Glycerophospholipid and/or sphingolipid second messengers resulting from the action of lipid metabolizing enzymes (e.g. lipid-kinases or lipases) are commonly found within cells. The importance of such mediating lipids in plants has become increasingly apparent. Responses to biotic and abiotic stresses, and to plant hormones, all appear to involve and require lipid signals. Likewise, developmental processes, in particular polarized growth, seem also to involve signalling lipids. Amongst these lipids, phosphatidic acid (PA) has received the most attention. It can be produced by phospholipases D, but also by diacylglycerol kinases coupled to phospholipases C. Proteins that bind phosphatidic acid, and for which the activity is altered upon binding, have been identified. Furthermore, other lipids are also important in signalling processes. PA can be phosphorylated into diacylglycerol-pyrophosphate, and plants are one of the first biological models where the production of this lipid has been reported, and its implication in signal transduction have been demonstrated. PA can also be deacylated into lyso- phosphatidic acid. The phosphorylated phosphatidylinositols, i.e. the phosphoinositides, can act as substrate of phospholipases C, but are also mediating lipids per se, since proteins that bind them have been identified. Other important lipid mediators belong to the sphingolipid family such the phosphorylated phytosphingosine, or long-chain bases. Many questions remain unanswered concerning lipid signalling in plants. Understanding and discussing current knowledge on these mechanisms will provide insights into plant mechanisms in response to constraints, either developmental or environmental.

Plants have evolved a wide range of mechanisms for coping with various abiotic stresses. In addition, genomics has become a necessary tool for the understanding of plant stress responses at the whole genome levels. The identification of stress-tolerant plant resources and the investigation of the functional role of the genetic variants is also a valuable tool in this research field. Recently, the advent of CRISPR/Cas genome editing technology, enables these variations to be introduced in crops for improved stress tolerance traits. Through the understanding of the molecular mechanisms involved in plant signaling in response to abiotic stress and crop performance characters under stress conditions, we hope to open new ways for the breeding of superior crops.

The model plant Arabidopsis thaliana is well-suited to study stress-driven compositional dynamics, metabolism, and functions of lipid metabolites. When Arabidopsis plants were subjected to wounding, infection by Pseudomonas syringae pv tomato DC3000 expressing AvrRpt2 (PstAvr), infection by Pseudomonas syringae pv. maculicola (Psm), and low temperature, and 86 oxidized and acylated lipids were analyzed using mass spectrometry, different sets of lipids were found to change in level in response to the various stresses. Analysis of plant species (wheat versus Arabidopsis), ecotypes (Arabidopsis Columbia 0 versus Arabidopsis C24), and stresses (wounding, bacterial infection, and freezing) showed that acylated monogalactosyldiacylglycerol was a major and diverse lipid class that differed in acyl composition among plant species when plants were subjected to different stresses. Mass spectrometry analysis provided evidence that oxophytodienoic acid, an oxidized fatty acid, is significantly more concentrated on the galactosyl ring of monogalactosyldiacylglycerol than on the glycerol backbone. A mass spectrometry method, measuring 272 lipid analytes with high precision in a relatively short time, was developed. Application of the method to plants subjected to wounding and freezing stress in large-scale experiments showed the method produces data suitable for lipid co-occurrence analysis, which identifies groups of lipid analytes produced by identical or inter-twined enzymatic pathways. The mass spectrometry method and lipid co-occurrence analysis were utilized to study the nature of lipid modifications and the roles of lipoxygenases and patatin-like acyl hydrolases in Arabidopsis during cold acclimation, freezing, and thawing.
