Book excerpt: This workshop was the second of this series held on the island of santorini in the Cycladic Sea. The first one ("Mechanism of Action of the Nicotinic Acetylcholine Receptor", NATO ASI Se ries H, vol. 10) took place in May 1986 and focused on what was at the time the best studied of all neuroreceptors. This second one, held only two years later, demonstrates the im mense progress achieved since then in the field of neurorecep tors and ion channels. Molecular cloning techniques have now made available the primary structures of a whole array of ion channel proteins, and this in turn has shed light on some gen eral principles of the structure-function relationships of these central elements of intercellular communication. The purpose of this workshop was to explore the common ele ments in gene and protein structure of already cloned ion channel proteins, and to assess the status of other cloning projects in progress. It explicitly focused on very recently published and unpublished results. All participants kept to these goals thereby demonstrating the very value of such work shops for the progress of science.

Available in PDF, EPUB and Kindle. Book excerpt: Ion channels are crucial components of living cells. Situated in the cell's membranes. they allow particular ions to pass from one side of the membrane to the other. In recent years the patch clamp technique has allowed the activity of individual channels to be measured, and recombinant DNA technology has led to fascinating detail on their structure. Together, these technical advances have produced a great flowering of knowledge and understanding about the subject, itself leading to further breakthroughs in science and medicine. Ion Channels provides an introduction to this scientific endeavour. It emphasises the molecular structure of channels as determined by gene cloning technology. This knowledge illuminates discussions of the permeability and selectivity of channels, their gating and modulation, their responses to drugs and toxins and the human diseases caused when they do not function properly.

Book excerpt: Ion channels are intimately involved in the everyday physiological functions that enable us to live a full and varied life. When disease strikes, malfunction of ion channels or their dependent is often involved, either as the cause or the effect of the illness. Thus, billions of dollars have been, and still are being, invested in research to understand the physiological and pathophysiological functions of ion channels in an attempt to develop novel therapeutic treatments for a wide range of diseases. This book provides a comprehensive overview of ion channel structure and function. It comprises two major parts. Part one is an introductory overview of the ion channel superfamily and the generic aspects of ion channel function. This part also reviews the methodologies by which ion channel function can be studied from the perspective of performing detailed biophysical characterization through to the deployment of high throughput approaches for identifying novel ion channel ligands. Part two of the book provides an in-depth review of the individual ion channel subfamilies and, as such, is subdivided into four broad sections: Voltage-Gated Ion Channels, Extracellular Ligand-Gated Ion Channels, Intracellular Ligand-Gated Ion Channels, and Polymodal-Gated Ion Channels, with each chapter focused on specific family members. These chapters have been written by world leading experts and provide a detailed overview of the structure, biophysics, localization, pharmacology, physiology, and disease relevance of each particular ion channel subfamily. Reviewing both the basic principles of ion channel function and providing a detailed up-to-date review of the phsyiological and pharmacological aspects of individual ion channel sub-families, this book constitutes both an excellent introduction to the field for non-specialists, as well as a highly valuable reference text for experienced researchers already working in the ion channel area.

Available in PDF, EPUB and Kindle. Book excerpt: Omnis cellula e cellula, "every cell from a cell," was dogma to the 19th century cellular physiologist and the cornerstone of Virchow's Cellular pathologie. "Spread out a cell into a layer and you will find that, in ceasing to be a cell, it has ceased to act as such," wrote the British 1 physiologist G . R. Lewes more than a century age. "The cell remains vital as long as its wall remains intact . . . " keeping its content "pure and clear" and thus preserving the "vital principle" within, echoed Claude 2 Bernard a few years later. The notion of the cell membrane as a pro tecting envelope held sway until it became clear that it could not account for the "coalescence" of poorly differentiated embryonic "vesicles" and for their transformation into "cell-like structures" capable of auto regulation and yet subject to what the grandfather of one of us defined as the "federal obligations imposed by the whole organism. ,,3 A new concept was needed, and soon the membrane was described as a structure capable of uniting as well as separating adjacent cells. Morphologic evidence for this dual function was obtained several years later when the electron microscope revealed the existence of tight and gap junc tions which, acting as intercellular bonds and channels, allowed the cells to communicate with one another and thus coordinate their biologic activities.
