Adipocytes are “smart” cells able within the tissue to communicate with surrounding cells, but also with various organs, particularly via leptin acting on the central nervous system. Brown adipose tissue (BAT) and white adipose tissue (WAT) are known to be distinct tissues, whereas the heterogeneity of WAT depots is well est- lished. Unfortunately, excess WAT leads to obesity, which is the most common health problem in industrialized countries. Therefore, from both a scientific and a technical point of view, the time has come to create a survey of adipose tissues and their neglected adipocytes. In Adipose Tissue Protocols, I have attempted to gather together chapters from all areas of adipose tissue research—from in vivo to in vitro studies—and to provide methods covering a wide variety of techniques, including the choice of adipose tissue depot and of morphological techniques for the study of BAT and WAT; the isolation, subcellular fractionation, and transfection of adipocytes where the low density of these cells must be taken into account; assays of nutrient and ion fluxes and the metabolic aspects of nutrient uptake; assays of lipid-related enzymes; biopsies and quantification of lipid-related mRNAs; cultures of adipose precursor cells from WAT and BAT of various species, including human tissue; measurements of adipose secretory products; and assessment of WAT metabolism in vivo.

Book excerpt: Abstract: Barrows and gilts, representing purebred Landrace (LL), Berkshire (BB), and their respective reciprocal crossbred pigs, were weighed and ultrasonic estimates of backfat thickness (BF) and loin muscle area (LMA) were taken at two week intervals beginning at an average live weight (LWT) of 19.5 and ending at an average of 127.0 kg to assess growth and corresponding changes in composition during the growing and finishing phases. Serial LWT, BF, and LMA were assessed in conjunction with a serial harvest process. Approximately ten pigs per line, representing nearly equal sex ratios, were harvested at an average of 82, 110, 152, and 180 d of age. At 82 d of age 365 pigs had a minimum of three serial measurements, but by 180 d of age, after pigs had been removed for serial harvest, 120 pigs remained that had ten serial ultrasound measurements. The study was conducted in two seasons representing spring and summer farrowed litters. Deposition patterns for BF and LMA across the weight range and LWT across the age range were non-linear. In all analyses the genetic line by quadratic weight (or age) effects were highly significant resulting in specific genetic line deposition patterns. Berkshire pigs had greater BF than LL pigs with the difference increasing with LWT, while BF of pigs from Berkshire sire x Landrace dam (BL) and Landrace sire x Berkshire dam (LB) had intermediate and significantly different BF from BB or LL pigs until 125 kg LWT where BF of LB pigs was not different from BB pigs. Beginning at 50 kg, BB and BL pigs had less LMA than EL and LB pigs and at 125 kg, EL pigs had greater LMA than all lines. Berkshire pigs weighed significantly less when compared with all other lines between 50 and 150 d of age. At 175 d of age, BL and LB pigs weighed more than BB pigs. However, the weight of LL pigs was not different from BB, BL, or LB pigs. Sex effects were significant for BF, LMA, and LWT at all periods, but the effect was additive and consistent across all genetic lines. Results of the present study indicate that lines containing Berkshire genetics resulted in greater BF when compared with pure Landrace. In contrast, for LMA the Berkshire-sired pigs (BB and BE) had similar and less EMA, while the Landrace-sired pigs (EL and LB) had similar and greater LMA in the weight range studied suggesting breed of sire effects were important for LMA. Growth rate patterns favored lines containing Landrace genetics when compared with pure Berkshires.
