Book excerpt: In this study, novel rhodium(I) carbene complexes were synthesized and fully characterized via a carbene ligand transfer methodology from Group 6 Fischer carbene complex precursors and subsequent ligand modification. The classic Fischer route was followed towards the isolation of mono- and biscarbene complexes of the form [M(CO)5{C(OR)R'}] [M = Cr, W; R = Me, Et; R' = 2-furyl, 2-thienyl, ferrocenyl] (complexes 1 6, 9 and 10), and [M(CO)5?{C(OR)-R''-C(OR)}M(CO)5] [M = Cr; R = Me, Et; R'' = 2,2'-bithien-5,5'-diyl, 2,5-furadiyl, 1,1'-ferrocendiyl] (complexes 7, 8 and 11), respectively. Analogous aminocarbene complexes [M(CO)5{C(NH2)R'}] [M = Cr, W; R' = 2-thienyl, ferrocenyl) (complexes 12 and 13) were prepared by simple aminolyses of the alkoxycarbene complex precursors and all isolated products were characterized using NMR and FT-IR spectroscopic methods, the results of which were comparable with literature values. Transmetallation techniques were employed in an attempt to transfer the carbene ligands to a rhodium(I) metal center of the dimeric [Rh(cod)Cl]2 precursor to result in novel 2-furyl, 2-thienyl and ferrocenyl Fischer carbene complexes of rhodium(I). Only the ferrocenylcarbene complex 15 [Rh(cod)Cl{C(OEt)Fc) were found to be stable enough to isolate, as the heteroaryl (thienyl, furyl) substituted carbene ligands dissociated in solution, with resultant decomposition dimerization to form the corresponding alkene and starting [Rh(cod)Cl]2 complex, as indicated by NMR spectroscopy. The ferrocenylcarbene complex 15 was then employed as a precursor for the syntheses of all other rhodium(I) carbene complexes via cod ligand substitution and aminolysis reactions to isolate mono- and dicarbonylcarbene complexes 16 23, [Rh(LL)Cl{C(X)Fc}] [LL = cod, (CO)2, (CO, PPh3), (CO, PCy3), (CO, P(OPh3)), (CO, AsPh3); X = OEt,NHnPr], with variable ?-acceptor properties. Full characterization of the novel complexes were achieved by single crystal XRD and spectroscopic methods. From the FT-IR data collected, the donor ability of the electronic environment around the rhodium(I) center was found to correlate with the known electron-donor ability of the coligands in the order PCy3>PPh3,AsPh3>P(OPh)3. This trend was corroborated by cyclic voltammetric methods through which the electron-withdrawing effects of the coligands were studied, and it was confirmed that the cod ligand is the most electrondonating whilst the dicarbonyls were found to be the least donating in the series. In addition, the increased electron donation of the aminocarbene ligands compared to the ethoxycarbene ligands was found to significantly influence the redox potentials of the metal centre in the studied complexes. The isolated rhodium(I) Fischer carbene complexes 15 - 22 were screened as catalyst precursors for the hydroformylation of 1-octene. Good to excellent catalytic activities, with selectivity toward the formation of the linear nonanal, was observed. These results were found to be comparable to results reported for rhodium(I) N-heterocyclic carbene complexes. A mercury-drop test was done to exclude a heterogeneous catalytic mode of action. Finally, the stability of the catalyst precursor 15 (and 17) was probed by an NMR experiment carried out under hydroformylation conditions. The Rh-Ccarbene bond is retained, although the presumed catalytically active species, the rhodium carbene carbonyl hydride complex could not be identified.

Book excerpt: The synthesis and electronic characterization of two new electrophilic carbenes is described. The synthesized carbenes were selected based on computational studies investigating the HOMO and LUMO energies of various new carbenes. The cyclic(amido)arylcarbene (CAmArY) could not be isolated due to rapid dimerization when the carbene was generated. The carbene dimer was characterized through NMR, mass spectrometry, and X-ray crystallography. A sulfur trapping experiment was performed to confirm the generation of the carbene. The pi-accepting ability of CAmArY was studied via selenium NMR. As of this writing, the CAmArY selenium adduct is the most pi-accepting carbene with a selenium NMR shift of 1240 ppm. Tolman Electronic Parameter (TEP) studies could not be completed due to decomposition of the iridium and rhodium metal complexes which is discussed in further detail below. A series of diiminocarbenes (DICs) were synthesized in good yields. DICs were isolated successfully and characterized through NMR and a single crystal X-ray structure of one free DIC was obtained. The pi-accepting ability of the DICs were also studied via 77Se NMR. The DIC selenium adducts displayed 77Se NMR shifts ranging from 640 - 699 ppm, which suggests a pi-accepting ability greater than CAAC but less than the DACs. TEP studies were performed by synthesizing the iridium (I) dicarbonyl chloride complex of the parent DIC and examining the stretching frequencies of the metal carbonyls via IR spectroscopy. The TEP value of the DIC was determined to be 2046 cm 1 which places its overall donating ability very similar to CAAC. To determine the effect of para-substitution of the arylimino substituents, DFT calculations were performed and a Hammett analysis was performed which showed that the level of the LUMO energy was more effected than the HOMO energy level. The selenium adducts of some of these DIC derivatives were prepared to conduct a Hammett analysis by plotting the 77Se NMR chemical shift versus the respective LUMO level. This Hammett analysis revealed a direct correlation of the LUMO energy level to the 77Se NMR shift. The synthesis of two other carbenes, an abnormal diamidocarbene (aDAC) and an indole derived carbene (IC) has been started.The synthetic routes currently being employed to synthesize the carbene precursors is described herein along with future efforts to prepare and characterize these carbenes.

