Book excerpt: Excerpt from Remarks on Cellular Separation: Read by Appointment of the American Association for the Improvement of Penal and Reformatory Institutions, at the Annual Meeting in New York, November 29, 1860 At the very opening of the Eastern Penitentiary, more than thirty years ago, the same views were expressed by one of the officers who signed the communication just cited, Mr. George W. Smith, in his Defence of the Separate System, republished in 1833 by order of the Philadelphia Society. In that valuable witness of the opinions and designs which were entertained and published at the period just mentioned, the period of most excited partisan ship, the author said, Religious and other instruo tion will be constantly and regularly administered; the visits of the virtuous and benevolent permitted and encouraged, under proper restrictions; unremit ted solitude, or separation from all society, will not be, therefore, permitted. Again, It was never ih tended by the friends of our system, even by thme who were opposed to the introduction of labor, todeprive the convicts of exercise, of books of instruc tion, and of suitable society. Book excerpt: This NATO Advanced Research Workshop held 25-30 September, 1988 at the Villa Gualino, Turin, Italy, was the first international meeting of its kind to be devoted solely to cell separation in plants. The partial or complete dissociation of one cell from another is an integral process of differentiation. Partial cell separations are basic physiological components of the overall programme of plant development. Complete cell separations are major events in the ripening of fruits, and the shedding of plant parts. Unscheduled cell separations commonly occur when tissues are subjected to pathogenic invasion. Environmental stresses too, evoke their own separation responses. Over the past five years much new knowledge has been acquired on the regulation of gene expression in specific stages of cell differentiation. Specific molecular markers have been identified that designate the competence of cells for achieving separation. Certain of the chemical signals (hormones, elicitors) that must be emitted or perceived by cells to initiate and sustain separation, are now known to us, and the resulting cell wall changes have come under close chemical scrutiny. The Turin meeting was a focus for those currently involved in such investigations. It assessed factors controlling cell separation in a wide spectrum of different cell types under a variety of conditions.

Book excerpt: Cell separation is at the core of current methods in experimental biology and medicine. Its importance is illustrated by the large number of physical and biochemical principles that have been evaluated for application to cell separation. The development of cell separation methods is driven by the needs of biological and medical research, and the ever-increasing demands for sensitivity, selectivity, yield, timeliness and economy of the process. The interdisciplinary nature of research in this area and the volume of information available in research publications and conferences necessitates a basic description of the fundamental processes involved in magnetic cell separation that may help the user in navigating this wealth of information available online and in scientific publications. This book will appeal to researchers in many areas utilizing this technique, including those working in cell biology, clinical research, inorganic chemistry, biochemistry, chemical engineering, materials science, physics and electrical engineering. Provides examples of how to calculate the volume magnetic susceptibility, a fundamental quantity for calculating the magnetic force acting on a cell, from various types of magnetic susceptibilities available in literature Introduces the elements of magnetostatics as they apply to cell magnetization and the magnetization of magnetic micro- and nano- particles used for cell separation Describes the parameters used to determine cell magnetophoresis

Book excerpt: Methods of Cell Separation brings to the attention of researchers at all levels the variety of methods available for separating viable populations of cells. Methods are grouped into 3 categories based on the criteria of separation, namely; size or density; non-specific surface properties; and specific surface properties. The principle of each method is described together with general and, where possible, specific protocols for conducting cell separation experiments. A central theme of the book is the separation of populations of blood lymphocytes which is used as an example for each method. Methods of Cell Separation is distinguished by three powerful assets: descriptions of the majority of cell separation methods currently being used; details of the experimental procedures involved in each method; and comparisons of the different methods for separating cell populations with particular reference to blood lymphocytes. An excellent addition to a distinguished series, and extremely useful as a laboratory manual.

Available in PDF, EPUB and Kindle. Book excerpt: The manipulation of cells and microparticles within microfluidic systems using external forces is valuable for many microscale analytical and bioanalytical applications. Acoustofluidics is the ultrasound-based external forcing of microparticles with microfluidic systems. It has gained much interest because it allows for the simple label-free separation of microparticles based on their mechanical properties without affecting the microparticles themselves. Microscale Acoustofluidics provides an introduction to the field providing the background to the fundamental physics including chapters on governing equations in microfluidics and perturbation theory and ultrasound resonances, acoustic radiation force on small particles, continuum mechanics for ultrasonic particle manipulation, and piezoelectricity and application to the excitation of acoustic fields for ultrasonic particle manipulation. The book also provides information on the design and characterization of ultrasonic particle manipulation devices as well as applications in acoustic trapping and immunoassays. Written by leading experts in the field, the book will appeal to postgraduate students and researchers interested in microfluidics and lab-on-a-chip applications.

Available in PDF, EPUB and Kindle. Book excerpt: In Volume I, Analysis of Cells and Tissues, we presented a range of protocols aimed at mapping and analyzing the expression of various molecules of pot- tial interest in metastasis research and for examining their production at the genetic level. In this second volume of metastasis research protocols, we move to the level of living cells and tissues and present methodologies applicable to examining metastatic behavior in vitro and in whole animal models. The methods described in the first section of this volume concentrate on the separation of cell lines with high and low metastatic potential, including the genetic modification of cell lines. The assay systems to test defined aspects of the metastatic cascade are then described in Part II and include cell migration assays, assays for matrix degrading enzymes, basement membrane degrading assays, adhesion assays, and assays of angiogenesis. The role of the specific elements of the metastatic cascade assayed in each of these systems in turn must of course be put into perspective relative to their roles in entire living organisms.

Book excerpt: Cell Separation: Methods and Selected Application, Volume 3 provides information pertinent to the design and application of methods for the separation of cells. This book covers a variety of topics, including lymphoma cells, lectins, purification of cells from lung tumors, macrophage electrophoretic migration test, tissue heterogeneity, and characteristics of cultured cells. Organized into 13 chapters, this volume begins with an overview of the approaches in examining particular cell-surface properties and their role in the metastatic process. This text then examines lectins as important tools for identification and separation of cells, particularly of lymphocyte subpopulations. Other chapters consider the various methods that have been used to disperse rat pituitary tissue into single cells. This book discusses as well the different methods for isolating type II cells. The final chapter deals with the significance of having cell cultures homogeneous for a given cell type. This book is a valuable resource for cell biologists, experimental oncologists, hematologists, immunologists, and endocrinologists.
