of granular cells in the human hippocampus. This thesis also provides optimized configurations to study neural tissues with laboratory instrumentation, and – finally – provides new correlative imaging approaches, in particular with scanning electron microscopy.

Book excerpt: The neuron doctrine, first formulated in 1891, states that the brain is constructed of individual neurons, organized into functioning circuits that mediate behavior. It is the fundamental principal that underlies all of neuroscience and clinical neurology. Foundations of the Neuron Doctrine gives an authoritative account of how this theory was the product of an explosion of histological studies and vigorous debates near the end of the nineteenth century by an extraordinary group of scientists, led by Santiago Ramon y Cajal of Spain, using a selective stain discovered by Camillo Golgi of Italy. They were the first to describe the distinctive branching patterns of nerve cells, providing evidence that the cells interact as individual units to form circuits, opposed however by Golgi, who held out for a view that the nerve cells form syncytial networks. Studies in the 1950s appeared to confirm the nerve cell as an individual unit, as embodied in the neuron doctrine, which became the basis for the rise of concepts of normal and disordered neural function since then. This 25th Anniversary Edition is timely. Recent studies are showing a much greater degree of complexity in neuronal organization, so that the debate of neuron versus network is again coming to the fore in neuroscience research. Book excerpt: The Spanish conquest of central Mexico in 1521 set in motion an evangelization campaign to convert the large indigenous populations to Catholicism. Franciscans, Dominicans, and Augustinians participated in the first stages of this campaign. The missionaries established doctrinas (missions) in many indigenous communities, and, during the sixteenth century, directed the construction of new sacred complexes, often on the site of pre-Hispanic temples. Many of the convent complexes still survive in various states of conservation. This Visual Catalog offers historical data regarding the convent complexes, as well as an extensive collection of photographs of the surviving buildings, murals, and design elements, and documents the Franciscan doctrinas. In the 1580s, Fray Antonio de Ciudad Real, O.F.M. accompanied the Comisario General Fray Alonso Ponce, O.F.M. on an inspection of the Franciscan installations in central Mexico and Central America. The book reproduces his descriptions of the Franciscan missions, and is accompanied by photographs of the convent complexes. It also documents the Dominican and Augustinian doctrinas, and discusses selected Jesuit colegios and missions in Mexico. The Jesuits first arrived in Mexico in 1572, and did not participate in the first evangelization campaign. They were active in urban missions and education, and also established missions on the far northern frontier of Mexico.
