Book excerpt: The ability to monitor allergic inflammation has improved considerably in recent years due to the identification of numerous factors involved in the regulatory network of inflammatory response. As research has progressed, it has become clear that the subject is more complex than was originally believed, and a symposium was organized to bring together leading experts in the area and to review current knowledge. This book contains reports that were presented at the meeting and includes discussions of human B cell activation, cytokines and anticytokines, neuropeptides, asthma, cellular and molecular processes, and the roles of mast cells, basophils, eosinophils, neutrophils, macrophages, and platelets in inflammation. It will be essential reading for all those engaged in research in immunology, allergy, and asthma. Research level Symposium proceedings covering important topics in: Allergy Inflammation Asthma

Book excerpt: Over the past 20 years, public concerns have grown in response to the apparent rising prevalence of food allergy and related atopic conditions, such as eczema. Although evidence on the true prevalence of food allergy is complicated by insufficient or inconsistent data and studies with variable methodologies, many health care experts who care for patients agree that a real increase in food allergy has occurred and that it is unlikely to be due simply to an increase in awareness and better tools for diagnosis. Many stakeholders are concerned about these increases, including the general public, policy makers, regulatory agencies, the food industry, scientists, clinicians, and especially families of children and young people suffering from food allergy. At the present time, however, despite a mounting body of data on the prevalence, health consequences, and associated costs of food allergy, this chronic disease has not garnered the level of societal attention that it warrants. Moreover, for patients and families at risk, recommendations and guidelines have not been clear about preventing exposure or the onset of reactions or for managing this disease. Finding a Path to Safety in Food Allergy examines critical issues related to food allergy, including the prevalence and severity of food allergy and its impact on affected individuals, families, and communities; and current understanding of food allergy as a disease, and in diagnostics, treatments, prevention, and public policy. This report seeks to: clarify the nature of the disease, its causes, and its current management; highlight gaps in knowledge; encourage the implementation of management tools at many levels and among many stakeholders; and delineate a roadmap to safety for those who have, or are at risk of developing, food allergy, as well as for others in society who are responsible for public health.

The concept of asthma being an "allergic disorder only" has dominated above others to an extent, that a re-evaluation of this issue in the light of very recent results from research in immunopathology seemed more than overdue. This volume provides a current concept of theory and clinical practice of immunopathology and therapy of asthma, comprising extensive and brand new laboratory data on atopy, intrinsic asthma, aspects of autoimmunology, as well as guidelines for allergen avoidance, immunotherapy and - last not least - pharmacological treatment of eosinophilic inflammation.

Available in PDF, EPUB and Kindle. Book excerpt: Erika Jensen-Jarolim and Manuel L. Penichet 1. 1 Background Infectious diseases, being the major burden in the history of mankind worldwide th until the beginning of the 20 century, were important triggers in the understanding of immunological mechanisms. In contrast to infectious diseases, reports of all- gies and cancers were less common, but increased tremendously within the last century. Based on the US mortality data of the National Center for Health Statistics, Centers for Disease Control and Prevention 2009, a recent report from the American Cancer Society indicated that the number of cancer deaths increased approximately from 100,000 to 550,000 per year between 1930 and 2006, paralleling the increase of the total population during this period. Leading causes of death from cancer are lung and bronchus cancer, in men prostate cancer, and in women breast c- cer [1, 2]. Normalization to population size shows that the cancer death rate for most malignancies has been generally stable, although the mortality rate of certain malignancies, such as lung and bronchus cancer, has increased over the last 50 years [1-3]. In allergy, the situation is less clear, because for the time period around the turn of th the 19 century, only imprecise information is available. However, within the last 30 years the incidences of allergies has doubled not only in industrial countries, but in developing countries as well [4].

In the last 10 years, substantial progress has been made in terms of understanding some of the mechanisms that drive severe asthma; there have also been concomitant advances in the recognition of specific molecular phenotypes. This ERS Monograph covers all aspects of severe asthma – epidemiology, diagnosis, mechanisms, treatment and management – but has a particular focus on recent understanding of mechanistic heterogeneity based on an analytic approach using various ‘omics platforms applied to clinically well-defined asthma cohorts. How these advances have led to improved management targets is also emphasised. This book brings together the clinical and scientific expertise of those from around the world who are collaborating to solve the problem of severe asthma.

Available in PDF, EPUB and Kindle. Book excerpt: This book explains how stress – either psychological or physical – can activate and/or paralyse human innate or adaptive immunity. Adequate immunity is crucial for maintaining health, both on Earth and in space. During space flight, human physiology is specifically challenged by complex environmental stressors, which are most pronounced during lunar or interplanetary missions. Adopting an interdisciplinary approach, the book identifies the impact of these stressors – the space exposome – on immunity as a result of (dys-)functions of specific cells, organs and organ networks. These conditions (e.g. gravitation changes, radiation, isolation/confinement) affect immunity, but at the same time provide insights that may help to prevent, diagnose and address immune-related health alterations. Written by experts from academia, space agencies and industry, the book is a valuable resource for professionals, researchers and students in the field of medicine, biology and technology. The chapters “The Impact of Everyday Stressors on the Immune System and Health”, “Stress and Radiation Responsiveness” and “Assessment of Radiosensitivity and Biomonitoring of Exposure to Space adiation” are available open access under a Creative Commons Attribution 4.0 International License via link.springer.com.

Book excerpt: Methods in Toxicology, Volume 2: Mitochondrial Dysfunction provides a source of methods, techniques, and experimental approaches for studying the role of abnormal mitochondrial function in cell injury. The book discusses the methods for the preparation and basic functional assessment of mitochondria from liver, kidney, muscle, and brain; the methods for assessing mitochondrial dysfunction in vivo and in intact organs; and the structural aspects of mitochondrial dysfunction are addressed. The text also describes chemical detoxification and metabolism as well as specific metabolic reactions that are especially important targets or indicators of damage. The methods for measurement of alterations in fatty acid and phospholipid metabolism and for the analysis and manipulation of oxidative injury and antioxidant systems are also considered. The book further tackles additional methods on mitochondrial energetics and transport processes; approaches for assessing impaired function of mitochondria; and genetic and developmental aspects of mitochondrial disease and toxicology. The text also looks into mitochondrial DNA synthesis, covalent binding to mitochondrial DNA, DNA repair, and mitochondrial dysfunction in the context of developing individuals and cellular differentiation. Microbiologists, toxicologists, biochemists, and molecular pharmacologists will find the book invaluable.

Available in PDF, EPUB and Kindle. Book excerpt: It is well established that asthma is an inflammatory disease of the airway mucosa and drugs like inhaled glucocorticoids are now commonly introduced early in therapy. A characteristic feature of this disease is the vast number of eosinophils in airway tissue, although many other migratory and resident inflammatory cells with the capacity to synthesize and release cytokines and putative asthma mediators are present in the inflamed mucosa. The cross-talk between lymphocytes and these cells and the role of cytokines in complex biological networks are currently areas of intense research. This volume gathers together chapters and discussions on the biology of immunocompetent and inflammatory cells, cellular interplay and communication, and on the relative importance of cells and mediators in disease. It should help contribute to further insights into the pathology of asthma and to the development of novel efficacious drugs for the treatment of asthma and related respiratory disorders.