Book excerpt: Since the discovery of the first stable silver N-heterocyclic carbene complex by Arduengo in 1991, the field of silver N-heterocyclic carbene complexes has expanded greatly. These complexes have been studied extensively for use in catalysis, as transmetallation agents, and more recently for pharmaceutical applications. Silver has been used for centuries as an anti-microbial agent, and more recently has been shown to have anti-tumor activity as well. Combination with the easily modified N-heterocyclic carbene ligand yields a drug that can be rationally designed for increased water solubility, increased lipophilicity, or to increase activity. These complexes have shown great results, and will be the main focus of this dissertation. Chapter 1 of this dissertation reviews silver N-heterocyclic carbene complexes as promising antimicrobial agents and the evolution of the field over the past two decades. Chapter 2 discusses the synthesis of a new N-heterocyclic carbene backbone, 4,5,6,7-tetrachlorobenzimidazole, and the corresponding silver complexes for antimicrobial use. These complexes were then examined in vitro against a panel of clinical bacteria, displaying inhibitory concentrations between 0.25-4 [micrograms]/mL. Additionally, two of the silver complexes displayed antimicrobial activity against a silver-resistant E. coli strain. Chapter 3 discusses the rational design of several new naphthalene containing imidazolium cations, the precursors to silver N-heterocyclic carbene complexes, for the use as anti-tumor drugs. These new imidazolium cations have been designed to increase water solubility, anti-tumor activity, and encapsulation efficiency into biodegradable delivery devices through modification of the substituents. These cations have demonstrated in vitro anti-tumor activity comparable to cisplatin against three types of lung cancer. The anti-tumor activity increased with the increase in the length of the alkyl chain substituent. Additionally, it was noted that the position of the alkyl chain on the naphthalene rings did not affect the anti-tumor activity of the cations. Chapter 4 discusses the in vitro anti-tumor activity of silver carbene complexes based on selected imidazolium cations from chapter 3. These complexes have demonstrated an increase in water solubility in comparison to previously reported silver N-heterocyclic carbene anti-tumor complexes. Additionally, the complexes have displayed anti-tumor activity at clinically relevant levels, demonstrating IC50 values ranging from 5-25 [micrograms]/mL. In general, the anti-tumor activity increased as the length of the alkyl chain on the ligand increased. The highest activity was displayed by SCC44, which unlike the other complexes contained two naphthalene rings.

Book excerpt: As carbon dioxide levels continue to rise in our atmosphere, scientific interest has peaked around the capture and utilization of CO2. Not only does CO2¬¬ have the potential to be used as a C1 building block for the production of value added chemicals, but CO2 also has the potential to be used as a carbon neutral hydrogen storage material in the form of formic acid. Although catalysts for CO2 reduction exist, many of these catalysts require the use of high temperatures and pressures and are not stable for prolonged exposure to the reaction conditions. Therefore, the challenge of making robust catalysts for CO2 hydrogenation that can operate under mild conditions with high activity remains outstanding. With the goal of generating a robust and highly active CO2 hydrogenation catalyst in mind, this thesis describes the fundamental metalation chemistry of a novel tripodal bis(protic N-Heterocyclic carbene)-phosphine ligand with ruthenium precursors and the reactivity of the resulting organometallic complexes with CO2. Chapter 1 provides a brief overview of CO2¬ in the earth’s atmosphere, a glimpse at CO2 hydrogenation chemistry, and an introduction to traditional and protic N-heterocyclic carbene (PNHC) chemistry. Chapter 2 describes the synthesis and characterization of PNHC Ru complexes utilizing [Cp*RuCl]4 as the ruthenium precursor. Chapter 3 investigates the coordination chemistry and synthesis of PNHC Ru complexes stemming from [(Arene)Ru] precursors. Chapter 4 describes both stoichiometric and catalytic reactivity studies of complexes synthesized in Chapters 2 and 3 with CO2. Finally, Chapter 5 dives into an entirely new subject and discusses the crystallographic structure determination of an unprecedented In¬¬37P20¬ nanocluster.
