Book excerpt: For more than a century mass spectrometry has been a well-known technique in the field of chemical analytics. Its selectivity and sensitivity has made it popular in various fields. From analysis of pure organics, its use is still being explored in the analysis of biomolecules, either purified or direct from tissue sections. For analyzing these vast arrays of molecules, typically the front end is modulated depending upon the need of the user. For direct analysis of a sample of interest, ionization techniques such as DESI, PS, MALDI, etc are incorporated into the front of the mass spectrometer. In this work, the ambient ionization techniques, DESI-MS and PS-MS was modulated for characterization, imaging and quantification of small molecules. The use of DESI-MS and PS-MS in the field of forensics, microbiology and pharmaceutics is described. With the optimization of the front end modes, better sensitivity, selectively and robustness was ensured.

Book excerpt: Mass spectrometry (MS) has become a widely used technique for the characterization of a wide range of substances in diverse fields. The selection of appropriate ionization techniques, source parameters, charge carriers based on the analyte’s polarity is essential in MS as only the ions are detected. When using a soft ionization technique, single stage-MS at best provides only chemical composition; thus, tandem MS is needed to determine structural information and dissociation pathways. This dissertation focuses on the characterization of various small molecules and ions using different ionization techniques, charge carriers, and collision-induced dissociation (CID). In Chapter II, commercially available ionic liquid (IL) cations, specifically imidazolium-based IL cations with different side-chain chemistries, were characterized by CID-MS. The imidazolium-based IL cations have diverse dissociations pathways depending on the nature of the side chain (aliphatic or aromatic) or the functional groups (allyl, vinyl, alcohol, methoxy, nitro) present on the side chain. Additionally, it was observed that some IL cations undergo thermal degradation under normal operating temperatures of electrospray ~275 °C. A variety of experimental and complementary computational chemistry results are presented. In Chapter III, cis-3-hydroxyproline (c3hPro) and cis-4-hydroxyproline (c4hPro) isomers were differentiated upon lithiation using CID-MS. The CID-MS of [c4hPro+Li]+ produces three primary fragmentation pathways, namely the neutral losses of H2O, CO, and CO2; whereas CID-MS of [c3hPro+Li]+ produces only loss of CO2 in this same region. These observations may have analytical utility, and in this work, the experimental observations were explored with thermodynamic and transition state calculations to better understand the processes. In Chapter IV, the accurate mass of synthetic monomers including PAH perfluorocycloalkene monomers and their precursors were measured and reported with a publication-quality mass accuracy using atmospheric pressure chemical ionization (APCI)-MS. Additionally, the effect of solution flow rate, ionization mode, source parameters (such as vaporization temperature, dry gas temperature and dry gas flow rate, nebulizer gas pressure), acidification, and different solvent systems on APCI-MS signal intensity were studied.

Book excerpt: Ambient ionization mass spectrometry (MS) approaches have revolutionized the way diagnostic molecular information can be obtained from biological and clinical samples, requiring minimal to no sample pretreatment, being operationally simple, and most importantly, providing near real time assessment of molecular information directly from biospecimens. Ambient ionization approaches have been widely applied to various clinical applications including screening of inborn errors of metabolism, therapeutic drug monitoring, and disease diagnosis and subtyping, with the goal of evaluating their use for addressing complex challenges in human health. Desorption electrospray ionization (DESI) MS is the most extensively used ambient ionization method for the investigation of molecular changes in tissue biospecimens, showing great potential for providing complimentary information to routine histopathology. Since its inception, a variety of other ambient ionization MS methods have been developed, with newer techniques, such as the MasSpec Pen, envisioned for the intraoperative analysis of in vivo and ex vivo tissue specimen during surgery. This dissertation describes the development and application of ambient ionization MS methods for the investigation of various human diseases. Chapter 2 provides discussion on the robustness of DESI-MS for breast cancer tissue section evaluation and receptor status determination in a multi-center study, providing evidence of its potential use to compliment routine histopathological assessment. Chapter 3 describes the optimization of DESI-MS to expand the molecular information obtained from a single tissue section, demonstrating for the first time the detection of proteins directly from tissue sections by DESI-MS. Chapters 4 and 5 discuss the clinical translation of the MasSpec Pen into an operating room for feasibility testing as well as the evaluation of the technology for improving intraoperative surgical margin evaluation during breast cancer surgeries. Lastly, Chapter 6 discusses the modification of the MasSpec Pen to better analyze clinical nasal swabs and its application toward screening of COVID-19 disease using lipid information

Book excerpt: Field Ionization Mass Spectrometry focuses on developments in field ionization (FI) mass spectrometry and describes its applications in physical chemistry, with emphasis on mass spectrometric problems. Physico-chemical problems as well as problems of chemical analysis are considered based on issues such as the probability of field ionization; field dissociation and charge distribution; kinetics of ion decomposition in high fields; negative ions; surface diffusion; activation of FI emitters; and elucidation of the structures of organic compounds. This book is comprised of four chapters and begins with a short review on some of the most important directions of research in FI mass spectrometry. Two main fields of research are discussed: physico-chemical investigations and quantitative analysis or structural determination of organic substances. The next chapter is devoted to focusing and non-focusing sources of FI and covers topics such as methods for production of FI tips and thin wires, together with the use of tips and carbon filaments as FI emitters. The last two chapters focus on the application of the FI mass spectrometer to physico-chemical problems and to quantitative analysis of homologous series of organic substances such as alkanes, alkenes, alkynes, amines, and alcohols. This monograph is intended primarily for chemists and mass spectrometrists.

