Book excerpt: By means of this ‘Frontiers in Genetics’ research topic, we are celebrating 30 years of the Comet Assay. The first paper on this single-cell gel electrophoresis assay was published in 1984 by O. Ostling and K.J. Johanson (Biochem. Biophys. Res. Commun. Vol.123: 291-298). The comet assay is a versatile and sensitive method for measuring single - and double-strand breaks in DNA. By including lesion-specific enzymes in the assay, its range and sensitivity are greatly increased, but it is important to bear in mind that their specificity is not absolute. The comet assay (with and without inclusion of lesion-specific enzymes) is widely used as a biomarker assay in human population studies - primarily to measure DNA damage, but increasingly also to assess the capacity of cells for DNA repair. Ostling and Johanson (Biochem. Biophys. Res. Commun., 1984) were also the first to report experiments to measure DNA repair, by simply following the decrease of DNA damage over time after challenging cells with ionising radiation. However, this approach is time-consuming and laborious as it requires an extended period of cell culture and is therefore not ideal for biomonitoring studies, which typically require high-throughput processing of many samples. As an alternative approach, the in vitro comet-based repair assay was developed: a cell extract is incubated with a DNA substrate containing specific lesions, and DNA incisions accumulate. The in vitro comet-based repair assay has been modified and improved over the past decade: it was first devised to measure base excision repair of oxidised purines in lymphocytes (Collins et al., Mutagenesis, 2001), but has since been adapted for other lesions and thus other repair pathways, as well as being applied to tissue samples in addition to cell suspensions. Even after 30 years, the comet assay is still in a growth phase, with many new users each year. Many questions are repeatedly raised, which may seem to have self-evident answers, but clearly, it is necessary to reiterate them for the benefit of the new audience, and sometimes being forced to think again about old topics can shed new light. Different applications of the comet assay are discussed in this special issue, including: genotoxicity testing in different organisms, human biomonitoring, DNA repair studies, environmental biomonitoring and clinical studies. Furthermore, we consider and where possible answer questions, including the ones raised by Raymond Tice at the 8th International Comet Assay Workshop in Perugia (Italy 2009): What is the spectrum of DNA damage detected by the various versions of the comet assay?; What are the limitations associated with each application?; What should be done to standardize the assay for biomonitoring studies?; Can the comet assay be used to monitor changes in global methylation status?; What cell types are suitable for detecting genotoxic substances and their effects in vivo and in vitro?; Can the assay be fully automated?; and more. So this ‘Frontiers in Genetics’ research topic is written for the beginner as well as for the experienced users of the comet assay.

Book excerpt: What are these graceful visitors to our skies? We now know that they bring both life and death and teach us about our origins. Comet begins with a breathtaking journey through space astride a comet. Pulitzer Prize-winning astronomer Carl Sagan, author of Cosmos and Contact, and writer Ann Druyan explore the origin, nature, and future of comets, and the exotic myths and portents attached to them. The authors show how comets have spurred some of the great discoveries in the history of science and raise intriguing questions about these brilliant visitors from the interstellar dark. Were the fates of the dinosaurs and the origins of humans tied to the wanderings of a comet? Are comets the building blocks from which worlds are formed? Lavishly illustrated with photographs and specially commissioned full-color paintings, Comet is an enthralling adventure, indispensable for anyone who has ever gazed up at the heavens and wondered why. Praise for Comet "Simply the best." —The Times of London "Fascinating, evocative, inspiring." —The Washington Post "Comet humanizes science. A beautiful, interesting book." —United Press International "Masterful . . . science, poetry, and imagination." —The Atlanta Journal & Constitution

Book excerpt: Optical Materials presents, in a unified form, the underlying physical and structural processes that determine the optical behavior of materials. It does this by combining elements from physics, optics, and materials science in a seamless manner, and introducing quantum mechanics when needed. The book groups the characteristics of optical materials into classes with similar behavior. In treating each type of material, the text pays particular attention to atomic composition and chemical makeup, electronic states and band structure, and physical microstructure so that the reader will gain insight into the kinds of materials engineering and processing conditions that are required to produce a material exhibiting a desired optical property. The physical principles are presented on many levels, including a physical explanation, followed by formal mathematical support and examples and methods of measurement. The reader may overlook the equations with no loss of comprehension, or may use the text to find appropriate equations for calculations of optical properties. Presents the optical properties of metals, insulators, semiconductors, laser materials, and non-linear materials Physical processes are discussed and quantified using precise mathematical treatment, followed by examples and a discussion of measurement methods Authors combine many years of expertise in condensed matter physics, classical and quantum optics, and materials science The text is written on many levels and will benefit the novice as well as the expert Explains the concept of color in materials Explains the non-linear optical behavior of materials in a unified form Appendices present rigorous derivations

Available in PDF, EPUB and Kindle. Book excerpt: Concerns about the adverse effects of chemicals present in the environment have created a need for better systems to assess their potential consequences on human health. One potential solution is the versatile and state-of-the-art Comet assay. Simple, sensitive, rapid and visual, this modern toxicological method allows quantitative and qualitative assessment of DNA damage in single cells. This assay is used in diverse fields ranging from clinical applications, human monitoring and environmental toxicology through to genetic toxicity testing. This updated and revised edition of The Comet Assay in Toxicology provides the latest information on this important tool. It addresses, in-depth, the different protocols, statistical analyses and applications used worldwide. It also includes the guidelines recommended by the Working Group on Comet Assay. The book begins with a review of the genesis of the assay for those new to the technique and goes on to explain procedures followed to assess different types of DNA damage, various applications of the assay, and guidelines for the conduct of the assay in in vitro and in vivo systems. New chapters written for this edition will provide information on the most contemporary approaches and applications, including in silico approaches, on meta-analysis of data and on the application of the Comet Assay in nanotoxicology. This book will serve as both a reference and a guide to students as well as investigators in the biomedical, biochemical and pharmaceutical sciences fields.

Book excerpt: In the gripping start to this young adult science fiction adventure series by popular Colorado radio host Dom Testa, the teenage crew of the starship Galahad must find a new home for humanity among the stars--if they fail, it will be the end of the human race.... When the tail of the comet Bhaktul flicks through the Earth's atmosphere, deadly particles are left in its wake. Suddenly, mankind is confronted with a virus that devastates the adult population. Only those under the age of eighteen seem to be immune. Desperate to save humanity, a renowned scientist proposes a bold plan: to create a ship that will carry a crew of 251 teenagers to a home in a distant solar system. Two years later, the Galahad and its crew—none over the age of sixteen—is launched. Two years of training have prepared the crew for the challenges of space travel. But soon after departing Earth, they discover that a saboteur is hiding on the Galahad! Faced with escalating acts of vandalism and terrorized by threatening messages, sixteen-year-old Triana Martell and her council soon realize that the stowaway will do anything to ensure that the Galahad never reaches its destination. The teens must find a way to neutralize their enemy. For if their mission fails, it will mean the end of the human race....
