Book excerpt: The use of N-heterocyclic carbenes (NHCs) as ligands in catalysis is one of the most significant developments in modern catalysis and organometallic chemistry. One way to extend the scope of NHC ligand tuning is by means of annulation of carbocyclic and heterocyclic rings to the NHC backbone. The bis(imino)acenaphthene-supported N-heterocyclic carbene [IPr(BIAN)] has been synthesized and can be regarded as originating from the fusion of a naphthalene ring to an NHC. Several metal complexes of IPr(BIAN), including those incorporating copper(I), silver(I), gold(I), or iridium(I) have been synthesized and characterized, including single-crystal X-ray diffraction studies. The doncity of IPr(BIAN) was investigated using the Tolmen Electronic Parameter (TEP) method. A TEP value of 2042 cm−1 was calculated for the IPr(BIAN) ligand using the Ir(CO)2Cl complex which indicates that IPr(BIAN) is a relatively strong electron donating NHC ligand. The well-behaved redox chemistry of the BIAN ligand class rendered IPr(BIAN) an excellent candidate for exploration of the relationship between ligand charge and carbene donicity. The electrochemical reduction of IPr(BIAN) was studied by cyclic voltammetry (CV) in a THF solution and a reversible reduction wave was detected at - 1.79 V vs SCE. Spectroelectrochemical IR studies were also undertaken to further characterize the nature of the reduced state. IPr(BIAN) was found to be a stronger electron donating ligand in the reduced state in comparison with the neutral state of the ligand. IPr(BIAN) was also chemically reduced using potassium graphite and the resulting radical anion was studied by electron paramagnetic resonance (EPR) techniques. An isotropic EPR signal was observed at a g value of 2.0112. Due to the known antimicrobial activities of silver and gold NHCs, the activities of the silver and gold complexes of IPr(BIAN) and the imidazolium salts of several BIAN ligands were investigated using the minimum inhibitory concentration test. The silver(I) and gold(I) complexes of IPr(BIAN) were found to be moderately active. The most active compounds were found to be the imidazolium salts, with MIC values ranging between

Book excerpt: "This dissertation discusses a newly growing area of the synthesis from xanthine ligands of new silver(I) N-heterocyclic carbene (NHC) complexes and their potential antimicrobial use in cystic fibrosis lung infections. In chapter I, the ligand properties of NHCs and the synthetic methods for the formation of silver(I) NHC complexes are outlined. An overview of silver antimicrobials and previously synthesized silver(I) NHC complexes from pyridine linked and tripodal imidazolium salts for the topical applications is discussed. Furthermore, a brief summary of cystic fibrosis is given. Chapter II presents the introductory studies to explore the NHC silver(I) chemistry of caffeine. Formation and characterization of mono and dinuclear bis(NHC) silver(I) complexes from different methylated caffeine salts are given. Chapter III focuses on the formation of an NHC-silver(I) acetate complex from the iodide salt of methylated caffeine. This complex demonstrates a promising antimicrobial activity against numerous resistant respiratory pathogens including members of the Burkholderia cepacia complex which causes a high rate of mortality in patients with cystic fibrosis. Synthesis of NHC silver(I) complexes from theophylline derivatives is given in chapter IV. The antimicrobial activity of these complexes against the same pathogens used in chapter III is presented. In chapter V, the use of a bis(NHC) silver(I) complex from methylated caffeine as a carbene transfer agent is demonstrated and formation of a Rh(I) NHC complex is outlined. Furthermore, the synthesis and characterization of Pt(II) and Pd(II) NHC complexes of methylated caffeine are presented."--Abstract.

Book excerpt: Bifunctional organometallic catalysts offer unique opportunities for new and improved reactivity and selectivity. The combination of a metal's d electrons and a pendant base or acid in the vicinity of an organometallic active site leads to substantial improvement of reaction rate in many key reactions. Here we report a synthesis of the first example of an imidazol-2-yl substituted bifunctional N-heterocyclic carbene ligand (NHC) and its coordination to a variety of metals. The metal complexes have been characterized by IR, NMR, and X-ray crystallography. The catalytic activity of these new NHC-metal complexes in hydrogenation reactions (e.g. Oppenauer-type alcohol oxidation, ß-alkylation of secondary alcohols with primary alcohols, N-alkylation of amines with alcohols), hydroamination, and azide-alkyne cycloaddition reactions is described.

