Book excerpt: Boron Lewis acids are a class of reagent that has been widely studied and utilized in organic chemistry for over sixty years. The popularity of these reagents comes from their diverse reactivity, functional group tolerance, and non-toxic nature. For these reasons, the continued development of chemistries involving boron reagents still receives much attention. This thesis examines the development of new methodologies for the functionalization of organic molecules based on boron reagents. Chapter 1 briefly describes general methodologies to synthesize azide compounds. Later, the mechanisms of photochemical decomposition of various azides will be discussed. Borontribromide-mediated dissociation of carbon-oxygen bonds has been extensively used to cleave aryl ethers. However, the regioselectivity is low when applying this method to unsymmetrical dialkyl ethers. The second chapter of this thesis describes the cleavage of unsymmetrical dialkyl ethers employing mixed boron trihalides. The selectivity profile of this new methodology is evaluated on various ether substrates, including benzyl, allyl, and propargyl ethers. Nitrenes are a high energy, reactive specie generated from thermolysis or photolysis of an azide precursor. Recently, it has been demonstrated that boryl nitrenes can insert into C−H bonds of unreactive substrates such as alkanes. The third chapter describes a one-pot three-step procedure to generate amino alcohols from simple or complex alcohols utilizing a boryl azide precursor. Using mixed boron halides, as described in chapter 2, many dialkoxyboryl chlorides were prepared and subsequently converted to the dialkoxyboryl azide. Chapter 3 describes the investigation of the photochemical decomposition of these azides. The Suzuki-Miyaura reaction is a versatile and powerful synthetic method to construct C−C bonds using aryl boronic acids. However, the application of this method in modification of biomolecule substrates is limited due to the instability of many biological molecules such as proteins in the presence of bases and high temperature. Chapter 4 will discuss the synthesis of new N-heterocyclic carbenes ligands to facilitate the Suzuki cross-coupling reaction in aqueous media in the absence of bases and elevated temperature.

These include the haloallylation of aryl aldehydes with boron trihalide; alkyne-aldehyde couplings mediated by boron trihalide to give either (Z, Z)-1,5-dihaloalkenes or (Z, E)-1,5-dihaloalkenes; dehydroxylation of benzylic alcohols and in situ generated benzylic alcohol salts with iron(III) chloride and triethylsilane to afford diarylmethanes; and, the alkenylation of allylic, benzylic, and propargylic alcohols with phenylacetylenes and titanium(IV) halide. The results of these studies strongly imply a cationic mechanism. The reaction methodology of the research described herein can be characterized as atom-efficient, environmentally friendly, and capable of generating the desired products in good to excellent yields.

Book excerpt: Carbon-rich, conjugated organic scaffolding is a popular basis for functional materials, especially for electronic and photonic applications. However, synthetic methods for generating these types of materials lack diversity and, in many cases, efficiency; the insistence of investigators focusing on the properties of the end product, rather than the process in which it was created, has led to the current state of the relatively homogeneous synthetic chemistry of functional organic materials. Because of this, there is plenty of room for improvement at the most basic level. Problems endemic to the preparation of carbon-rich scaffolding can, in many cases, be solved with modern advances in synthetic methodology. We seek to apply this synthesis-focused paradigm to solve problems in the preparation of carbon-rich scaffolds. Herein, the development and utilization of three methodologies: iridium-catalyzed arene C-H borylation; zinc- mediated alkynylations; and Lewis acid promoted Mo nitride-alkyne metathesis, are presented as improvements for the preparation of carbon-rich architectures. In addition, X-ray crystallographic analysis of two classes of compounds are presented. First, an analysis of carbazole-containing arylene ethynylene macrocycles showcases the significance of alkyl chain identity on solid-state morphology. Second, a class of rigid zwitterionic metal-organic compounds display an unusual propensity to crystallize in the absence of inversion symmetry. Hirshfeld surface analysis of these crystalline materials demonstrates that subtle intermolecular interactions are responsible for the overall packing motifs in this class of compounds.

Benefits include step and atom economy, as well as the ubiquitous presence in organic molecules. Advances to this exciting realm of synthesis are described in this thesis with an emphasis on the development of catalytic, selective reactions under mild conditions. Additionally some applications of the methodologies are demonstrated. In Chapter 1, the first examples of inter-and intramolecular enantioselective conjugate alkenylations employing organostannanes are reported. A chiral, cationic Rh(I)-diene complex catalyzed the enantioselective conjugate addition of alkenylstannanes to benzylidene Meldrum's acids in moderate enantiomeric ratios and yields. Notably, the cationic and anhydrous conditions required for the asymmetric alkenylation are complementary to existing protocols employing other alkenylmetals. In Chapter 2, a domino, one-pot formation of tetracyclic ketones from benzylidene Meldrum's acids using Sc(OTf)3 via a [1,5]-hydride shift/cyclization/Friedel-Crafts acylation sequence is described. Respectable yields were obtained in accord with the ability to convert to the spiro-intermediate, and considering the formation of three new bonds: one C-H and two C-C bonds. An intriguing carbon-carbon bond cleavage was also serendipitously discovered as part of a competing reaction pathway. In Chapter 3, the pursuit of novel C-H bond transformations led to the development of non-carbonyl-stabilized rhodium carbenoid Csp3-H insertions. This methodology enabled the rapid synthesis of N-fused indolines and related complex heterocycles from N-aziridinylimines. By using a rhodium carboxamidate catalyst, competing processes were minimized and C-H insertions were found to proceed in moderate to high yields. Also disclosed is an expedient total synthesis of (±)-cryptaustoline, a dibenzopyrrocoline alkaloid, which highlights the methodology. In Chapter 4, the Lewis acid promoted substitution of Meldrum's acid discovered during the course of the domino reaction was explored in detail. The protocol transforms unstrained quaternary and tertiary benzylic Csp3-Csp3 bonds into Csp3-X bonds (X = C, N, H) and has even shown to be advantageous with regards to synthetic utility over the use of alternative leaving groups for substitutions at quaternary benzylic centers. This reaction has a broad scope both in terms of suitable substrates and nucleophiles with good to excellent yields obtained (typically>90%).
