L'assemblage des centres Fe-S et leur insertion dans des protéines acceptrices requièrent l'activité d'une machinerie protéique dédiée. Bien que les protéines de la biogenèse des centres Fe-S soient conservées, plusieurs aspects fonctionnels et mécanistiques restent inconnus. Notre travail de thèse a consisté à caractériser les protéines mammifères de type A, ISCA1 et ISCA2, qui sont impliquées dans la biogenèse mitochondriales des centres Fe-S. En utilisant une approche couplant l'immunoprécipitation avec une analyse protéomique par spectrométrie de masse, plusieurs interactions protéiques d'ISCA1 et ISCA2 ont pu être identifiées. En plus d'une interaction entre ISCA1 et ISCA2, nous avons ainsi montré l'existence d'interactions spécifiques à chacune de ces protéines. Une approche de knockdown dans la souris via l'injection de virus adéno-associés, a permis de montrer l'absence de redondance fonctionnelle entre ISCA1 et ISCA2 puisque seul ISCA1 se trouve être nécessaire dans la maturation d'une catégorie de protéines à centre Fe-S.

Book excerpt: Abstract: Members of the IscU family of proteins are among the most conserved of all protein groups, extending across all three kingdoms of life. IscU is believed to be involved in iron-sulfur cluster delivery to apo iron-sulfur proteins. However, most of the evidence supporting the function of IscU stems from genetic and cellular biological studies. Therefore, we set out to biochemically characterize human, yeast, and prokaryotic IscU proteins. A variety of spectroscopic techniques were used to evaluate IscU including, UV-visible absorption, Mössbauer, near- and far- UV circular dichroism, mass spectrometry, atomic absorption, and nuclear magnetic resonance. Herein we demonstrate that IscU proteins coordinate reductively labile [2Fe-2S]2 centers and are capable of mediating delivery of intact cluster to apo protein targets. Furthermore, extensive structural and dynamic data of a hyperthermophilic homologue, Thermotoga maritima IscU, revealed that IscU adopts a mobile molten globule-like state that is vastly different from the previously identified ferredoxin-like fold that has thus far been characterized for other metallochaperones. Such a dynamic molecule may allow for the flexibility that is necessary for the multiple roles of Fe-S cluster assembly, and recognition and delivery of that cluster to a target protein. Additionally, we utilized X-ray crystallography to elucidate a high resolution structure of an oxygen ligated [4Fe-4S] high potential iron protein.

Book excerpt: Iron is the second most abundant metal on earth, and therefore it is not surprising that throughout the kingdoms of life iron is a vital metal in biological processes. A common form of iron that is conserved throughout all organisms is the iron-sulfur cluster. Examples include the [2Fe-2S] and [4Fe-4S] clusters that bind to a number of proteins with specific Fe-S cluster binding sites. Such sites normally consist of four cysteine residues, with occasional replacement of Cys with histidine or aspartate. Iron-sulfur clusters have been connected to several functions in the body, including metabolite synthesis, metabolism, DNA synthesis, biodegradation, and isomerization. The biosynthesis and trafficking of [2Fe-2S] clusters has been studied extensively within the mitochondria, but mysteries remain concerning the cellular chemistry of Fe-S clusters, particularly the cellular formation of [4Fe-4S] clusters and the relationship and possible trafficking of [2Fe-2S] clusters between mitochondria and cytosol of the cell. One of the largest families of proteins, the S-adenosylmethionine (SAM) family, binds a [4Fe-4S] cluster. Lipoyl synthase (LIAS) is a member of the radical S-adenosylmethionine (SAM) superfamily that catalyzes the final step of lipoic acid biosynthesis. The enzyme contains two [4Fe-4S] centers (reducing and auxiliary clusters) that promote radical formation and sulfur transfer, respectively. Most information concerning LIAS and its mechanism has been determined from prokaryotic enzymes. We have made progress in expressing, isolating and characterizing human LIAS, its reactivity, and evaluation of natural iron-sulfur (Fe-S) cluster reconstitution mechanisms. Cluster donation by a number of possible cluster donor proteins and heterodimeric complexes has also been evaluated. [2Fe-2S]-Cluster-bound forms of human ISCU and ISCA2 were found capable of reconstituting human LIAS such that complete product turnover was enabled for LIAS, as monitored via a liquid chromatography-mass spectrometry (LC-MS) assay. EPR studies of native LIAS and substituted derivatives that lacked the ability to bind one or the other of LIAS’s two [4Fe-4S] clusters revealed a likely order of cluster addition, with the auxiliary cluster preceding the reducing [4Fe-4S] center. These results detail the trafficking of Fe-S clusters in human cells and highlight differences with respect to bacterial LIAS analogs. Likely in vivo Fe-S cluster donors to LIAS are identified, with possible connections to human disease states, and a mechanistic ordering of [4Fe-4S] cluster reconstitution is evident. Further studies into the trafficking proteins, ISCA1 and ISCA2 also revealed novel cellular pathways with implications for assembly of [4Fe-4S] centers in cluster proteins. While the metabolic enzyme aconitase could not be reconstituted by ISCA1, ISCA2, or an ISCA1/ISCA2 heterodimer, both ISCA1 and ISCA2 could receive cluster from several trafficking proteins, including ISCU, GLRX5, and Nfu, as confirmed using in vitro CD transfer experiments. Aconitase was readily reconstituted by ISCU, GLRX5, and Nfu. Investigation of the Chaetomium thermophilum (C. thermophilum) protein, Atm1p, also provided insight on an ABC transporter protein implicated in transporting a glutathione-bound [2Fe-2S] cluster across the plasma membrane. Atm1 is a protein that is conserved throughout numerous organisms including humans, and has been linked to various disease states; most notably, sideroblastic anemia. By use of site-directed mutagenesis, and substituting multiple residues in the protein connected to ATPase activity and substrate binding, it was found that the protein was still able to transport the [2Fe-2S] glutathione cluster across a proteoliposome, but the ability to bind the [2Fe-2S] cluster impacted the ability to bind and consume ATP, indicating that residues outside the substrate binding pocket are involved in mediating conformational changes between the substrate binding pocket and nucleotide binding domain.

Among these cofactors are the molybdenum (Moco) and tungsten (Wco) cofactors. Both Moco/Wco biosynthesis and Fe-S cluster assembly are highly conserved among all kingdoms of life. After formation, Fe-S clusters are transferred to carrier proteins, which insert them into recipient apo-proteins. Moco/Wco cofactors are composed of a tricyclic pterin compound, with the metal coordinated to its unique dithiolene group. Moco/Wco biosynthesis starts with an Fe-S cluster-dependent step involving radical/S-adenosylmethionine (SAM) chemistry. The current lack of knowledge of the connection of the assembly/biosynthesis of complex metal-containing cofactors is due to the sheer complexity of their synthesis with regard to both the (genetic) regulation and (chemical) metal center assembly. Studies on these metal-cofactors/cofactor-containing enzymes are important for understanding fundamental cellular processes. They will also provide a comprehensive view of the complex biosynthesis and the catalytic mechanism of metalloenzymes that underlie metal-related human diseases.
