In fact one of the initial functions attributed to gap junction channels was the passage of growth regulatory signals. This hypothesis was supported by the negative correlation between intercellular communication and the rate of cell growth in many cell lines. Decreases in gap junctional coupling have consistently been associated with the induction of rapid or synchronous cell division in both normal and pathological conditions. Unfortunately, the precise role of junctional communication in growth regulation still remains a mystery. Here we have attempted to explore the complex crosstalk which occurs between cell surface receptor and non-receptor tyrosine kinases and Cx43 gap junctional communication. Cells transformed by the v-src oncogene have offered a particularly useful system for investigation of the mechanisms) that can mediate both acute and chronic closure of gap junction channels. Using the Xenopus oocyte expression system we have dissected multiple signaling pathways by which the initial (acute) closure of pre-formed Cx43 channels in response to v-src occurs. In the second study, using biochemical and fluorescence microscopic techniques we have attempted to investigate the raison d'étre for the acute inhibition of junctional communication during mitogenic stimuli.

More importantly, antibodies to three major gap junction (GJ) proteins were generated and used to characterize the GJ proteins of various tissue culture systems. Progress has also been made in understanding the biochemical and molecular basis of the action of certain tumor promoting chemicals, such as TPA, mezerein and bryostatin, which indicated that protein kinase C (PKC), an import component of cellular second messenger system, was activated. Since gap junction protein is considered to be affected by PKC, the observations we made suggest that PKC activating toxicants can exert their action as tumor promoters through abolishing GJ protein function. Another study suggested that certain oncogenes, ras, neu and src, induce cellular transformation and the resulting transformed cells have very poor GJIC. Studies are underway to identify the mechanisms of gap junction protein regulation.

This stain targeted intermediate filaments that we now know consist mainly of glial fibrillary acidic protein, a protein used today as an astrocytic marker. Cajal described the morphological diversity of these cells with some ast- cytes surrounding neurons, while the others are intimately associated with vasculature. We start the book by discussing the heterogeneity of astrocytes using contemporary tools and by calling into question the assumption by classical neuroscience that neurons and glia are derived from distinct pools of progenitor cells. Astrocytes have long been neglected as active participants in intercellular communication and information processing in the central nervous system, in part due to their lack of electrical excitability. The follow up chapters review the “nuts and bolts” of ast- cytic physiology; astrocytes possess a diverse assortment of ion channels, neu- transmitter receptors, and transport mechanisms that enable the astrocytes to respond to many of the same signals that act on neurons. Since astrocytes can detect chemical transmitters that are released from neurons and can release their own extracellular signals there is an increasing awareness that they play physiological roles in regulating neuronal activity and synaptic transmission. In addition to these physiological roles, it is becoming increasingly recognized that astrocytes play critical roles during pathophysiological states of the nervous system; these states include gliomas, Alexander disease, and epilepsy to mention a few.
