Book excerpt: Polymeric nanoparticle (NP) drug carriers present a promising technology for controlled release since they are capable of improving the encapsulation efficiency and stability of the drugs inside the NPs and also able to provide effective drug levels over a longer period of time, compared to traditional therapy. However, before the NP drug delivery technology becomes a reality, important parameters of NPs like size, drug loading ability and sustained release kinetics must be well investigated and optimized in order to minimize the adverse effects of chemotherapeutic compounds and prolong the drug releasing profile in a controlled manner. In order to accomplish this objective, this thesis proposed two novel methods for synthesis of NPs as drug delivery carriers, with assistance from bulk and microfluidic technologies, for hydrophobic and hydrophilic drugs, individually. For encapsulation of hydrophobic drugs, a modified flow focusing method was developed on a glass capillary microfluidic platform. Unlike conventional microfluidic flow focusing using two miscible phases, an insoluble component (DCM) was introduced into the dispersed phase to form a partially water-miscible precursor, and a transformation phenomenon of “jet-micro droplets--nanoparticles” was firstly observed instead of the “jet-micro droplets” or “jet-nanoparticles” from traditional flow focusing. Using Doxorubicin as a drug model, size-tunable Doxorubicin-PLGA NPs (80~170 nm) were synthesized by adjusting the flow rates, polymer concentration and the volume fraction of DCM in dispersed phase with an excellent monodispersity (PDI=0.1~0.2) which was superior to those from conventional flow focusing. We also found that drug loading content increased when volume ratio of DCM/DMSO in dispersed phase increased, with a considerable mass loading ratio up to 26.3%. In addition, Doxorubicin-PLGA NPs synthesized with DCM/DMSO precursor exhibited a slower drug release profile than those synthesized with pure DMSO precursor. This modified flow focusing method can also be extended to encapsulate inorganic compounds, such as iron oxide (Fe2O3) for a combination of chemotherapy and thermo-therapy, and showed a better loading ability of Fe2O3 than conventional research using pure DMSO. This method successfully combined the advantages from previous classical drug encapsulation techniques: small particle size, ease to operation-like nanoprecipitation; monodispersity, high drug encapsulation efficiency-like emulsion-based methods, provided us a promising tool for preparing nanoparticle carriers for multiple drug loading of both organic drugs and inorganic compounds For encapsulation and release of hydrophilic drugs, a modified bulk drop-wise nanoprecipiation method was designed by separating drug and polymer into aqueous and DMSO phases, respectively. In this case, we successfully solved the problem of the poor solubility of hydrophilic drug in organic solvents, for which reason the traditional nanoprecipitation method was limited to the application of hydrophilic drug encapsulation. Monodisperse ciprofloxacin-loaded PLA (poly (D,L-lactide))-Dextran and PLGA-PEG (poly (lactide-co-glycolide)-block-poly (ethylene glycol)) NPs were prepared of a tunable size range (80~200 nm). The drug loading ability, up to 18.6% (w/w), was found having an excellent linear correlation with the original feed of the ciprofloxacin drug, which indicated that drug content encapsulated by the NPs could be precisely controlled and an in-vitro sustained release was achieved up to 95.4% in 6 days. This thesis demonstrated the design and mechanism of different drug encapsulation and release systems; and synthesis, characterization, and optimization of drug-loaded polymeric nanoparticles. Our novel drug delivery systems significantly improved the encapsulation efficiency of various therapeutic compounds and exhibited a sustained-release profile. These nano-drug-delivery systems exploited intrinsic properties of NPs for controlled release, and will not only benefit the field of nanobiomedicine, but also could be further applied to food, flavor, fragrance and cosmetics industry.

