Book excerpt: In winter, all drought stress treatments induced dehydin-like proteins at mid-seed development and reduced the level of soluble sugars in the mature seeds. The reduction in soluble sugars in seeds from drought stressed plants coincided with the reduction in AA-germination. In winter and field experiments, drought stress treatments increased the concentration of P, K, Ca, Mo, Mn, and Cu in seed similar in size and weight to seeds from well-watered plants; but, the concentrations varied between the two experiments. Drought-stress treatments reduced seed size in the winter experiment, but not in field experiment. Medium seeds had lower nutrient content than large seeds. In conclusion, all drought stress treatments reduced yield, yield components, increased production of small and medium seeds, and induced expression of dehydrins at mid-seed development. Well-watered and gradual stress treatment improved seed vigor (AA-germination) of large seeds over that observed in the SS treatment and this reduction in seed vigor was coincided with the reduction in soluble sugars.

Book excerpt: With the increasing scarcity of water resources, soil moisture stress is the single most threat to global soybean production causing extensive yield losses. The objectives of this study were to investigate soil moisture stress effects on all aspects of soybean growth and development processes and to develop functional algorithms that could be used for field management decisions and in soybean crop modeling. To fulfill these objectives, six experiments were conducted; one in vitro osmotic stress study on seed germination, four studies by imposing five soil moisture treatments, 100, 80, 60, 40, and 20% of daily evapotranspiration of the control at different growth stages using sunlit plant growth chambers, and one transgenerational study on seed germination at different osmotic levels and offspring growth at three irrigation treatments (100, 66, and 33% based on field capacity) for plants grown at different soil moisture levels. Two cultivars from maturity group V, Asgrow AG5332 and Progeny P5333RY, with different growth habits were used in all these studies. Midday leaf water potential, plant height, mainstem nodes, gas-exchange traits, canopy reflectance, and several yield components including pod weight, seed yield, and seed quality were measured. Soil moisture stress decreased biomass, net photosynthesis, yield, individual seed weight, maximum seed germination, protein, fatty acids, sucrose, N, and P and increased oil, stachyose, Fe, Mg, Zn, Cu, and B contents. Overall, Asgrow AG5332 was more tolerant to drought stress than Progeny P5333RY. Soil moisture stress induced changes in seed quality that were correlated with seed germination and seedling vigor in the F1 generation. These data can be used to build a model-based decision support system capable of predicting yield under field conditions.

Additionally grain Mn and B from the same plants increased by an estimated 23 and 41%, respectively. However, plants fertilized during seed fill had an estimated 18% reduction in grain Fe concentration, and no change in grain Cu concentration. Fertilizing maternal plants during seed fill resulted in a significant increase of progeny germination rates, but had no influence on seedling vigor. Average germination rates were 67 and 61% from fertilized and non-fertilized plants, respectively. We found no evidence that root temperatures of 18 to 36°C during grain fill influenced soybean seed formation and development, nutrient concentration, germination and vigor. However, maternal plant nutrient stress during this stage can affect yield determinants, dry weights, seed nutrient concentration and germination rates.

Also, there were variations among soybean varieties on viability and vigour as well as among the environmental conditions. However, cold storage gave the best in preserving viability and vigour of soybean seed. The result of this work will give the farmers researchers and seed producers a better option of material and environmental condition to be used for storing seeds especially in the tropics, because seeds are frequently stored from one year to another.

Book excerpt: Soybean (Glycine max L.) is an important legume crop often exposed to heat and drought stresses during reproductive and early-seed setting stages, resulting in lower yields and seed quality. Ten soybean cultivars were phenotyped for individual (drought or heat) and combined stress tolerance. Under drought, reduced stomatal conductance and increased canopy temperature significantly reduced seed number (46%) and weight (35%). Heat stress alone reduced seed number (19%) and weight (23%) compared to control. Moreover, a degree increase in daytime temperature above 32 ℗ʻC during the reproductive stage reduced seed weight by 4% and 7% under well-watered and drought conditions, respectively. Seed protein was increased under drought, while it declined under heat and combined stress compared to control. In contrast, oil content showed the opposite trend. Weak correlations between phenotypic traits under individual and combined stress suggest that selecting cultivars for individual stress tolerance may not work under combined stress conditions.

Rapid and effective methods of screening large numbers of soybean lines for drought tolerance are urgently needed. Two experiments were conducted to evaluate the effects of drought in soybean during reproductive stages. In the first experiment five genotypes from maturity groups 2 through 5 were tested under well-irrigated and drought conditions. Beginning at R5, leaf samples were taken for nitrogen concentration analysis. Pictures were taken across the top of each plot to determine the intensity of greenness using the Dark Green Color Index (DGCI). Aerial photographs were also taken to determine aerial DGCI values. Leaf nitrogen concentration decreased as plants approached maturity and was closely related to ground DGCI. Additionally, ground DGCI and aerial DGCI values followed similar trends. The aerial DGCI measurements had advantages over ground DGCI measurements in that it allowed discernment between both water treatments. This opens up the possibility of using aerial DGCI to screen genotypes that senesce more slowly under drought. In the second experiment, the effects of drought in soybean were evaluated by aerial infrared image analysis, carbon isotope discrimination (Delta 13C) and oxygen isotope composition (delta 18O). Five fast-and five slow-wilting genotypes derived from a cross of Benning × PI416937 were evaluated under three water treatments that included a full and two deficit-irrigation treatments of increasing severity (deficit 1, and 2). After canopy closure, aerial infrared images were taken to determine the relative canopy temperature. Soybean leaves sampled at late R5 and seed at harvest were collected to measure Delta 13C (leaf and seed) and delta 18O (seed) as surrogate measurements for water use efficiency (WUE) and transpiration, respectively. As water availability decreased, the Delta 13C values from leaf and seed generally decreased (i.e., higher WUE). In contrast, the delta 18O values and relative canopy temperature generally increased with increasing drought stress. Moreover, slow-wilting genotypes generally had lower Delta 13C, delta 18O and canopy temperature than fast-wilting genotypes. However, delta 18O values were not consistent over years. The results from these two experiments indicate that the determination of DGCI, Delta 13C, and canopy temperature were promising tools for rapid characterization of drought-related traits in soybean.
