Book excerpt: This report considers the biological and behavioral mechanisms that may underlie the pathogenicity of tobacco smoke. Many Surgeon General's reports have considered research findings on mechanisms in assessing the biological plausibility of associations observed in epidemiologic studies. Mechanisms of disease are important because they may provide plausibility, which is one of the guideline criteria for assessing evidence on causation. This report specifically reviews the evidence on the potential mechanisms by which smoking causes diseases and considers whether a mechanism is likely to be operative in the production of human disease by tobacco smoke. This evidence is relevant to understanding how smoking causes disease, to identifying those who may be particularly susceptible, and to assessing the potential risks of tobacco products.

This book was released on 2019-10-18 with total page 429 pages. Available in PDF, EPUB and Kindle. Book excerpt: CD1 and MR1 are major histocompatibility complex (MHC) class I-related proteins that bind and present non-peptide antigens to subsets of T cells with specialized functions. CD1 proteins typically present lipid antigens to CD1-restricted T cells, whereas MR1 presents vitamin B-based ligands and a variety of drugs and drug-like molecules to MR1-restricted T cells. The CD1 family of antigen presenting molecules has been divided into two groups: Group 1 contains CD1a, CD1b and CD1c, and Group 2 contains CD1d. Additionally, CD1e is expressed intracellularly and is involved in the loading of lipid antigens onto Group 1 CD1 proteins. Humans express both Groups 1 and 2 CD1 proteins, whereas mice only express CD1d. Group 1 CD1 proteins present lipid antigens to T cells that generally express diverse T cell receptors (TCRs) and exhibit adaptive-like functions, whereas CD1d presents lipid antigens to subsets of T cells that express either diverse or highly restricted TCRs and exhibit innate-like functions. CD1d-restricted T cells are called natural killer T (NKT) cells, which includes Type I or invariant NKT (iNKT) cells expressing semi-invariant TCRs, and Type II NKT cells expressing more diverse TCRs. CD1-restricted T cells have been implicated in a wide variety of diseases, including cancer, infections, and autoimmune, inflammatory and metabolic diseases. Additionally, NKT cells have been targeted for immunotherapy of disease with ligands such as α-galactosylceramide for iNKT cells, or sulfatide for Type II NKT cells. Like iNKT cells, MR1-restricted T cells express semi-invariant TCRs and display innate-like functions. MR1-restricted T cells, also called mucosal-associated invariant T (MAIT) cells, have been implicated in immune responses against a variety of pathogens such as Mycobacterium tuberculosis, Pseudomonas aeruginosa, Helicobacter pylori, hepatitis C virus and influenza virus. Moreover, these cells contribute to autoimmune and inflammatory diseases, including colitis, rheumatoid arthritis, psoriasis, lupus, and diabetes.

While FFA have long been considered byproducts of cell metabolism, they are now recognized as ligands that regulate cell and tissue function via G-protein-coupled receptors. At least three receptors have been identified for which FFA appear to be the endogenous ligands.

Available in PDF, EPUB and Kindle. Book excerpt: Even though initially considered as a passive means for storing energy, lipids are now regarded as multifaceted molecules with crucial structural and functional activities. For instance, some of them play essential roles as key components of cell membranes whereas others act as signaling molecules in the regulation of cell homeostasis. In recent years, lipid research has attracted increasing interest because of the involvement of this class of compounds in human health. Indeed, a plethora of pathological conditions are characterized by alterations in lipid metabolism, such as cardiovascular diseases and brain disorders. This Special Issue is a collection of papers from different experts in lipid research, with the aim of providing new insights into the physiopathological involvement of lipids and their impact on human health. This collection also demonstrates the usefulness of interdisciplinary approaches in the development of novel methods to study and manipulate lipid metabolism, which may represent an attractive target for designing effective therapeutic strategies to counteract numerous pathologies.

Available in PDF, EPUB and Kindle. Book excerpt: Many physiological conditions such as host defense or aging and pathological conditions such as neurodegenerative diseases, and diabetes are associated with the accumulation of high levels of reactive oxygen species and reactive nitrogen species. This generates a condition called oxidative stress. Low levels of reactive oxygen species, however, which are continuously produced during aerobic metabolism, function as important signaling molecules, setting the metabolic pace of cells and regulating processes ranging from gene expression to apoptosis. For this book we would like to recruit the experts in the field of redox chemistry, bioinformatics and proteomics, redox signaling and oxidative stress biology to discuss how organisms achieve the appropriate redox balance, the mechanisms that lead to oxidative stress conditions and the physiological consequences that contribute to aging and disease.

Available in PDF, EPUB and Kindle. Book excerpt: The PI3Ks control many key functions in immune cells. PI3Ks phosphorylate PtdIns(4,5)P2 to yield PtdIns(3,4,5)P3. Initially, PI3K inhibitors such as Wortmannin, LY294002 and Rapamycin were used to establish a central role for Pi3K pathway in immune cells. Considerable progress in understanding the role of this pathway in cells of the immune system has been made in recent years, starting with analysis of various PI3K and Pten knockout mice and subsequently mTOR and Foxo knockout mice. Together, these experiments have revealed how PI3Ks control B cell and T cell development, T helper cell differentiation, regulatory T cell development and function, B cell and T cell trafficking, immunoglobulin class switching and much, much more. The PI3Kd inhibitor idelalisib has recently been approved for the treatment of B cell lymphoma. Clinical trials of other PI3K inhibitors in autoimmune and inflammatory diseases are also in progress. This is an opportune time to consider a Research Topic considering when what we have learned about the PI3K signalling module in lymphocyte biology and how this is making an impact on clinical immunology and haematology.

Book excerpt: This book presents state of the art knowledge on severe asthma with the aim of providing readers with a clear understanding of, first, the heterogeneity of the condition and of patients’ symptom profiles and responses to therapy and, second, the future implications of this heterogeneity for individualized patient care. After an opening section that offers an overview of severe asthma, including its clinical significance, the pathogenesis, available diagnostic approaches, and treatment options are described in detail. The sections on diagnosis and treatment cover the role of biomarkers, the use of radiologic diagnostic modalities, and both pharmacologic and non-pharmacologic therapies, including emerging options that will address hitherto unmet needs of patients. The outcomes of cutting-edge preclinical and clinical research are carefully documented and numerous useful tips provided on patient management. The inclusion of many informative schematic figures will assist readers in grasping the contents easily. The book will be of high value for medical students, researchers, general physicians, specialists, and paramedical staff.