The combination of the two disciplines created the modern field of nanomedicine, in which drug delivery is one of the most prominent branches. Several aspects are involved in drug delivery; this work will focus on the drug delivery vehicle. In particular three aspects will be investigated: building material, internal structure and material compatibility. In a first project DNA was proposed as an innovative building material. DNA nanoparticles were made from self-folding of long concatameric repeats of a single strand sequence. Nanoparticles with different sequences created a library that was bio- panned against dendritic cells (DC). Particles from the enriched library were sequenced and individually tested for affinity towards DC. The use of DNA as building material offers several advantages. For instance DNA binding drugs (such as Doxorubicin) can be easily incorporated, and immunostimulatory sequences (such as GpC) and any other encoding sequence can be integrated within the concatamers. In addition, any other molecule or small particle of interest can be conjugated to a short complementary sequence and hybridized on the outer layer of the DNA nanoparticle. DNA nanoparticles' payloads are limited to hydrophilic drugs. In addition to an hydrophobic payload, some therapies require a high loading and steady release. To achieve such results a gradient structure was created within the core of a polymeric nanoparticle. Physical and chemical gradient were considered. A chemical gradient was created by combining a low molecular weigh polycaprolactone (PCL) to a higher molecular weigh poly(lactic-co-glycolic acid) (PLGA). PCL and PLGA have different degradation rate and hydrophobicity. The particles created by combining the two polymers showed properties (such as loading) dependent on the two polymers' proportion into the composition. The chemical gradient nanoparticles are characterized by high loading and steady release of hydrophobic drugs. A new technique was examined to encapsulate hydrophilic oligonucleotides into hydrophobic nanoparticles. PMAL-C8, a zwitterionic polymer, interacts with oligonucleotides of different length and improves their loading into the hydrophobic core. The nanoparticles created were successfully used in transfection experiments, showing the bioavailability of the loaded oligonucleotides.

For the most part, these consisted of a matrix of poly(lactic-co-glycolic acid) (PLGA), polyethylene glycol (PEG), and polyvinyl alcohol (PVA) containing encapsulated anticancer drugs as chemotherapy agents. The drug release from biodegradable nanoparticles was analyzed mathematically using new approaches that simultaneously incorporates the three major mechanisms of release: initial burst, nanoparticle degradation-relaxation, and diffusion. The theoretical release studies were corroborated experimentally by evaluating the cytotoxicity effectiveness of PHT-427-loaded nanoparticles over pancreatic cancer cells in vitro. These studies showed that the encapsulated PHT-427 drug in the nanoparticles is more accessible and thus more effective when compared with the drug alone. Also, the PHT-427-loaded nanoparticles cytotoxicity was evaluated in vivo studies with pancreatic tumors. The results show that the drug is more effective when is loaded into polymeric nanoparticles compared to drug alone, by reducing orthotopic pancreatic tumor growth. In addition, a selection of hydrophobic to hydrophilic drugs were encapsulated into polymeric nanoparticles to find optimal drug loadings by using single or double emulsification techniques. The release of these drugs from PLGA nanoparticles was evaluated to determine the overall release profile characteristics. The encapsulation of the drug pemetrexed was improved by using polyethileneimine. The high positive charge density of polyethileneimine causes a strong electrostatic interaction with the carboxylic acids of pemetrexed; this complex decreases the solubility of pemetrexed and boosts the encapsulation efficiency. Additionally, a drug release mathematical analysis that considers the effects of the temperature of release was effectively established. The analysis was performed by using two different models: the first one simultaneously incorporates the mechanisms of initial burst and nanoparticle degradation - relaxation, and the second model, besides of the mechanisms of the first model, includes the diffusion of the drug. Both models were successfully employed to describe the experimental release of rhodamine 6G from PEGylated nanoparticles at different temperatures. From the parameters obtained by the fit using each model, it was possible to define a set of new relations of the form of Arrhenius to estimate the parameters of release at other temperatures.

This dissertation focuses on the rational design, development and application of well-defined polymeric nanoparticles, capable of high loading of both hydrophobic and hydrophilic therapeutic agents, toward the effective treatment of lung diseases. In the first study, cisplatin was loaded into non-degradable poly(acrylic acid)-b-polystyrene-based SCKs through the formation of coordination bonds between platinum and carboxylate groups in the nanoparticle shell domain. The effects of crosslinking were investigated by comparing drug loading & release, in vitro cytotoxicities, and immunotoxicities. In another study, degradable polyphosphoester-based polymeric micelles and SCKs, each derived from non-cytotoxic, amphiphilic block-graft terpolymers, were specifically designed and synthesized for anti-cancer drug paclitaxel (PTX) delivery toward the treatment of osteosarcoma lung metastases. PTX could be encapsulated into either micelles or SCKs, with overall PTX concentration as high as 4.8 mg/mL vs. the low solubility for free PTX in water of less than 2.0 [lowercase Mu symbol]g/mL. In vivo biodistribution indicated that both micelles & SCKs underwent extravasation from the lung in a controlled manner, while crosslinking slowed the rate of extravasation significantly. Moreover, hydrophilic silver cations were also attached to the nanoparticles via the interaction between silver and alkyne as a potential treatment for bacterial pulmonary infections. The well-defined Ag-loaded nanoparticles released silver in a controlled and sustained manner over 5 days, and displayed enhanced in vitro antibacterial activities against cystic fibrosis-associated pathogens and decreased cytotoxicity to human bronchial epithelial cells, in comparison to silver acetate.
