Book excerpt: The Hudson Raritan Estuary (HRE) was once home an abundant population of the eastern oyster, Crassostrea virginica. Years of habitat degradation, via removal of habitat and shell substrate, overfishing of the population, and inputs of organic and inorganic contaminants, all led to the decline of this previous keystone species. The HRE today is a highly eutrophic environment, with increased sediment inputs, periods of low dissolved oxygen, algal blooms, and hotspots of contaminants throughout. The current study was designed to understand how a chronically degraded habitat, as is present in the HRE now, affects both juvenile and adult oyster physiology. There are three parts to this study: in the first, a large scale field transplant study was deployed to determine how juvenile oyster health and subcellular physiology are altered over a continuum of sites across the HRE, and if subcellular metal accumulation related to alterations in physiology. Using eight sites across the HRE, it was apparent that there are many site-specific factors that affect oyster physiology, and the synergistic effects of these abiotic and biotic factors together influence oyster physiology the most. There was no one factor that could be isolated as a key parameter to determine future oyster restoration. The second part used a field transplant study to examine the role of a degraded habitat on adult oysters and reproduction. Using Vitellogenin protein and energy expenditures to estimate oyster reproduction, it was seen that adult oysters respond much slower than juveniles and no differences were seen between highly degraded habitats and less degraded habitats. Thirdly, both juveniles and adults were observed to accumulate non-essential metals (Cd and Hg) in the field. In order to determine if metal accumulation is the sole cause of physiological alterations, a laboratory exposure was designed to determine if changes in subcellular physiology could be correlated specifically to subcellular accumulation of Cd or Hg, when no other abiotic factors are able to influence oyster health. It was observed that Cd accumulation lead to physiological changes, but Hg accumulation did not. Using this information about the site-specific nature of oyster physiology and how metal accumulation can alter physiology will allow researchers to choose future restoration sites and set up projects that will allow for maximum growth and survival.

While cadmium (Cd) has been shown to alter specific immune defense mechanisms, the direct effect of Cd on the inactivation and degradation of bacteria in this animal is unknown. Furthermore, the roles of individual tissues in the distribution, inactivation and degradation of live bacteria introduced into the host remain unclear. First, we determined the effect of Cd on the immune responses of C. virginica at the organismal, cellular and molecular level. Following a chronic Cd exposure, oysters were injected in the adductor muscle with 105 live Vibrio campbellii. Quantitative real-time polymerase chain reaction (Q-PCR) was used to quantify the number of intact bacteria, while selective plating was used to quantify the number of injected bacteria that remained culturable within the tissues and fluids of the whole animal at 10, 30 and 60 min after injection. While Cd did not alter the oyster's ability to inactivate bacteria, Cd exposure induced a significant decrease in the numbers of circulating hemocytes and significant changes in the expression of selected genes as determined by microarray analysis. The gills, digestive gland and hemocytes mounted unique responses to chronic Cd exposure, yet the functional integrity of immune defense at the organismal level was maintained under these conditions. Second, we determined the relative contribution of individual tissues to the antibacterial response of C. virginica following injections of V. campbellii. Numbers of intact bacteria were used to identify sites of bacterial accumulation, while percentages of culturable bacteria were quantified to determine sites of bacteriostasis within the digestive gland, mantle, gills, adductor, gonadal tissues, labial palps and hemolymph at 10, 30, 60 and 120 min after injection. The gonadal tissues contained the greatest concentration of intact bacteria and the lowest percentage of culturable V. campbellii compared to all other tissues. We suggest that the gonadal tissues, when present, are the main site of bacterial accumulation and bacteriostasis. Overall, this study demonstrates the resilience of oysters to bacterial infections by their ability to inactivate bacteria.

Book excerpt: Bivalve aquaculture is increasingly being considered as a viable bioremediation strategy in eutrophied estuaries. Bivalves provide a variety of ecosystem services, including water filtration and nutrient removal from the water column. Filtration rates vary according to species and location-specific hydrological conditions. To explore the potential benefits of using eastern oyster (Crassostrea virginica) aquaculture as part of a nutrient trading plan, I examined nitrogen (N) accumulation across the Delaware Inland Bays, a series of small estuaries on the Mid-Atlantic coast. Two different size classes of oysters were deployed in multiple locations to monitor N content across space and N accumulation rate according to size. I found that N content varied with location but not with size class. N content in oyster tissue (% DW) was similar to previous findings for C. virginica, but content in shell (% DW) was up to an order of magnitude greater. My results confirm that N accumulation varies depending on location and suggests that N content within the shell may be higher than previously reported. My attempts to quantify the nutrient bioassimilation services that oyster aquaculture provides could be used to help inform future nutrient management plans in the Delaware Inland Bays.

