Advances in molecular genetic technology and understanding of the bovine genome have led to the development of tools that can be used to enhance profitability on cow-calf enterprises. Factors Affecting Calf Crop: Biotechnology of Reproduction provides a detailed compilation of current and forthcoming technology for managing reproduction in cattle. The book discusses topics such as: approved techniques for controlling the estrous cycle in cattle; managing follicular growth with progesterone, estrogens, and prostaglandins; freezing, thawing, and transfer of cattle embryos; application of embryo transfer to the beef cattle industry; embryo transfer in topically adapted cattle; new factors affecting bull fertility; embryo collection and utilization technology, in vitro fertilization, somatic cell cloning, and genetic technologies; uses of real-time ultrasound; and sexed semen. Over 25 leading animal scientists have combined their expertise to produce the first single-source reference that covers successful reproductive techniques that will, most likely, be the wave of the future. Expansive in scope, the book addresses current biotechnologies as they impact the production of beef cattle. Written at a level to appeal to the researcher, commercial producer, or student, Factors Affecting Calf Crop: Biotechnology of Reproduction presents you with a wealth of technologies applicable to animal agriculture.

Book excerpt: Less than 10% of beef cow-calf producers in the United States currently implement reproductive technologies such as estrous synchronization and artificial insemination (AI) as a component of their breeding management program. Synchronization protocols aid in the synchrony of estrus and ovulation to improve cattle fertility, however, many protocols often require multiple hormone injections and animal handlings. Development of estrous synchronization protocols that limit number of injections, cost, and labor inputs, without compromising fertility, may improve the adoption of fixed-time artificial insemination (FTAI) within the beef industry. The main objective of the first study was to compare the effect of a single injection of high-concentration (HICON) prostaglandin F2[alpha] (PGF) or two conventional PGF injections (2PGF) administered 8 h apart at controlled internal drug release insert (CIDR) removal, on reducing serum progesterone (P4) before FTAI and on subsequent pregnancy per artificial insemination (P/AI) in postpartum beef cows (n=404) synchronized with a 5-d CO-Synch + CIDR breeding program. Serum P4 concentration at the time of insemination is a determining factor of cattle fertility in FTAI protocols, as lower concentrations have been associated with increased P/AI. A secondary objective of the first study was to assess the relationship between P4 concentration at the time of insemination and probability of pregnancy to AI. Progesterone concentrations at AI were lesser in 2PGF than HICON, however, P/AI was not different between treatments. Probability of pregnancy decreased as P4 concentration at AI increased and the optimal P4 cutoff concentration to optimize fertility was determined at 0.43 ng/mL. Intramuscular (i.m.) route of injection is commonly used to administer products used in estrous synchronization in beef cattle; however, this route causes greater incidence of injection-site lesions and tissue damage associated with reduced beef carcass quality and tenderness than subcutaneous (s.c.) routes. Therefore, use of s.c. administration is favorable to reduce carcass damage. The objective of the second study was to compare the ability of a single s.c. injection of high-concentration PGF (HICON-SC) with two i.m. injections of conventional PGF 6 h apart (2PGF-IM) to reduce P4 concentration and induce luteolysis by the time of AI in a 5-d CO-Synch + CIDR synchronization program in postpartum beef cows. A similar decline in P4 concentration occurred in both treatments following PGF, indicating s.c. administration of HICON could be an effective alternative to i.m. administration of the conventional double PGF dose to induce luteolysis before AI in the 5-d CO-Synch + CIDR protocol.

Book excerpt: The objectives of the studies presented in Section I were to evaluate insemination dynamics, time to pregnancy, and profitability of replacement dairy heifers managed with first service programs that relied primarily on insemination at detected estrus (AIE), timed-artificial insemination (TAI), or a combination of both. The objectives of the experiments presented in Section II were; to develop and validate a tool integrating a disposable fluorescence-based lateral flow immunoassay (LFIA) coupled with a portable imaging device to determine the reproductive status of cows based on circulating progesterone (P4) concentration; to test the efficacy of intravaginal (IVG) administration of prostaglandin-F2_ (PGF) to induce corpus luteum (CL) regression; and to develop and validate an electronically-controlled hormone delivery device for reproductive control of cattle. The studies presented in Section I demonstrated that a reproductive management strategy designed to submit all heifers to TAI reduced time to first AI and time to pregnancy when compared with a strategy that relied primarily on AIE after induction of estrus with PGF treatments. The program that combined AIE and TAI resulted in intermediate performance. In spite of greater reproductive cost, programs that used TAI in combination with AIE or solely TAI reduced rearing cost and generated more revenue during first lactation, which translated in numerically greater cash flow under fixed or simulated market conditions. The first experiment of Section II demonstrated that the developed LFIA system can accurately predict the presence of a functional CL (i.e., P4 ≥ 1 ng/mL) using bovine plasma samples. Additional improvements of this assay may lead to the development of a rapid, low-cost, cow-side tool for determination of reproductive status of cattle. The other experiments presented in Section II demonstrated that two IVG treatments of PGF 12 hours apart resulted in similar luteal regression risk, estrus expression, ovulatory response to GnRH, and pregnancy per AI compared with that observed after a single intramuscular dose of PGF. Further, we demonstrated that the current IVG prototype device for hormone delivery can be programmed to automatically release PGF for successful induction of luteal regression in cattle. Once optimized, the developed IVG device may be an alternative too to the needle-injection methods presently used to synchronize ovulation of cows.
