It is composed of a wide variety of cell types that establish specific connections to form functional circuits. During development, circuits in the retina generate spontaneous waves of activity that instruct the wiring of the visual system. We study the circuits that produce stage III waves and desynchronize the firing of neighboring ganglion cells with opposite light responses (ON and OFF RGCs), a feature that is thought to help establish parallel ON and OFF pathways in downstream visual areas. We find that intersecting lateral excitatory and vertical inhibitory circuits give rise to precisely patterned stage III retinal waves. The retina can encode a wide range of visual features due to the variety of signals generated by its circuits. Amacrine cells (ACs) are the most diverse class of neurons in the retina yet of the 30 50 AC types in mammalian species, few have been studied in detail. Here, we identify and morphologically characterize three VIP expressing GABAergic AC types (VIP1 , VIP2 and VIP3 ACs) in mice. We find that the somata of VIP1 , VIP2 and VIP3 ACs are asymmetrically distributed along the dorso-ventral axis of the retina and that their neurite arbors differ in size and stratify in distinct sublaminae of the inner plexiform layer. Next, we target VIP ACs under 2 photon guidance for patch clamp recording to analyze light responses and underlying synaptic inputs. We find that VIP1 , VIP2 and VIP3 ACs differ in response polarity and spatial tuning, and contribute to the diversity of inhibitory and neuromodulatory signals in the retina.Finally, to examine the contribution of VIP ACs to visual possessing, we probe how conditional suppression of neurotransmitter release alters circuit function. First, we identify a RGC synaptic pattern using a combination of optogenetics and patch clamp recordings. Then, we chronically silence GABAergic synapses in VIP ACs by genetic deletion of vesicular GABA transporter (VGAT). Preliminary data indicate that this manipulation does not significantly alter downstream light responses. We are currently pursuing additional RGC partners as well as alternative tools to silence VIP ACs.

Neural circuits combine an intrinsic program, that manifest itself in the form of spontaneous activity and molecular markers, with input from the environment to mature into a perfect arrangement of circuits that define how we interact with the world. The immature mammalian retina is a perfect example of the intricacies of neural circuit development. The developing retina exhibits a robust pattern of spontaneous activity, known as retinal waves. Retinal waves are necessary for proper development of the neural connections in early parts of the visual pathway. Recent research on this topic establishes that specific spatiotemporal properties of waves instruct specific aspects of visual map formation. In addition, retinal waves have recently found roles in cell differentiation, cell migration, and synapse formation. Thus, understanding the underlying mechanisms of the spatiotemporal properties of waves and exploring new ways in which waves could influence developing neural circuits will contribute to our understanding the role of spontaneous activity in developmental processes of the nervous system. In mice during postnatal days 1-9, retinal waves are generated by cholinergic starburst amacrine cells (SACs), which are excitatory retinal interneurons at this stage. Starburst amacrine cells exhibit slow afterhyperpolarizations that are thought to set a refractory period and thus the periodicity and frequency of waves. Using an array of electrophysiological methods such as patch-clamp and multi-electrode array recordings, and calcium imaging, we showed that the 2- pore potassium channel TREK1 underlies SACs slow afterhyperpolarization and thus wave frequency. First, we found that the SACs afterhyperpolarization is physiologically and pharmacologically consistent with a slow potassium current generated by a 2-pore potassium channel. Second, gene expression analysis of SACs revealed the expression of TREK1. Lastly, either silencing the putative TREK1-activated signaling pathways or knocking out TREK1 increases the frequency of retinal waves. Our work provides an example of how molecular mechanisms generate specific spatiotemporal properties of waves, which are important for the development of the visual system. In addition to instructing the wiring of retinal inputs to the visual centers of the brain, retinal waves influence developmental aspects within the retina. Prior to the onset of vision, which happens after cessation of waves, the retina relies on intrinsically photosensitive retinal ganglion cells (ipRGCs) for light detection. We explored the influence of spontaneous retinal waves on the ipRGC-dependent light response of the developing retina. We combined patch- clamp recordings, calcium imaging, multi-electrode array recordings, and tracer coupling, among other methods to determine interactions between cholinergic waves and ipRGC circuits. First, we found that ipRGCs form gap junction connections with other retinal neurons, including other ipRGCs. Second, blocking cholinergic waves induces ipRGCs to form more gap junction connections. Third, blocking waves dramatically increases the number of light-responsive cells. Fourth, waves evoke dopamine release in the developing retina. Lastly, blocking dopamine signaling increases the number of light-responsive cells, similar to blocking waves. We concluded that retinal waves evoke dopamine release that act on ipRGC to down regulate their gap junction coupling. Thus, retinal waves modulate ipRGC gap junction coupling via dopaminergic signaling to regulate the overall light response of the developing retina. To further explore the properties of the light responses of developing ipRGCs, we characterized their light-evoked calcium transients. There are 5 different types of ipRGCs identified in adult mice, and 3 physiological types in developing mice. Pooling data from a large population of ipRGCs we analyzed amplitude, latency, and response decay of ipRGC light responses. We found that although amplitude and latency exhibited a large variation across cells, they could not be used to classify cells into different subtypes of ipRGCs. In contrast, response decay showed multimodal distributions and thus potential as a classifier for developing ipRGC types. This work contributes to our understanding of light-sensing in the developing retina.