Book excerpt: In the early 1990s, the introduction of suspended culture methods and the development of a market for cocktail oysters (65-75 mm) propelled the oyster aquaculture industry in New Brunswick forward, now producing over 140,000,000 oysters (337,000 floating bags) on 2,649 hectares of private shellfish leases. Recently, the industry has been threatened by an oyster disease outbreak, known as MSX in the Bras D'Or Lakes region (Nova Scotia). The possibility of the MSX parasite spreading into New Brunswick waters, as well as an increasing number of aquatic invasive species in the area, is a real concern for the industry and has prompted an investigation on possible mitigation strategies. The benefits of incorporating an upriver site into the production cycle of oyster aquaculture operations were evaluated. Seed (~ 26 mm shell height) and adult (~ 66 mm shell height) oysters were taken from downriver (salinity ~ 20-30‰) aquaculture sites and transferred upriver (salinity ~ 5-20‰) and monitored for shell and meat growth (May-October). At the same time, the impact of the relay fishery (harvest of oyster in closed marginally contaminated areas for relay to tenures in open areas) on oyster productivity was evaluated; wild upriver oysters were transferred downriver and monitored. Productivity data revealed that seed transferred upriver grew and survived as well as seed that remained downriver, while the mortality rates of adult oysters were lower at the upriver site. Meat content was unaffected in adult oysters transferred upriver. However, adult oysters transferred upriver had a gain in shell height (least squares means ± standard error) over the oysters that remained downriver (2.7 ± 0.5 mm vs. 1.8 ± 0.5 mm) and, in shell width (Median [95% CI]; 2.8 mm [1.9, 3.6] vs. 1.0 mm [0.3, 1.2]). Final organic meat content were approximately 35% less in adult oysters transferred downriver compared to those that remained upriver, confirming anecdotal reports from fishers of the relay fishery. Stress response of adult oysters was also assessed (80 and 150 days post transfer). Stress response measured revealed that regardless of their origin (upriver, downriver), oysters maintained upriver showed high levels of lysosomal destabilization and tissue atrophy. By contrast, the transfer downriver had the opposite effect, i.e. low lysosomal destabilization and low tubule atrophy. Results suggest that there exists no association between productivity parameters and the level of stress response measured by the biomarkers. The field investigation prompted further exploration on the impact of hemolymph fluid osmolality on neutral red retention assay (NRA) outcome. The prescribed saline solution for the assay assumes animals are isosmotic to surrounding water; however oysters sampled were hyperosmotic. Hemolymph osmolality was manipulated under laboratory conditions by subjecting adult oysters to temperature (0, 5, 10, 15, 20oC) and salinity (6, 27‰) regimes. At 10-20oC, oysters remained isosmotic to ambient waters, while hyperosmotic at 0-5oC. When mimicking spring freshets (salinity 6‰), hyperosmotic condition was observed in all temperature groups, and was inversely proportional to temperature. Monitoring of valve activity confirmed a restricted exchange at 6‰ salinity (e.g. spring freshet). With these laboratory results, use of filtered hemolymph fluid (NRAMOD) is recommended instead of the prescribed standard solution (standard NRA) adjusted to ambient seawater.

Book excerpt: The explosion of the Deepwater Horizon oil platform on April 20, 2010 initiated the release of 640 million liters of oil into the Gulf of Mexico (GOM). In efforts to prevent the oiling of coastlines, response officials applied nearly 7.5 million liters of the oil dispersant Corexit. Despite these efforts, oil reached over 2100 km of coastline along the northern GOM, including estuaries that serve as important habitats and nurseries. These estuaries are dynamic ecosystems, with constant fluctuations in salinity and dissolved oxygen. While it is well-established that oil is toxic to marine species, gaps remain in the understanding of how the impact of oil exposure may have been exacerbated by environmental (hypoxia and low salinity) and anthropogenic (oil dispersants) stressors. The impacts of oil and oil dispersants were investigated in an economically and ecologically important GOM invertebrate, the eastern oyster (Crassostrea virginica). Oysters were exposed to Corexit, oil, or dispersed oil to evaluate potential toxic effects on immunological (phagocytosis and respiratory burst) and physiological (feeding rate) endpoints. Exposure resulted in significant modulation of immune functions and feeding rates, and oysters were most sensitive to dispersed oil. The impacts of environmental stressors on the physiological responses to oil exposure were investigated in sheepshead minnows (Cyprinodon variegatus), small estuarine fish native to the GOM with tolerance for a wide range of environmental conditions. Adult sheepshead minnows were exposed to oil in three environmental scenarios: normoxic, hypoxic, and hypoxic with low salinity. Oil exposure reduced egg production and fertilization rate, but only when sheepshead minnows were exposed in hypoxic and hypoxic with low salinity scenarios. Parental exposure in the normoxic scenario resulted in developmental effects in F1 and F2 generations, demonstrating transgenerational effects. F1 developmental effects tended to be altered in the hypoxic and hypoxic with low salinity scenarios. The combination of reduced reproductive capacity and altered development could lead to long-term population level impacts for sheepshead minnow. These data suggest that oil spill risk assessments that fail to consider anthropogenic (chemical dispersants) and environmental stressors (hypoxia and salinity) may be underestimating risk to the health of eastern oysters and sheepshead minnows.

This decline is in spite of being considered a resilient species. Causes of the decline include overharvesting, disease, and habitat loss. While efforts have been made to combat each cause, the key element to recovering the population is coordinated habitat restoration. This study aims to develop a GIS-based habitat model for the eastern oyster in the Chesapeake Bay. The first goal of this study was to determine the water quality parameters necessary for successful oyster population restoration and to locate where those conditions exist concurrently. To identify these areas that are most suitable for restoration efforts, a habitat analysis was performed using GIS data of water quality parameters in the bay consisting of water temperature, salinity, dissolved oxygen, pH, and bathymetry. Data obtained from monitoring stations throughout the bay were used to estimate the water quality parameters. Those parameters were interpolated using inverse distance weighting to create continuous rasters of seasonal averages for each parameter. Then rasters were combined and analyzed using geospatial processing to determine the areas that contain the most favorable conditions for oyster growth at both larval and adult life stages. Additionally, the conditions for proliferation of oyster diseases MSX and Dermo were assessed geospatially. The second goal of this study was to determine the potential changes to suitable oyster habitat area and Dermo proliferation following the climate change experts' prediction of a 2°C water temperature increase. Resulting models showed a decrease in total area of high quality habitat for larval and adult models with the 2°C increase, as compared to the current condition models. Disease proliferation of Dermo also exhibited increases in higher risk areas in the 2°C increase model when compared to the current condition model.
