Several studies in animal models have demonstrated the efficacy of these vaccines for triggering the immune system to induce specific and protective immunity against tumors of different origin. In fact, anti-idiotypic mAbs have been used successfully in cancer therapeutics to overcome the poor immunogenicity of some tumor-associated antigens, in particular those of non-protein origin. In some clinical trials, including patients with different tumors, anti-Id specific humoral and/or cellular responses following immunization were associated with improved clinical outcome. This number of Frontiers will be dedicated to anti- idiotypic antibodies and their use in cancer treatment. Book excerpt: The phenomenon of idiotypy was discovered almost thirty years ago, but it was only during the past decade that it attracted widespread interest and became the subject of numerous research investigations. From the outset, much of the interest in idiotypy was based on its implications with respect to the repertoire of antibodies. Kunkel showed, for example, that idiotypes associated with certain human myeloma or Bence-Jones proteins were present in normal human globulins at levels of less than one part per million. Also, Oudin's original definition of idiotypy implied that idiotypes could be uniquely associated with individual rabbits as well as with particular antigen-binding specificities. Such observations provided some of the earliest evidence for an extensive repertoire of immunoglobulin molecules. The implications of these findings have been amply confirmed by recent studies of protein struc ture and molecular genetics; many of these studies are reviewed in the present volume. It is known now that the diversity of antibodies is based on the presence of numerous V and L V H genes, on recombinatorial events involving D and] segments, on somatic mutations, and on processes involving deletion of DNA followed by repair with errors, including inser tions. Each of these parameters is capable of influencing the idiotype expressed by the final immunoglobulin product. Regulation of the immune response is another area in which idiotypy has significantly influenced modern immunology.

Solid-phase synthesis of peptides is leading to an array of synthetic vaccines, an approach that is expected to attain its full potential once the sequences activating suppressor cells are discovered and the rules for presentation of antigens to T and B cells are better worked out. A new breed of vaccines is on the horizon that seeks to control fertility.

This is true even for the antigen binding sites of antibodies called idiotypes, which are bound by complementary bind ing sites of other antibodies called anti-idiotypes. Thus, anti-idiotypes are structural homologues of antigens. These idiotypic-anti-idiotypic interactions constitute a network within the immune system. Since one lymphocyte produces only one type of antibody molecule, this network is in fact a network of cells. We expect that the network is functional: the appearance of antigen will disturb the equilibrium of the network at the point where it competes with the anti idiotypic lymphocyte for binding to the idiotypic lympho cyte. It has been known for quite some time that anti idiotypic antibody can be used to prime the immune system for memory to an antigen that it has never seen. This phe nomenon is now being explored for possible use in immuni zation against viruses, bacteria, parasites and tumors as well as for the modulation of autoimmunity. The ability of anti-idiotypes to mimic, both antigenically and function ally, the corresponding biologically active molecules seen by an idiotypic antibody was first demonstrated for the hormone insulin and is now being observed in many other systems. The papers assembled in this volume· bring the reader to the cutting edge of the potential practical applica tions of the network theory of the immune system.