This phenomenon has been studied extensively in the developing retina. Before vision is possible in the retina, neighboring retinal ganglion cells, the retina's projection neurons, spontaneously fire correlated bursts of action potentials separated by long periods of silence. This bursting activity propagates across the retina in the form of "retinal waves". My overall goal was to elucidate the transient features of the developing retina that are the synaptic basis of retinal waves. In the adult retina, circuitry is organized vertically, such that signals travel from the light-sensitive photoreceptors, through a class of excitatory interneuron known as bipolar cells, to retinal ganglion cells, whence the signal is transmitted to the brain. Hence, in the adult retina, each point in visual space is represented by excitation in a small group of cells, with minimal lateral communication among neighboring groups of cells. In contrast, during development, retinal waves propagate laterally across many degrees of visual space. Using a combination of transgenic mice and physiological recordings from acutely isolated mouse retinas, I tested two hypotheses regarding how activity propagates laterally across the retina during development. First, I tested whether extrasynaptic glutamate provides a source of coupling among neighboring bipolar cells. Second, I tested whether specific neuronal connexins, which are the proteins that form electrical synapses, are involved in wave generation. These experiments will help us to better understand the mechanisms by which cells in the brain form the correct connections during development. The knowledge gained from these experiments may help to explain congenital defects of the visual system and could be used to help reestablish connections in the injured brain.

Book excerpt: Across the developing nervous system, immature networks generate spontaneous activity that is highly correlated amongst neighboring cells, which is required for the correct establishment of adult neural circuits. Remarkably, correlated activity persists following disruption of the underlying circuits that mediate it, indicating that plasticity mechanisms exist to ensure correlated activity is maintained. Here, we examine this phenomenon in the developing mouse retina, where correlated activity is mediated by cholinergic transmission and propagates across the retina as a wave. The absence of cholinergic signaling leads to the generation of "recovered" waves that propagate through a distinct, gap junction mediated circuit. Our findings show that stimulation of melanopsin-expressing intrinsically photosensitive retinal ganglion cells (ipRGCs) facilitates recovery of correlated activity in the absence of cholinergic waves, which results in the emergence of a light-sensitive network. We tested whether pharmacological blockade of cholinergic waves altered retinal light-response properties. We observed an increase in the duration of light-evoked activity and number of light-responsive cells, which arose from signaling via gap junctions. These observations suggest that electrical coupling of ipRGCs increases in the absence of cholinergic input, allowing melanopsin-driven signals to propagate to other neurons. Furthermore, we show that light-sensitive waves are strongly modulated by dopamine signaling--a potent neuromodulator of gap junction coupling. We determine that this light-sensitive wave circuit is present but latent in wild type retina, where it is usually suppressed by a combination of cholinergic and dopaminergic signaling. Our observations indicate that dopamine signaling acts as a switch for network reconfiguration, where high dopamine silences the light-sensitive, gap junction coupled network under cholinergic waves and reduced dopamine activates it in the absence of cholinergic waves. We conclude that the wiring diagram of the developing retina includes several overconnected circuits, in which some circuits are closed and others activated depending on the internal state of the system.
