Book excerpt: Thirty years ago, the group of Baulieu and colleagues discovered that certain steroid hormones were present in higher amounts in the brain than in the plasma, and also found that suppression of circulating steroids by adrenalectomy and castration did not affect the concentration of pregnenolone, dehydroepiandrosterone and their sulfate esters in the rat brain. These seminal observations led to the concept that the brain, in very much the same way as the adrenal cortex, testis, ovary and placenta, was capable of synthesizing steroids. These brain born steroids, called neurosteroids, have been found to exert a vast array of biological activities. A number of steroidogenic enzymes have now been identified in the central nervous system by immunohistochemistry and in situ hybridization, and the neuronal and hormonal mechanisms regulating the biosynthesis of neurosteroids have been partially elucidated. The aim of this Research Topic is to celebrate three decades of research on neurosteroids by gathering a bouquet of review papers and original articles from leading scientists in the flourishing field of neurosteroids.

Its later history is well known, yet comparatively little is known about the city before it became Constantinople, and then Istanbul. Although it was just a minor Greek polis located on the northern fringes of Hellenic culture, surrounded by hostile Thracian tribes and denigrated by one ancient wit as the -armpit of Greece, - Byzantium did nevertheless possess one unique advantage--control of the Bosporus strait. This highly strategic waterway links the Aegean to the Black Sea, thereby conferring on the city the ability to tax maritime traffic passing between the two. Byzantium and the Bosporus is a historical study of the city of Byzantium and its society, epigraphy, culture, and economy, which seeks to establish the significance of its geographical circumstances and in particular its relationship with the Bosporus strait. Examining the history of the region through this lens reveals how over almost a millennium it came to shape many aspects of the lives of its inhabitants, illuminating not only the nature of economic exploitation and the attitudes of ancient imperialism, but also local industries and resources and the genesis of communities' local identities. Drawing extensively on Dionysius of Byzantium's Anaplous Bosporou, an ancient account of the journey up the Bosporus, and on local inscriptions, what emerges is a meditation on regional particularism which reveals the pervasive influence that the waterway had on the city of Byzantium and its local communities and illustrates how the history of this region cannot be understood in isolation from its geographical context. These alterations can be drug class-specific, and are not confined to neurons, as drugs of abuse also induce molecular and cellular alterations in various glial cell types such as astrocytes and microglia. The phenomenon of drug-induced plasticity includes changes in dendritic branching and architecture, dendritic spine density and morphology, astrocyte-neuronal interactions, dysregulation of glutamatergic and GABAergic signaling, and alterations in myelination or microglial phenotype. This drug-induced "rewiring" of the brain at numerous levels can contribute to the development, maintenance, and persistence of the addicted state, as well as associated deficits in normal cognitive functioning. The aim of this Research Topic is to collect recent and important findings related to the structural alterations produced by drug of abuse in neurons, glial, and other cell types of the central nervous system.
