Book excerpt: Excerpt from Proceedings of the American Society of Biological Chemists, 1916, Vol. 4 Although it has been recognized since the time of Magendie that blood possesses active diastatic properties, no studies of a conclusive nature appear to have been published relative to the activity of the blood diastase in human subjects suffering from diabetes. This has probably been due to the lack of a sufficiently delicate and accurate method. It has been found that the procedure introduced by Lewis and Benedict' for the estimation of the sugar of the blood, may be excellently utilized in the estimation of its diastatic activity. Two 2 cc. Samples of oxalated blood are taken, one being employed as a control. The control tube is made up to 10 cc. With distilled water, and the tube to be employed for the test to 9 cc. Both tubes are now placed in a water bath at 40°c. As soon as the contents of the tubes have been brought to this temperature, 1 cc. Of 1 per cent soluble starch is added to the second tube, the con tents are mixed, and incubation is then carried out for exactly 15 minutes at 40°c. After the incubation has been completed, about gm. Of dry picric acid is at once added to each tube and the mixtures are stirred. When the proteins are precipitated, the tubes are centrifuged and the supernatant fluid is filtered. The sugar in 3 cc. Portions of the filtrates is now estimated according to the technique described by Myers and Bailey.2 Correction is made for the sugar originally present in the blood (with the aid of the control) and for the slight reducing action of the soluble starch. The results have been recorded in terms of the percentage of the soluble starch (10 mg.) transformed to reducing sugars (calculated as glucose) by the 2 cc. Of blood employed.
