Book excerpt: Non-selective inhibitors of cyclic nucleotide phosphodiesterase (PDE), such as theophylline, have been used extensively since 1958. In the decade of the '70s, various PDE isoenzymes were defined which led to the development of the second generation of PDE inhibitors. Currently a variety of these new inhibitors are under test as potential anti-inflammatory drugs. During the past five years, molecular biology has revealed a superfamily of these phosphodiesterase isoenzymes. This book summarizes the present state of knowledge, as well as giving a comprehensive description of the compounds available. It will be invaluable for everyone who wants to choose the most suitable PDE inhibitor for their research or who is dealing with such drugs in a clinical setting. - Utilizes actual testing and research of new PDE inhibitors - Valuable for researchers and students alike

Available in PDF, EPUB and Kindle. Book excerpt: The elucidation of the cellular and molecular bases underlying the inte grated function of the central nervous system, both in disease and in health, must ultimately come from the combined efforts of scientists from many disciplines, including biology, chemistry, histology, pathology, physiology, pharmacology, and psychology. Communication between scientists from these various disciplines-vital to the advancement of our understanding of the function of the nervous system-has become more and more difficult in recent years. Both increasing specialization and the incredible increases in publications pertinent to brain research in a wide spectrum of journals, in symposium volumes, in monographs, in abstracts, and in reviews contrib ute to the problems of cross-communication and even of communication within a scientific discipline. Research on the significance of cyclic nucleo tides to the function of nervous systems is particularly illustrative of the communication problem. Since the initial publications by Sutherland, Rall, and Butcher in the late fifties and early sixties on high levels of adenylate cyclase, phosphodiesterases, and cyclic AMP in brain, the ensuing litera ture of this field has expanded exponentially. At the present time, from five to ten publications relevant to cyclic nucleotides and the nervous system appear each week. Indeed, these are minimal numbers based mainly on examination of literature titles and key index words. Many articles concerned with some aspect of central function contain, buried within their text, experiments with or related to cyclic nucleotides.

Presented in step-by-step detail, these readily reproducible methods allow the measurement of cyclic nucleotide variations in living cells, as well as their visualization in a spatio-temporal manner, the localization and characterization of their activities in tissues and living cells, and the assessment of targeted PDEs in creating specific tools and drugs.

Book excerpt: In the present volume of the Handbook of Experimental Pharmacology well known experts describe the actions of different xanthines with a focus on caffeine and theophylline. A special chapter is devoted to theobromine, an active component of chocolate, the actions of which are less well characterized. This book also presents the pharmacology of one xanthine derivative, propentofylline, as an example of a xanthine that has gone through extensive development for a novel therapeutic area.

This book was released on 2014-05-28 with total page 253 pages. Available in PDF, EPUB and Kindle. Book excerpt: Lena Daumanns's thesis describes structural and functional studies of the enzyme Glycerophosphodiesterase (GpdQ) from Enterobacter aerogenes. It also examines the properties of small mimics of this enzyme and related binuclear metallohydrolases such as the metallo-ß-lactamases to enhance our understanding of hydrolytic cleavage of important substrates like phosphoesters and β-lactams. Overall, this project has led to a better understanding of the metal ion binding and active site structural features of the enzyme GpdQ. Daumann describes how she successfully immobilized phosphoesterase and related biomimetics on solid supports for potential applications in the area of bioremediation of organophosphate pesticides. Analysis shows that both the enzyme and biomimetics can be stored on the solid support without loss of activity. Furthermore, the author specroscopically and mechanistically characterized a number of Zn(II), Cd(II) and Co(II) complexes, some of which are among the most active biomimetics towards organophosphates reported to date. This thesis makes excellent reading for non-specialists because each chapter includes a short introduction section.

Available in PDF, EPUB and Kindle. Book excerpt: PDEs are a family of enzymes that catalyze the hydrolysis of intracellular cyclic nucleotides. They are implicated in a number of disorders and dysfunctions and PDE inhibitors have already proven to be effective therapies for erectile dysfunction, COPD, and psoriatic arthritis. This family of enzymes also plays a role in diseases and disorders of the CNS such as depression, anxiety, schizophrenia, and Alzheimer's Disease. Unfortunately no effective PDE inhibitors have been developed for the treatment of these diseases. The proposed book will be a comprehensive overview of the current state of basic and translational research on PDE inhibitors written by internationally recognized experts. Authors will also discuss potential PDE subtypes and splice variants in the hopes that this will spur more creative approaches to PDE targeting drugs.

Book excerpt: This report from the Committee on Military Nutrition Research reviews the history of caffeine usage, the metabolism of caffeine, and its physiological effects. The effects of caffeine on physical performance, cognitive function and alertness, and alleviation of sleep deprivation impairments are discussed in light of recent scientific literature. The impact of caffeine consumption on various aspects of health, including cardiovascular disease, reproduction, bone mineral density, and fluid homeostasis are reviewed. The behavioral effects of caffeine are also discussed, including the effect of caffeine on reaction to stress, withdrawal effects, and detrimental effects of high intakes. The amounts of caffeine found to enhance vigilance and reaction time consistently are reviewed and recommendations are made with respect to amounts of caffeine appropriate for maintaining alertness of military personnel during field operations. Recommendations are also provided on the need for appropriate labeling of caffeine-containing supplements, and education of military personnel on the use of these supplements. A brief review of some alternatives to caffeine is also provided.
