The successfully synthesized ZnO powder from the sol-gel centrifugation and sol-gel storage methods were characterized and investigated by X-ray diffraction, field emission scanning electron microscopy, transmission electron microscopy, Fourier-transform infrared spectroscopy, UV–visible spectroscopy, and photoluminescence test to compare the properties of the nanoparticles. The best characteristic of the ZnO powder from both methods was observed when the powders were coated on an ITO glass to fabricate a PEC. The current density–voltage performances of both PECs were investigated under luminescent and dark conditions.

Book excerpt: The clinical incidence of large-scale bone defects has increased over the past few decades to both an increasingly aging population and an associated rise in the number of traumatic injuries. These defects require surgical intervention and can be complicated by post-operative surgical site infections; while these infections were originally managed with intravenous antibiotics, overuse of antibiotic drugs in both livestock and hospital environments has contributed to the creation of antibiotic resistant pathogens such as methicillin resistant Staphylococcus aureus. In craniofacial applications, especially, the lack of a synthetic, biocompatible and osteoconductive graft material that could address infections via a non-pharmaceutical approach provided a niche for exploration. The objective of this study was to synthesize a novel glass series, characterize the series to determine whether it would be biologically feasible and if so, identify the antibacterial efficacy and cytocompatibility of the material. A novel Zn-based bioactive glass series was created, as Zn has proven antibacterial capacity in previous studies. Molten mixtures of SiO2, Na2O, SrO, CaO and ZnO with varying CaO and ZnO concentrations, from 0-30% Zn, were quenched in room temperature water and ground to dental cement standards of sub-45 microns. X-ray diffraction revealed broad humps for each composition indicating that each was an amorphous solid. Scanning electron microscopy, energy dispersive spectroscopy and particle size analyses validated that the particle size of each composition was 45 microns, containing a uniform distribution of both large and small particles of the appropriate elemental compositions. BET surface area analysis confirmed that the surface areas of the samples were between 0.4163 m2/g and 0.6671 m2/g from RC-Control to RC-3; similarity in surface areas implied that reactivity differences were likely attributable to other factors. Differential thermal analysis indicated that the glass transition temperature of the series dropped from 635 degrees C to 578 degrees C from RC-Control to RC-3, indicating an increase in reactivity with Zn concentration. This was validated by MAS-NMR where samples shifted from -77ppm for RC-Control, indicating a tendency towards Q1, to -72.1ppm for RC-3, indicating Q0 tendency. The reactivity of the samples was high enough to indicate potential as a bioactive material, and a cement was synthesized with glass powder, polyacrylic acid and DI water at a ratio of 1.3:1 and optimized to working times of ~5min for each glass composition. Co-culture of E. coli with glass powders and subsequent plating onto LB agar revealed that significant antibacterial efficacy was possible for 20% and 30% Zn samples, RC-2 and RC-3. This trend was corroborated when zones of inhibition were measured for cement disks in S. aureus plates. RC-3 had a notably greater antibacterial capacity, but despite this, in a mammalian cell environment with murine MC3T3 pre-osteoblastic cells, showed no cytotoxicity. The antibacterial capacity and cytocompatibility of the RC series is promising and further exploration of RC-3 as an antibacterial bone graft substitute should be conducted.

The rate at which these glasses generate a response, or their reactivity, is an important parameter for healing efficacy. Owing to their tetrahedral coordination, silicate-based glasses exhibit slow and incomplete solubility, which limit their application in soft tissue repair. Therefore, there is great interest in developing more soluble and reactive bioactive glass formulations based on the borate network forming oxide, which can be both trigonally and tetrahedrally coordinated. It has been shown that borate-based glasses have higher reactivity than silicate-based glasses attributed to their glass network structure. Limitations in conventional methods such as the lack of accuracy and time-consumption to examine reactivity/bioactivity and surface properties of bioactive and soluble glasses have led to an increased interest in finding and investigating new techniques, such as dynamic vapour sorption (DVS) and inverse gas chromatography (IGC). It was demonstrated that DVS has the ability to measure and predict the bioactivity/reactivity of silicate-, phosphate- and borate- based glasses with various specific surface areas/particle sizes or formulations. It was shown that DVS was able to measure and correlate the vapour isotherm values of melt-quench derived Bioglass® 45S5: (46.1)SiO2-(26.9)CaO-(24.4)Na2O-(2.6)P2O5 (mol %) of three different particle sizes (surface areas) with the reactivity, bioactivity and ions release rates of glasses. Higher surface area led to the higher vapour sorption and reactivity. Additionally, IGC was utilized to examine the surface properties of melt-quench derived phosphate glasses (PGs) doped with both SiO2 and TiO2 (50P2O5-40CaO-xSiO2-(10 − x) TiO2, where x = 7, 5, 3, and 0 mol%). It was shown that IGC was more accurate and sensitive than other conventional techniques such as contact angle measurement in determining the effect of silica content on the surface properties of PGs. In conclusion, this dissertation has assessed the potential complementarity of DVS and IGC in the characterization of aqueous reactivity and surface properties of bioactive and soluble glasses.Moreover, complications in the healing of chronic wounds in patients who are at constant risk of infections have led to a great demand for the development of biomaterial based wound dressing replacement. Borate-based glasses have recently been approved for wound repair which are melt-quench derived, yet borate glasses fabricated through the sol-gel process have the benefits of higher dissolution rate, which could potentially lead to the more rapid wound repair. However, the potential of sol-gel derived borate glasses, in particular, silver doped borate-based glasses (AgBGs) for soft tissue repair has so far not been investigated. In this thesis, anti-bacterial AgBGs in the B2O3Ag2O-CaO-P2O5 system were fabricated through optimization of sol-gel process and glass composition. Silver ion release rate and anti-bacterial activity were correlated and demonstrated dose dependent efficacy against Escherichia coli or Staphylococcus aureus and Pseudomonas aeruginosa bacteria. In addition, AgBGs were systematically examined to determine their effects on cellular functions such as metabolic activity, cell viability and migration through in vitro cell culture of human derived keratinocyte (HaCat) and mouse derived fibroblastic (NIH/3T3) cell lines.
