Book excerpt: The biological membranes of cellular organization enfold an important group of membrane proteins called the ATPases, which are not only versatile in maintaining chemical gradient and electrical potential across the membrane but also bring metabolites necessary for cell metabolism and drive out toxins, waste products and solutes that otherwise can curb cell functions. ATPases are distributed virtually in all live forms starting from unicellular to multicellular and also in viruses. There are different types of ATPases, which differ in function and structure and in the type of ions they transport. The three main types of the ion pump ATPase family are: (i) P-type ATPases that transport different ions across membranes and Ca2+ATPases belongs to this catagory (ii) F-type ATPase in mitochondria, chloroplasts and bacterial plasma membranes produce ATP using the proton gradient; and (iii) V-type ATPase catalyzes ATP hydrolysis to transport solutes and maintains acidic pH in organelles like lysosomes. Genetic defects in either of the ATPases cause several diseases and a number of researches have demonstrated the involvement of the members of ATPases in the cell pathology and diseases, thereby penetrating exciting new areas of our understanding. In this book, the authors summarize recent knowledge about the molecular mechanisms associated with Ca2+-ATPase, V-ATPase and F-ATPase in intracellular and extracellular Ca2+ transport, mitochondrial ATP synthase, vesicular H+ transport, and lysosomal pH regulation. This book thereby bridges the gap between fundamental research and biomedical and pharmaceutical applications. We have not attempted to include material on all types of plasma and intracellular membranes, but rather have emphasized structures which have been studied relatively thoroughly. Four chapters describe transport of different types of molecules and ions across the plasma membranes of mammalian red cells. Two essays concern the excitable membranes of nerve and muscle cells while the remaining four chapters treat transport across several types of intracellular membranes. Water makes up more than two-thirds of the mass of most living cells. The transport of water between the inside and outside of cells and organelles is important for the function of these structures. As a result of investigations in many laboratories over the past four decades, our picture of the water permea bility of the red cell membranes is rather detailed when compared to the water permeability of other biological membranes. In Chapter 1, R. I. Macey describes this picture and also considers the permeability of red cell membranes to non electrolytes, including metabolic substrates such as sugars, amino acids, purines and nucleosides.

The aim of the colloquium was to bring into focus data relating to cell membranes that might contribute to understanding the pathogenic mechanism of Duchenne muscular dystrophy. A major impediment to progress in understanding the patho genesis of muscular dystrophy has been the failure, so far, to identify the basic genetic defect. Pending the identification of the genetic lesion in Duchenne dystrophy and, in view of scattered but persistent indications of a basic membrane disturbance, it seemed worthwhile to explore in open dialogue the current state of knowledge of membrane morphology and chemistry with an eye to possible leads for further investigation. The participants, drawn from a variety of interested disciplines, attempted to synthesize and reconcile their findings and to identify crucial areas of ignorance in need of exploration. For the most part they avoided specialized jargon and spoke in a language that could be understood by the rest of the group. Apart from providing a review of widely varying approaches to the study of the composition and behavior of cell membranes, the discussions brought together current think~g on strategies and approaches to the study of the pathogenesis of muscular dystrophy. Already the personal contacts made at the colloquium have led to new inter-institutional collabora tive investigations.
