Book excerpt: Three distinct types of contractions perform colonic motility functions. Rhythmic phasic contractions (RPCs) cause slow net distal propulsion with extensive mixing/turning over. Infrequently occurring giant migrating contractions (GMCs) produce mass movements. Tonic contractions aid RPCs in their motor function. The spatiotemporal patterns of these contractions differ markedly. The amplitude and distance of propagation of a GMC are several-fold larger than those of an RPC. The enteric neurons and smooth muscle cells are the core regulators of all three types of contractions. The regulation of contractions by these mechanisms is modifiable by extrinsic factors: CNS, autonomic neurons, hormones, inflammatory mediators, and stress mediators. Only the GMCs produce descending inhibition, which accommodates the large bolus being propelled without increasing muscle tone. The strong compression of the colon wall generates afferent signals that are below nociceptive threshold in healthy subjects. However, these signals become nociceptive; if the amplitudes of GMCs increase, afferent nerves become hypersensitive, or descending inhibition is impaired. The GMCs also provide the force for rapid propulsion of feces and descending inhibition to relax the internal anal sphincter during defecation. The dysregulation of GMCs is a major factor in colonic motility disorders: irritable bowel syndrome (IBS), inflammatory bowel disease (IBD), and diverticular disease (DD). Frequent mass movements by GMCs cause diarrhea in diarrhea predominant IBS, IBD, and DD, while a decrease in the frequency of GMCs causes constipation. The GMCs generate the afferent signals for intermittent short-lived episodes of abdominal cramping in these disorders. Epigenetic dysregulation due to adverse events in early life is one of the major factors in generating the symptoms of IBS in adulthood.

Available in PDF, EPUB and Kindle. Book excerpt: The endothelium, a monolayer of endothelial cells, constitutes the inner cellular lining of the blood vessels (arteries, veins and capillaries) and the lymphatic system, and therefore is in direct contact with the blood/lymph and the circulating cells. The endothelium is a major player in the control of blood fluidity, platelet aggregation and vascular tone, a major actor in the regulation of immunology, inflammation and angiogenesis, and an important metabolizing and an endocrine organ. Endothelial cells controls vascular tone, and thereby blood flow, by synthesizing and releasing relaxing and contracting factors such as nitric oxide, metabolites of arachidonic acid via the cyclooxygenases, lipoxygenases and cytochrome P450 pathways, various peptides (endothelin, urotensin, CNP, adrenomedullin, etc.), adenosine, purines, reactive oxygen species and so on. Additionally, endothelial ectoenzymes are required steps in the generation of vasoactive hormones such as angiotensin II. An endothelial dysfunction linked to an imbalance in the synthesis and/or the release of these various endothelial factors may explain the initiation of cardiovascular pathologies (from hypertension to atherosclerosis) or their development and perpetuation. Table of Contents: Introduction / Multiple Functions of the Endothelial Cells / Calcium Signaling in Vascular Cells and Cell-to-Cell Communications / Endothelium-Dependent Regulation of Vascular Tone / Conclusion / References

The distinguished scientists and clinicians open a new area of endocrinology, which sees the vasculature and kidney as endocrine organs and sheds new light on the hormonal regulation of the cardiovascular system. They also add significantly to our current understanding of the autocrine, paracrine, and endocrine regulation of vascular function in health and in those vascular disease states that accompany diabetes mellitus, dyslipidemia, and hypertension. Endocrinology of the Vasculature is the first book to comprehensively and systematically explore the vasculature as an endocrine organ.

Book excerpt: Functional Magnetic Resonance Imaging (fMRI) has become a standard tool for mapping the working brain's activation patterns, both in health and in disease. It is an interdisciplinary field and crosses the borders of neuroscience, psychology, psychiatry, radiology, mathematics, physics and engineering. Developments in techniques, procedures and our understanding of this field are expanding rapidly. In this second edition of Introduction to Functional Magnetic Resonance Imaging, Richard Buxton – a leading authority on fMRI – provides an invaluable guide to how fMRI works, from introducing the basic ideas and principles to the underlying physics and physiology. He covers the relationship between fMRI and other imaging techniques and includes a guide to the statistical analysis of fMRI data. This book will be useful both to the experienced radiographer, and the clinician or researcher with no previous knowledge of the technology.

Available in PDF, EPUB and Kindle. Book excerpt: The study of the actions of drugs on smooth muscle has been a preoccupation of many pharmacologists almost from the beginning of the discipline. To a con siderable degree, the development of theories to explain drug actions on smooth muscle has occurred somewhat independently of the development of our knowledge of the physiology, biochemistry, and biophysics of smooth muscle. This knowledge has developed rapidly in the past decade, and some of its consequences for our understanding of drug-receptor interactions in smooth muscle have not always been fully appreciated or accepted. One of the purposes of this volume is to provide pharmacologists with some understanding of the physiology, biophysics, and bio chemistry of smooth muscle and of related advances in methodology so as to facilitate the incorporation of such knowledge and related methods into future pharmacological studies of smooth muscle and drug interactions. Another purpose of the book is to provide both graduate students and in vestigators in pharmacology and related disciplines with a summary of the numerous methods that have evolved or are available for the study of drug and smooth muscle interactions, and, in particular, to highlight their possible uses and limitations. Perhaps, because of the diversity in content and difficulty of these methods, there has to our knowledge never been a previous attempt to bring them together in one place. We have not, of course, succeeded entirely in this objective.

Available in PDF, EPUB and Kindle. Book excerpt: The three different types of muscle tissue found in the animal kingdom are cardiac, skeletal, and smooth. The muscle cells are not only complex but also fascinating. In recent years there has been substantial advances in our understanding of muscle cell biology, especially in areas of molecular anatomy, basic physiology, understanding disease mechanisms, and therapeutic targets. Consequently, this book mainly focuses not only on the biology of myocytes, but also on all-encompassing disciplines pertaining to muscle tissue, such as fundamental physiology, molecular mechanisms of diseases, muscle regeneration, etc. for all three types of muscle, namely, skeletal, cardiac, and smooth muscle. As a result, the goal of this book is to consolidate the recent advances in the area of muscle biology/diseases/regeneration covering a broad range of interrelated topics in a timely fashion and to disseminate that knowledge in a lucid way to a greater scientific audience. This book will prove highly useful for students, researchers, and clinicians in muscle cell biology, exercise physiology/science, stem cell biology, developmental biology, cancer biology, pathology, oncology, as well as tissue engineering and regenerative medicine. This quick reference will benefit anyone desiring a thorough knowledge pertaining to recent advances in muscle biology in the context of health and disease.
