Book excerpt: Women with estrogen receptor positive breast tumors respond to adjuvant hormonal therapy with an enhancement of disease free survival and overall survival. A substantial fraction of tumors are dependent upon estrogen for growth, and antiestrogen has been shown to be effective for the treatment of breast cancer. The precise mechanism of estrogen and anti-estrogen action in breast tumors still remain unclear. We postulated that estrogen and anti-estrogen act by activating the function of the endogenous estrogen receptor. Using rat mammary gland as the model, we examined the function of ER in response to estrogen and antiestrogen. Our data suggested that estrogen and anti-estrogen are able to activate and regulate the function of estrogen receptor in a cell context dependent fashion. We conclude that estrogen and anti-estrogen may exert their effects on the growth of target tissues, at least in part, by increasing the functional activity of the endogenous estrogen receptor. Abstracts for the research done in this funding period were submitted to the Annual Endocrine Meeting in June, 1997 and Era of Hope DOD BCRP Meeting in October, 1997 for presentation.

Regulation of gene expression by hormone-bound steroid receptors Is mediated by members of the steroid receptor coactivator (SRC) family. The purpose of this study was to examine the role of SRC-1 in the regulation of gene expression by steroid receptors. In breast cancer SRC-1 was found to exist in a multifactor complex which was stably associated with liganded progesterone receptor, implicating an active SRC-1 complex as a primary target of liganded PR in breast cancer cells. In an effort to further define the role of SRC-1 in breast cancer, we characterized a member of the active SRC-1 complex, SRA (steroid receptor RNA coactivator), which was shown to function as an RNA. Significantly, SRA Is a limiting factor required for efficient transcriptional activation by steroid hormone receptors in breast cancer cells.

Book excerpt: In Breast Cancer: Cellular and Molecular Biology [Kluwer Academic Pub lishers, 1988], we tried to present an introduction to the emerging basic studies on steroid receptors, oncogenes, and growth factors in the regulation of normal and malignant mammary epithelium. The response to this volume was superb, indicating a tremendous interest in basic growth regulatory mechanisms governing breast cancer and controlling its malignant progres sion. In the two years since its publication, much new and exciting in formation has been published and the full interplay of regulatory mechanisms is now beginning to emerge. We have divided this book into four sections that we hope will unify important concepts and help to crystallize areas of consensus and/or disagreement among a diverse group of basic and clinical scientists working on the disease. The first section is devoted to studies on oncogenes, antioncogenes, proliferation, and tumor prognosis. The first chapter, by Sunderland and McGuire, introduces the characteristics of breast cancer as studied by patho logists to establish prognostic outcome. Of particular interest is a new proto oncogene called HER-2 (or neu), which is rapidly becoming accepted as a valuable new tumor marker of poor prognosis. The second chapter, by Lee Bookstein and Lee, introduces the best known antioncogene, the retinoblas toma antioncogene, whose expression is sometimes lost in breast cancer. Malignant progression appears to be influenced by the balance of proto oncogene and antioncogene expression.

This volume brings together an international team of prominent investigators who discuss their most recent work on the basic and clinical aspects of steroid/nuclear receptors. The contributions represent updated versions of the invited presentations made at The Second Meadow Brook Conference on Steroid Receptors in Health and Disease. Book excerpt: Members of the steroid receptor coactivator family have been implicated in the regulation of nuclear receptor function by enhancing ligand-dependent transcriptional activation of target genes. We have previously isolated the third member of the SRC family, receptor-associated coactivator 3 (RAC3). In this study, we investigated the mechanisms by which RAC3 may interact with and modulate the transcriptional activity of nuclear receptors. We found that VDR, ERBeta, and retinoid receptors interact with different a-helical LXXLL motifs of RAC3. Peptides corresponding to these motifs have diverse affinities for the VDR and ERBeta and mutation of specific motifs differentially impairs the ability of RAC3 to interact with receptors in vitro. Consequently, these mutations inhibit the enhancement of transcriptional activation by these receptors in vivo. Furthermore, we found that the activation function-2 (AF-2) domain of RXR interferes with RAC3 binding to DNA-bound VDR/RXR or RAR/RXR heterodimers, while the VDR or RAR AF-2 domains are required for this interaction. Finally, we demonstrated that RAR and RXR differentially contribute to coactivator recruitment and transcriptional regulation by the RAR/RXR heterodimer. These results suggest a receptor-specific binding preference for the different LXXLL motifs of RAC3, which may provide flexibility for RAC3 to regulate the function of different receptors.
