Book excerpt: Coronary heart disease (CHD) remains a major cause of human mortality and morbidity worldwide. Classical risk factors for atherosclerosis fail to account fully for the wide variations in CHD prevalence and/or severity between differing populations. The search for hitherto unrecognised risk factors has recently focused on the potential role of chronic inflammation and common infections. Chronic Infection, Chlamydia and Coronary Heart Disease is the first book to review the wealth of evidence linking various infective agents with CHD, focusing particularly on the proposed pathogenetic role of the main candidate microorganism, Chlamydia pneumoniae. Written by two leading investigators in the field, the book provides a concise, clearly written and up-to-date account of the diverse lines of evidence - seroepidemiological, pathological, animal and in vitro data and clinical antibiotic trials. The possibility of an infective basis to atherosclerosis and CHD has already attracted much scientific interest. Chronic Infection, Chlamydia and Coronary Heart Disease highlights the key published works on the topic, and concludes with information on existing, ongoing, and future lines of enquiry.

Available in PDF, EPUB and Kindle. Book excerpt: Chlamydia pneumoniae is a bacteria that is most commonly known for causing colds and pneumonia. However, researchers have recently found a link between C. pneumoniae and atherosclerosis, a clogging of the arteries that causes heart attack and stroke. In addition, ongoing research is showing that certain strains may play a role in asthma, multiple sclerosis, Alzheimer's disease, and arthritis. This volume, part of the Infectious Agents and Pathogenesis series, is a complete portrait of C. pneumoniae and what is currently known about it.

Available in PDF, EPUB and Kindle. Book excerpt: A flood of data indicate the importance and the relevance in both respiratory and extrapulmonary disease of Chlamydia pneumoniae infection today. Antibody prevalence rates in Western countries reach 50% in the adult population and remain high in old age suggesting a high rate of reinfection. Chlamydia pneumoniae can cause upper respiratory tract infections, like pharyngitis and sinusitis as well as bronchitis and more than 10% of community-acquired pneumonia. Recent data suggest a possible association of Chlamydia pneumoniae infection with wheezing and adult-onset asthma. However, the most intriguing hypothesis pertains the possible association between Chlamydia pneumoniae infection and atherosclerosis.

Available in PDF, EPUB and Kindle. Book excerpt: Infectious agents have been recognized to involve the heart and vascular system for well over a century. Traditional concepts and teachings of their involvement in the pathogenesis of disease have been by a few established mechanisms. Since the last decade of the 20th century there has been renewed interest in the medical and public media on infectious diseases affecting the cardiovascular and cerebrovascular systems, through their relationship with the development of acceleration of atherosclerosis. This volume highlights and reviews new perspectives of infections on the cardiovascular system as never before. It is a truly valuable resource for scientists, researchers, residents, and fellows in the fields of infectious disease, cardiology, and microbiology.

Book excerpt: International specialists in Microbiology, Infectious Diseases, Internal Medicine, Cardiology Immunology, Pneumonology, Neurology and Epidemiology gathered to this workshop to discuss and enhance interdisciplinary knowledge on the possible etiological role of Chlamydia pneumoniae, a widespread human respiratory infection, in the pathogenesis of chronic inflammatory diseases with major public health impact such as atherosclerosis, cardiovascular disease, adult-onset asthma bronchiale, chronic obstructive pulmonary diseases, reactive arthritis, morbus Alzheimer and multiple sclerosis. Current deficits and goals in the standardisation of diagnostic tools, basic and applied research, design of epidemiological and monitoring of therapy studies were identified. A special feature of this book is the comprehensive collection of state-of-the-art review articles in the various fields with many references.

Available in PDF, EPUB and Kindle. Book excerpt: It has been known for over 150 years that hallmarks of inflammation can be observed in the wall of atherosclerotic vessels. It was, however, not clear if this inflammation is the cause or the consequence of atherogenesis. More recently, it has become evident that inflammation mediated both by innate and adaptive immunity is instrumental even in the earliest stages of the development of atherosclerotic lesions, i.e., that it plays an important pathogenetic role. In this volume, international experts in the field discuss the pathogenetic, diagnostic, preventive and possible therapeutic relevance of inflammation in atherogenesis. This book is intended for researchers and physicians in the fields of vascular biology, immunology and atherosclerosis.

Book excerpt: Strong clinical and experimental evidence has suggested the involvement of Chlamydia pneumoniae (C. pneumoniae) in the development of atherosclerosis. However, the direct role of C. pneumoniae infection in vascular remodelling processes in the absence of a host immune response remains undetermined. To study the direct effect of this pathogen within the arterial wall, we developed a novel ex vivo porcine coronary artery model that supported bacterial growth for up to two weeks in culture. Employing this approach, we demonstrated that C. pneumoniae infection could alter vascular functions parameters, including endothelial-dependent relaxation responses. This impairment was associated with a decrease in eNOS expression and increased oxidative stress, changes that are also noted in atherosclerotic plaques. We further demonstrated that C. pneumoniae infection initiates medial thickening via vascular smooth muscle cell (VSMC) proliferation. This proliferative response was associated with an increase in expression of endogenous heat shock protein 60 (Hsp60) and alterations in nuclear protein import machinery. Additionally, C. pneumoniae infection and Hsp60 overexpression in primary VSMCs resulted in alteration in nuclear protein import parameters leading to the cell proliferation. Using a rabbit atherosclerotic model, we demonstrated that Hsp60 is induced during atherosclerotic lesion growth and correlated with both the proliferative status and the expression of protein involved in nuclear protein import within the atherosclerotic vessel. In summary, our work has demonstrated the feasibility of studying the molecular mechanisms of infection-induced atherosclerosis using an ex vivo coronary culture system. Importantly, our data has provided the first direct evidence that an active C. pneumoniae infection alone, without contributions from a host immune system, can mediate endothelial dysfunction and stimulate arterial thickening, two key remodelling processes present during atherosclerotic progression. Our findings further suggest the involvement of Hsp60 as a key contributor in growth-based pathologies like C. pneumoniae-mediated atherosclerosis possibility through modulation of nuclear protein import.
