Of particular interest is the finding that, in several tissues, the cell surface is differen tiated at the intercellular region, thereby providing communicating chan nels between apposing cells. Although our actual knowledge of the precise structures and mecha in the complex process of intercellular communication is still nisms involved scanty, evidence has been presented that ions and molecules diffuse from cell to cell, establishing a physiological continuum. Embryonic differentia tion, cell growth, neoplasia, electrical synchronization in nerve and mus cles, as well as immune response seem to be related to ceIl communication. In organizing this volume, it has been our intention to provide the reader with an actual review of the processes involved in intercellular communication in normal tissues as well as in neoplasia. The study of cell-signaling m- ecules in this context is a central component of cancer research. From the knowledge of such targets, investigators have been able to productively advance many insightful hypotheses about how a particular cancer cell may misinterpret, or respond inappropriately to, growth regulatory cues in their environment. Despite these key insights, the rapidly evolving nature of cell signaling research in cancer has necessitated a continuous revision of these theoretical constructs and the updating of methods used in their study. One contemporary example of the evolution of this field is provided by an analysis of the Human Genome Project data, which reveal a previously unsuspected diversity in the multigene families encoding for most signaling pathway int- mediates. In assessing the usefulness of a particular methodological approach, therefore, we will need to keep in mind that there is a premium on those p- tocols that can be easily adapted for the analysis of multiple members within a gene family. A large class of proteins called kinases are enzymes required by cancer cells to grow, proliferate, and survive apoptosis (death) by the immune system. Two important kinases are MET and RON which are receptor tyrosine kinases (RTKs) that initiate cell signaling pathways outside the cell surface in response to extracellular ligands (growth factors.) Both kinases are oncogenes which are required by cancer cells to migrate away from the primary tumor, invade surrounding tissue and metastasize. MET and RON reside on both cancer cells and the support cells surrounding the tumor, called the microenvironment. MET and RON are activated by their particular ligands, the growth factors HGF and MSP, respectively. Blocking MET and RON kinase activation and downstream signaling is a promising therapeutic strategy for preventing tumor progression and metastasis. Written for cancer physicians and biologists as well as drug discovery and development teams in both industry and academia, this is the first book of its kind which explores novel approaches to inhibit MET and RON kinases other than traditional small molecule kinase inhibitors. These new strategies target key tumorigenic processes on the outside of the cell, such as growth factor activation by proteases. These unique strategies have promising potential as an improved alternative to kinase inhibitors, chemotherapy, or radiation treatment.

Book excerpt: Malignant growth of cells is often characterized by disorganization of tissue structure, abnormal blood vessel development, and insuffi cient vascular supply. As a consequence, the cancer cells grow in a three-dimensional pattern in atypical microenvironments which include physical, chemical, and nutritional stresses. Necrosis often develops some distance away from the blood vessels. In association with an inherent instability in malignant cell populations, and also because of the changing micromilieu, significant cellular heteroge neity emerges with regard to various phenotypic characteristics. Both biological behavior and responses to therapeutic agents can be affected. A variety of in vitro and in vivo experimental models exist for research on properties of cancer cells during growth. The multicell spheroid model was developed as a system of intermediate complexity in which three dimensional growth of cells enhances cell-cell interactions and creates micro environments that simulate the conditions in intervascular microregions of tumors or microme tastatic foci. Spheroids may change their cellular characteristics with changing environments during growth. These can be studied under controlled conditions in vitro. Interest in details of experimental methods for this model system stimulated the organization of the First International Conference in Rochester, NY in 1980, the Proceedings of which were summarized in Cancer Research in 1981. Since then there has been a rapid increase in the use of this model system, and increased research on the significance of cell-cell and cell-microenvironment interactions in biology in general.
