Book excerpt: There have been very few developments that markedly affect the need to greatly revise the text from the last version of this book. This is testament to the fact that hetero- neous enzyme-linked immunosorbent assays (ELISA) provide ideal systems for dealing with a wide range of studies in many biological areas. The main reason for this success is test flexibility, whereby reactants can be used in different combinations, either attached passively to a solid phase support or in the liquid phase. The exploitation of the ELISA has been increased through continued development of specifically produced reagents, for example, monoclonal and polyclonal antibodies and peptide antigens coupled with the improvement and expansion of commercial products such as enzyme-linked conjugates, substrates and chromogens, plastics technology and design of microwell plates, inst- mentation advances and robotics. However, the principles of the ELISA remain the same. There has been some rearrangement of chapters plus addition of three new ones dealing with charting methods for assessing the indirect ELISA, ruggedness and robustness of tests-aspects of kit use and validation, and internal quality control and external quality management of data, respectively. These reflect the need to control what you are doing with ELISA and to exploit the method to its full extent. I do not apologize for dealing with the same areas in different ways a number of times, as it is imperative that principles are understood to allow planning, operation, and control of ELISA.

Available in PDF, EPUB and Kindle. Book excerpt: This book offers comprehensive information on all aspects of ELISA, starting with the fundamentals of the immune system. It also reviews the history of analytical assays prior to the advent of ELISA (enzyme-linked immunosorbent assay) and addresses the materials of choice for the fabrication of the platforms, possible biomolecular interactions, different protocols, and evaluation parameters. The book guides readers through the respective steps of the analytical assay, while also familiarizing them with the possible sources of error in the assay. It offers detailed insights into the immobilization techniques used for protein attachment, as well as methods for evaluating the assay and calculating the key parameters, such as sensitivity, specificity, accuracy and limit of detection. In addition, the book explores the advantages and shortcomings of the conventional ELISA, as well as various approaches to improving its performance. In this regard, merging and integrating other technologies with widely known ELISAs have opened new avenues for the advancement of this immunoassay. Accordingly, the book provides cutting-edge information on integrated platforms such as ELISpot, plasmonic ELISAs, sphere-/bead-based ELISAs, paper-/fiber-based ELISAs and ELISA in micro-devices.

Book excerpt: This volume is a practical biochemical guide to the Enzyme-Linked Immunosorbent Assay (ELISA), used to detect a target substance in a liquid sample. The ELISA is an important and widely used diagnostic tool in medicine, animal health, botany and quality assurance processes in food and beverage production. An introductory chapter orients the reader on the basic structure and function of immunoglobulins and their fragments while subsequent chapters outline the methodology to generate monoclonal antibodies using hybridoma technology and the general methods used to purify antibodies. Multiple chapters demonstrate how to creatively use the properties of the antibody to identify, localize and quantify target analytes to answer questions and resolve problems. The reader will learn how to use a variety of immunoassay strategies, reporters and detection systems that will undoubtedly facilitate their efforts to gain answers to their own questions. Written in the successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible protocols and notes on troubleshooting and avoiding known pitfalls. Authoritative and easily accessible, ELISA: Methods and Protocols seeks to provide both professionals and novices with the technical information necessary for the reader to successfully use the immunoassay as part of the discovery process.

One of the essential goals of this book is to delineate vital information about different types of ELISA and their applications. The beginning chapter describes the history and background of enzyme-linked immunosorbent assay (ELISA), as well as the remarkable changes that were brought about by this technique. The chapter also describes the commercial application techniques used in food and diagnostic industries. The second chapter, on the other hand, describes various types of ELISA and their steps to perform ELISA. The third chapter discusses the practical applications and their mechanisms from theory. It also discusses possible multiple sources of error which can exist at each stage of the process; how to minimize the error; and the principles of data analysis. The fourth chapter discusses the applications of ELISA in various fields, which also include the uses of ELISA in the diagnosis of infectious diseases, parasitic diseases, and non-communicable disease caused by a variety of pathogenic microorganisms. While Chapters One to Four describe a detailed history, a variety of ELISA, their steps, their sources of application error, their solutions and their applications in different fields, the remaining chapters discuss more specific issues, including: The role of ELISA in advanced glycerin end products and their cell surface receptors in metabolic diseases, indirect ELISA for the diagnosis of Brucella Canis, ELISA based detection in cancer research, etc.

Available in PDF, EPUB and Kindle. Book excerpt: From an expert in the field comes the definitive guide to managing breast cancer in the information age—a comprehensive resource for diagnosis, treatment, and peace of mind. The breast cancer cure rate is at an all-time high, and so is the information, to say nothing of the misinformation, available to patients and their families. Online searches can lead to unreliable sources, leaving even the most resilient patient feeling uneasy and uncertain about her diagnosis, treatment options, doctors, side effects, and recovery. Adding to a patient’s anxiety is input from well-meaning friends and family, with stories, worries, and opinions to share, sometimes without knowing the details of her particular case, when in reality breast cancer treatment has gone well beyond a “one size fits all” approach. Elisa Port, MD, FACS, chief of breast surgery at The Mount Sinai Hospital and co-director of the Dubin Breast Center in Manhattan, offers an optimistic antidote to the ocean of Web data on screening, diagnosis, prognosis, and treatment. Inside you’ll discover • the various scenarios when mammograms indicate the need for a biopsy • the questions to ask about surgery, chemotherapy, radiation, and breast reconstruction • the important things to look for when deciding where to get care • the key to deciphering complicated pathology reports and avoiding confusion • the facts on genetic testing and the breast cancer genes: BRCA-1 and BRCA-2 • the best resources and advice for those supporting someone with breast cancer From innovations in breast cancer screening and evaluating results to post-treatment medications and living as a breast cancer survivor, Dr. Elisa Port describes every possible test and every type of doctor visit, providing a comprehensive, empathetic guide that every newly diagnosed woman (and her family) will want to have at her side. Praise for The New Generation Breast Cancer Book “One book you need . . . If you’re considering your options for treatment or know someone who is, this step-by-step guide, The New Generation Breast Cancer Book, is essential reading.”—InStyle “Elisa Port, M.D., is the doctor every patient deserves: brilliant and compassionate. Her book will be a sanity saver and, quite possibly, a life saver.”—Geralyn Lucas, author of Why I Wore Lipstick to My Mastectomy “As up-to-date as one can get, with lots to offer people facing a cancer diagnosis or hoping to support someone with the disease.”—Library Journal (starred review) “The New Generation Breast Cancer Book helps you sort through all the information you’ve gathered, clarify the terminology, consider the options, and make the right decisions for your unique case.”—Edie Falco “A lifeline for many women in need of today’s most up-to-date choices for treatment . . . Everyone should read this book for themselves, their mothers, grandmothers, daughters, and friends.”—Kara DioGuardi, Grammy-nominated songwriter, music executive, and Arthouse Entertainment co-founder “The book is teeming with easy-to-understand medical explanations, tips, takeaways, and pro-and-con discussions of various courses of action. Port also includes two extremely useful appendices that respectively take on common myths and answer questions frequently asked by friends and family. This is a vital read that will empower men and women alike.”—Publishers Weekly
