Diphonix Resin (Eichrom Industries), is used to pre-concentrate the actinides from digested fecal samples. A rapid microwave digestion technique is used to remove the actinides from the Diphonix Resin, which effectively extracts plutonium and americium from acidic solutions containing hydrofluoric acid. After resin digestion, the plutonium and americium are recovered in a small volume of nitric acid that is loaded onto small extraction chromatography columns, TEVA Resin and TRU Resin (Eichrom Industries). The method enables complete dissolution of plutonium oxide and provides high recovery of plutonium and americium with good removal of thorium isotopes such as thorium-228.

The new method separates plutonium, neptunium, uranium, americium and strontium-90 with high chemical recovery and excellent thorium removal. The method uses calcium phosphate precipitation and stacked TEVA Resin(R) and TRU Resin(R) cartridges to separate and purify the actinides. Plutonium and neptunium are separated on TEVA Resin(R), while uranium and americium are simultaneously retained and separated on TRU Resin(R). Plutonium-236 tracer can be used to allow simultaneous separation and measurement of both plutonium and neptunium using TEVA Resin(R). Strontium-90 can also be separated on Sr Resin(R) by evaporating and redissolving load and rinse solutions collected from the TEVA/TRU column and separating strontium on Sr-Resin(R). Fast flow rates are achieved by using small particle size resin cartridges and a vacuum box separation system that will separate 24 samples at a time. This unique approach can be used with urine samples because iron is not present at significant levels in urine and plutonium reduction is accomplished without adding iron (II) to the sample. The advantage of this approach is that actinides can be loaded onto two separate resins in a single load step with simultaneous extraction and assay of neptunium and plutonium with high chemical recovery and excellent removal of matrix interferences.

Book excerpt: A rapid gas-liquid chromatographic (GLC) method was developed for the confirmation of benzoylecgonine (BE) positive urine samples screened by the enzyme multiplied immunoassay technique (EMIT®) assay. The procedure is performed by solvent extraction of BE from 0.1 or 0.2 mL of urine, followed by an aqueous wash of the solvent and evaporation. The dried residue was derivatized with 50 μL of pentafluoropropionic anhydride and 25 μL of pentafluoropropropanol at 90°C for 15 min. The derivatizing reagents were evaporated to dryness, and the derivatized BE, and cocaine if present, were reconstituted and injected into the gas chromatograph. The column was a 15-m by 0.2-mm fused silica capillary column, coated with 0.25 μm of DB-1, terminating in a nitrogen phosphorus detector (NPD) Cocaine and the pentafluoro BE derivatives retention times were 3.2 and 2.6 min, respectively. Nalorphine was used as reference or internal standard with a retention time of 4.78 min. The complete procedure can be performed in approximately 1.5 h. The EMIT cutoff between positive and negative urine samples is 300 ng/mL of BE. The lower limit of sensitivity of this method is 25 ng of BE extracted from urine. Validation studies resulted in confirmation of 101 out of 121 EMIT cocaine positive urine samples that could not be confirmed by thin-layer chromatography (TLC) This represents 84% confirmation efficiency.