Book excerpt: Direct analysis using mass spectrometry (MS) techniques provide a powerful approach to investigate the molecular composition and spatial distribution of complex samples with high sensitivity and specificity. In particular, these techniques have been widely applied to achieve rapid molecular analysis of complex clinical samples including native tissue samples and biospecimens. Several types of direct analysis MS techniques have been developed for tissue analysis including a variety of ambient ionization MS techniques and matrix-assisted laser desorption ionization (MALDI) MS. Notably, both approaches are highly capable of performing molecular imaging of endogenous metabolites, lipids, and proteins to characterize tissue samples. Furthermore, through applying ambient ionization and MALDI MS imaging techniques, new insights into disease state can be learned by investigating the correlation between spatially resolved molecular information with histological evaluation in tissue samples. Although several other types of direct analysis MS techniques have been developed, this dissertation focuses on advancements using solvent-based ambient ionization MS (e.g. desorption electrospray ionization (DESI)) and MALDI MS imaging methods for tissue analysis and disease characterization. In Chapter 2, a fundamental and systematic investigation of the affect of solvent properties on molecular extraction processes in solvent-based ambient ionization was performed. In Chapter 3, the performance of DESI MS imaging for the analysis and statistical classification of thyroid fine needle aspiration (FNA) biopsies using different solvents, sources, and high-performance MS systems was evaluated. Next, Chapter 4 describes the development of a sample preparation method for MALDI MS imaging to improve detection of various endogenous proteins by selectively reducing signal from hemoglobin proteins in blood-rich tissues. Lastly, in Chapter 5, various MALDI imaging MS methods were applied to characterize the protein and N-glycan molecular signatures in endometriosis tissues and provide new insights into possible mechanisms of endometriosis development and progression. Collectively, this dissertation provides insights into the fundamental aspects, method developments, and applications of direct analysis MS techniques including solvent-based ambient ionization MS and MALDI MS imaging for tissue analysis. Furthermore, this dissertation showcases the exciting and significant potential of direct analysis MS techniques to solve current challenges in the clinic and provide new insights in healthcare and medicine

This book was released on 2009 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: Ambient mass spectrometry (MS) is a new and growing sub-field in MS which has opened new research avenues, particularly for applications relating to the analysis of solid samples. Results on the implementation and application of ambient MS techniques including: desorption electrospray ionization (DESI) and direct analysis in real time (DART) indicated that these techniques could serve as complementary tools for the rapid qualitative screening of pharmaceuticals, allowing up to two orders of magnitude improvement in throughput compared to traditional methods such as liquid chromatography MS. The selectivity of DESI could be enhanced by performing the experiment in the reactive mode. In this mode, complexation reactions between reagents added to the spray solvent and analytes on the sample surface resulted in analyte stabilization, inhibiting fragmentation. They also resulted in a concomitant enhancement in the analyte surface activity, facilitating their evaporation from secondary droplets culminating in an improvement in sensitivity. Also for drug tablets analysis, the analyte signal dependency on DESI geometrical set-up variables could be mitigated following the careful and controlled addition of an isotopically labeled internal standard (IS) to the sample or by spraying samples with a pair of reagents with different affinities for the analyte. Either of these approaches resulted in an analyte-to-IS signal ratio (in the former) or an analyte complex ratio (in the later), which was largely independent of DESI experimental variables allowing quantitative analysis using this technique. DESI MS was also observed to be a very powerful tool for determining the 2-D distribution of various pharmaceutically important compounds on tablet and tissue surfaces. The ability to map the distribution of molecules of interest by DESI MS has very great implications in drug tablet quality control and in determining the role of chemical signals presented on tissue surfaces. DESI was observed to be limited to ionizing molecules of medium to high polarities without much limitation in terms of mass range, whereas DART was better suited for the analysis of molecules within a broader range of polarities, but within a more limited mass range (up to 800 Da approximately). These limitations were circumvented by implementing a novel multimode ambient ion source, desorption electrospray/metastable-induced ionization (DEMI), which combines various aspects of DESI and DART. Initial experiments with the DEMI ion source demonstrated its ability to enable the simultaneous analysis of molecules within a broader range of polarities and masses than DESI and DART alone.
