It deals both experimentally and theoretically with the question of symptom specificity in psychosomatic research – why some individuals respond to psychological stress with gastric disorders, others with sexual impotence, and still others with high blood pressure. As the author notes in summarizing his conclusions, "The repeated pairing of activation of a given organic system with intense nervous stress directs the pathological influence of the stressor primarily upon the system activated; subsequently the natural stimuli which would ordinarily activate the system in a normal manner sustain the pathological stressor’s effect as a conditioned stimulus for the stressor effect." The translation of this work from the original Russian brings to the attention of Western investigators new and useful models of stress-induced disorders, and sheds new light on the pervasive problem of psychosomatic disease.

The workshop focused on the availability of marmosets in the United States and abroad; animal welfare and ethical considerations stemming from the use of wildtype and genetically modified marmosets; and standards of housing and care, dietary needs, and feeding requirements for marmosets in captivity. Most CNS drugs fail because of efficacy, and the core issue underlying these problems is a poor understanding of disease biology. Concern about the poor productivity in neuroscience drug development has gained intensity over the past decade, amplified by a retraction in investment from the pharmaceutical industry. This retreat by industry has been fueled by the high failure rate of compounds in advanced clinical trials for nervous system disorders. In response to the de-emphasis of CNS disorders in therapeutic development relative to other disease areas such as cancer, metabolism, and autoimmunity, the National Academies of Sciences, Engineering, and Medicine initiated a series of workshops in 2012 to address the challenges that have slowed drug development for nervous system disorders. Motivated by the notion that advances in genetics and other new technologies are beginning to bring forth new molecular targets and identify new biomarkers, the Academies hosted the third workshop in this series in September 2016. Participants discussed opportunities to accelerate early stages of drug development for nervous system disorders in the absence of animal models that reflect disease and predict efficacy. Within the biomedical research community, there is little doubt about the utility of such models. But with increasing frequen cy, individuals concemed with animal rights question the validity of such models and argue that they actually produce no useful information for understanding or developing treatments for human disorders. Each of the chapters will address this issue, considering the utility, validity, generalizability, and limitations of the models presented. Chapter 1 begins with a brief review of the ethics of animal use in neuro biological research, inc1uding a discussion of the criteria that may be used to evaluate animal models of human disease and extrapolate from the model to appropriate questions regarding humans. The limitations of such extrapolation are also discussed, with special consideration of issues specific to the use of neurotoxins. Chapter 2 extends this theme with further consideration of issues and strategies involved in developing neurotoxin-induced models of neuro degenerative disorders and assessing risk factors associated with neurotoxin ex posure. It then narrows to evaluate the use of a model systems approach to explore neurotoxin-induced leaming and memory deficits in animals as related to humans with dementia.
