Book excerpt: Cancer is a leading cause of death in the United States and accounts for about $209.8 billion of the financial burden on the United States healthcare system. One of the most common occurrences in more than 90% of the cancers is the inactivation of Retinoblastoma (RB) tumor suppressor gene. In normal cells, RB is responsible for the negative regulation of cell cycle by inhibiting the E2 promoter binding (E2F) proteins. In addition to this, it has also been shown that RB can regulate multiple physiological processes such as apoptosis, differentiation, metabolic pathways and immune response by mediating transcriptional and proteomic changes. Previous studies outlining RB-loss have approached this question in the context of long-term loss of RB function in a transformed, cancer cell line or a tumor in a model organism. To understand the cellular changes immediately after RB-loss, we developed a model system to deplete RB1 over a shorter period of six days. We then performed experiments to profile changes to both RNA and protein levels. Our data suggests that many proteins that are involved in the cellular metabolic processes and other pathways are altered in presence of RB-loss. In addition, we observe poor correlation between the mRNA and protein levels in our dataset, suggesting additional layers of post-transcriptional regulation. Previous studies from our lab have identified post-transcriptional factors such as RNA-binding proteins that can alter tumor phenotypes in Drosophila. In addition, we have also performed high-throughput sequencing analysis of human Retinoblastoma tumors with control retinas. From this study, we identified several long non-coding RNAs that were altered in our dataset which are involved in post-transcriptional regulation. Understanding this mechanism of regulation will help us develop therapeutic strategies adapted to target the specific sensitivities of cancer cells. In continuation of our studies in pediatric tumor disposition syndromes, our lab also identified a novel mode of post-transcriptional regulation in a cancer-predisposition disease called DICER1 syndrome. Previous studies have identified mutations on the coding sequence of the DICER1 protein that can cause the rare tumors found in DICER1 syndrome patients. However, not all DICER1 syndrome patients have mapped mutations on the DICER1 protein. In our study, we identified a human DICER1 syndrome patient with pathogenic allele containing a mutation on the 3’UTR of the DICER1 transcript. This mutation falls on the binding site of the Pumilio (PUM) family of RNA binding proteins and can affect the binding of this protein to the DICER1 transcript. Our study shows that disruption of this regulatory site can alter the levels and function of the DICER1 protein. This can also alter the miRNA levels in the cell lines with altered DICER1 protein levels which can affect cellular phenotypes. DICER1 expression is a double-edged sword in cancer, and it is important for the cells to maintain the right amount of DICER1 making it hard to develop therapeutic strategies directly targeting DICER1. Understanding PUM-mediated post-transcriptional regulation of DICER1 may provide new opportunities to target DICER1 activity in tumors.