Book excerpt: "New symmetric and asymmetric types of N-heterocyclic carbene (NHC) precursors, grouped into mono-dentate, bi-dentate and tri-dentate chelating ligands, were synthesized. The reaction of the carbene precursors with silver(I) oxide in methanol at room temperature affords the silver(I) N-heterocyclic carbene complexes. The solubility and stability of silver(I) NHC complexes in water depends on the ligand and counter anion. In-vitro test of water soluble silver(I) NHC complex, [AgC1--N5O2H21] [OH]− 3a shows antimicrobial activity as good as 0.5 % AgNO3 using the same amount of silver on pathogenic bacteria and fungi of clinical importance. The encapsulation of Ag(I) NHC complexes, [Ag2C34N10O4H]2 2[OH]− 6 and [Ag3C54N12O2H]3+[Br]− 2[OH]− 8a respectively by electrospinning technique, delivers bioactive nanosilver ions which improved the antimicrobial activity of the Ag(I) NHC over the un-encapsulated forms. The electrospun mat of 6 sustained the release of bioactive nanosilver ions for at least seven days. Electrospun fiber mat containing 75% of 6 showed improved antimicrobial efficacies in-vitro over silver sulfadiazine cream (SSD), with the rate of bactericidal activity six fold faster than SSD and ca. the same rate as 0.5% AgNO3. The amount of silver in the fiber mat is six times lower than SSD and 8 fold lower than 0.5% AgNO3. The size of the nanosilver ions of electrospun mat of 6 is

Available in PDF, EPUB and Kindle. Book excerpt: Direct Synthesis of Metal Complexes provides in-depth coverage of the direct synthesis of coordination and organometallic compounds. The work is primarily organized by methods, but also covers highly relevant complexes, such as metal-polymer coordination compounds. This updated reference discusses recent developments in cryosynthesis, electrosynthesis, and tribosynthesis (popular as it doesn’t require organic solvents), with special attention paid to ‘greener’ methodologies and approaches. Additionally, the book describes physical methods of zero-valent metal interaction with organic matter, including sputtering, ultrasonic treatment and synthesis in ionic liquids. The book presents completely new content as a follow-up to the 1999 Elsevier Science publication Direct Synthesis of Coordination and Organometallic Compounds that was edited by Dr. Garnovskii and Dr. Kharisov. Covers current methods and techniques of metal interactions with organic media leading to metal chelates, adducts, di- and polymetallic complexes, metal-containing macrocycles, supported coordination compounds (i.e., metal complexes on carbon nanotubes), and more Describes reactivities of distinct forms of elemental metals (powders, sheets, nanoparticles (including a host of less-common metal nanostructures) with organic phase (liquid, solid and gaseous) and water Includes experimental procedures, with examples of direct synthesis, at the end of each chapter

Book excerpt: Since the development of the first metal N-heterocyclic carbenes (NHC) by Ofele and Wanzlick in the 1960s, investigation into this area has grown dramatically, especially after the synthesis of the first stable free metal carbene in 1991 by Arduengo. Since that time, metal carbenes have been synthesized for applications related to catalysis and more recently therapeutics. Research in the Youngs group has focused on applying the synthetic routes of Lin to make a variety of silver NHCs for medicinal purposes. More recently, encapsulation of the silver NHCs into nanoparticles have been investigated for sustained release and systemic delivery of the therapeutics. This dissertation will focus on both the synthesis of some new silver NHCs as well as their encapsulation into biodegradable nanoparticles. Chapter 1 discusses the advantage of using silver as an antimicrobial and anticancer agent, as well as why NHCs were chosen. It will also provide an overview of research in the area of nanoparticles that are relavent to encapsulation of our silver NHC complexes. Chapter 2 details the synthesis of two new silver NHCs with methylbenzoate substituents attached, as well as the antimicrobial activity of these compounds. Chapter 3 describes a series of mono-halogenated compounds that were synthesized in order to determine their water-stability using 1H NMR spectroscopy. Chapter 4 will discuss the synthesis of one of our most active compounds to date as well as its encapsulation into the biodegradable, FDA approved nanoparticle poly(ethylene glycol)-poly(lactic acid) (PEG-PLA). The efficacy of the silver NHC-encapsulated nanoparticle on a small-cell lung cancer line is also presented. Finally, Chapter 5 will describe the in vivo toxicity results of the same nanoparticle system performed on two different strains of mice utilizing two different delivery methods, including nebulization and intravenous injection, to demonstrate the potential of this silver NHC-nanoparticle for antimicrobial and anticancer applications.
