Book excerpt: Fluorescence and phosphorescence are proving to be extremely sensitive probes for elucidating conformation of proteins and nucleic acids and for studying molecular interactions. Newer instrumentation and techniques hold forth great promise for the future of these luminescence methods in biopolymer research. It must be noted, however, that the discovery that certain amino acids, purines, and pyrimidines emit fluorescence or phosphorescence is relatively recent, occurring within the last decade. Professor Konev is one of the pioneers in the application of these procedures to biopolymers and is highly qualified to write about this subject. This book, though written largely as a monograph of the author's own contributions, is also an excellent review of the subject. Of particular interest are the references to many important Russian papers in this field which have not been recognized in the Western literature. It is apparent from this book that fluorescence and phosphorescence methods are being used about as widely in Russia as elsewhere in the world and that we must not overlook these im portant contributions. Konev's studies on protein fluorescence have been widely recognized. It is of interest to learn about these and other of his applications. The last part of the book, which deals with fluorescence as a means to probe into the structure and conforma tion of macromolecules in intact cells, is most interesting. Aside from published symposia this book is the first written specifically about luminescence of biopolymers. Sidney Udenfriend Bethesda. Available in PDF, EPUB and Kindle. Book excerpt: Many physiological conditions such as host defense or aging and pathological conditions such as neurodegenerative diseases, and diabetes are associated with the accumulation of high levels of reactive oxygen species and reactive nitrogen species. This generates a condition called oxidative stress. Low levels of reactive oxygen species, however, which are continuously produced during aerobic metabolism, function as important signaling molecules, setting the metabolic pace of cells and regulating processes ranging from gene expression to apoptosis. For this book we would like to recruit the experts in the field of redox chemistry, bioinformatics and proteomics, redox signaling and oxidative stress biology to discuss how organisms achieve the appropriate redox balance, the mechanisms that lead to oxidative stress conditions and the physiological consequences that contribute to aging and disease.

Book excerpt: Fluorescence in situ hybridization (FISH) has been developed as a powerful technology which allows direct visualisation or localisation of genomic alterations. The technique has been adopted to a range of applications in both medicine, especially in the areas of diagnostic cytogenetics, and biology. Topics described in this manual include: FISH on native human tissues, such as blood, bone marrow, epithelial cells, hair root cells, amniotic fluid cells, human sperm cells; FISH on archival human tissues, such as formalin fixed and paraffin embedded tissue sections, cryofixed tissue; simultaneous detection of apoptosis and xpression of apoptosis-related genes; comparative genomic ybridization; and special FISH techniques.

Book excerpt: John Walker and Ralph Rapley have collected a wide-ranging group of molecular and biochemical techniques that are the most frequently used in medical and clinical research, especially diagnostics. The authors-well-established investigators who run their own research programs and use the methods on a regular basis-outline the practical procedures for using them and describe a variety of pertinent applications. Among the technologies presented are southern and western blotting, electrophoresis, PCR, cDNA and protein microarrays, liquid chromatography, in situ hybridization, karyotyping, flow cytometry, bioinformatics, genomics, and ribotyping. The applications include assays for mutation detection, mRNA analysis, chromosome translocations, inborn errors of metabolism, protein therapeutics, and gene therapy.

Book excerpt: In Confocal Microscopy Methods and Protocols, Stephen Paddock and a highly skilled panel of experts lead the researcher using confocal techniques from the bench top, through the imaging process, to the journal page. They concisely describe all the key stages of confocal imaging-from tissue sampling methods, through the staining process, to the manipulation, presentation, and publication of the realized image. Written in a user-friendly, nontechnical style, the methods specifically cover most of the commonly used model organisms: worms, sea urchins, flies, plants, yeast, frogs, and zebrafish. Centered in the many biological applications of the confocal microscope, the book makes possible the successful imaging of both fixed and living specimens using primarily the laser scanning confocal microscope. The powerful hands-on methods collected in Confocal Microscopy Methods and Protocols will help even the novice to produce first-class cover-quality confocal images.
