Since we have hundreds of miRNAs, the major challenge is now to understand their specific biological function. In fact the experimental evidence suggests that signaling pathways could be ideal candidates for miRNA-mediated regulation. Several studies suggest that miRNAs affect the responsiveness of cells to signaling molecules such as WNT, Notch, TGF-β and EGFR. Altered expression of particular miRNAs has been implicated in the onset and development of cancer and could be used as potential biomarkers for the disease. Recently, many studies have found miRNAs have crucial regulatory roles in Cancer stem cells (CSCs) a kind of tumor initiating cells (TICs) and dormancy. Findings also suggest that DNA methylation may be important in regulating the expression of many miRNAs in several cancer initiating cells. Several miRNAs are known to either upregulated or downregulated in CSCs when compared to non-cancerous cells from the same tissues. CSCs are a small subpopulation of cells identified in a variety of tumors and involve in self-renewal, differentiation, chemoresistance and tumorigenesis. Book excerpt: The discovery of microRNA (miRNA) involvement in cancer a decade ago, and the more recent findings of long non-coding RNAs in human diseases, challenged the long-standing view that RNAs without protein-coding potential are simply “junk” transcription within the human genome. These findings evidently changed the dogma that “DNA makes RNA makes protein” by showing that RNAs themselves can be essential regulators of cellular function and play key roles in cancer development. MiRNAs are evolutionarily conserved short single-stranded transcripts of 19–24 nucleotides in length. They do not code for proteins, but change the final output of protein-coding genes by regulating their transcriptional and/or translation process. Ultraconserved genes (UCGs) are non-coding RNAs with longer length (>200bp) that are transcribed from the ultraconserved genomic region. Both miRNAs and UCGs are located within cancer-associated genomic regions (CAGRs) and can act as tumor suppressors or oncogenes. In this chapter, we present principles and concepts that have been identified over the last decade with respect to our understanding of the function of non-coding RNAs, and summarize recent findings on the role of miRNAs and UCGs in cancer development. Finally, we will conclude by discussing the translational potential of this knowledge into clinical settings such as cancer diagnosis, prognosis and treatment.

Since the first description of prominin in 1997, in mouse neuroepithelial cells and in human hematopoietic stem cells as AC133 antigen, this molecule has aroused a large interest especially, as a stem cell marker, that gave rise to an ever growing body of publications and more recently its expression in cancer stem cells. Controversies as to its role as a cancer stem and its detection in different models, as well as its use as a prognostic marker have emerged. Yet, beyond its use as a stem cell and cancer stem cell marker, prominin-1/CD133 displays unique biological features and appears of importance in other processes like for example in retinal biogenesis. Indeed, this five-transmembrane plasma membrane glycoprotein, which marks membrane protrusions is associated with several essential processes like cell polarity, asymmetric cell division and membrane remodeling. We propose to review current knowledge about this intriguing molecule and present pertinent information to determine the biological role of prominins and assess their importance in medicine and cancer research. The primary audience for this book is geared towards scientists and researchers with interest in cancer stem cells, stem cells, cell biology, neurobiology, and regenerative medicine.
