Available in PDF, EPUB and Kindle. Book excerpt: Recent Advances in the Pharmacology of Toxins brings together the proceedings of the Second International Pharmacological Meeting of the International Union of Physiological Sciences, held in Prague, Czechoslovakia, on August 20-23, 1963. The meeting provided a forum for discussing advances in the understanding of the pharmacology of toxins and addressed topics ranging from antagonists of bradykinin to the mode of the spread of tetanus toxin in experimental animals. This volume is comprised of 28 chapters and begins with an analysis of the effects of toxins from microorganisms on mammalian cellular metabolism and structure, followed by a review of SRS-A, a slow-reacting substance of anaphylaxis, as a potential factor in the symptoms produced by venoms and toxins. Subsequent chapters explore the release of pharmacologically active lipids by enzymes contained in toxins; the structure and function of some enzymes in snake venoms, with reference to disulfide linkages; biochemistry and pharmacology of small-molecular compounds of cobra venom; and the role of ganglioside in the mode of action of tetanus toxin. This book will be of particular value to pharmacologists and physiologists.

Book excerpt: Improving and Accelerating Therapeutic Development for Nervous System Disorders is the summary of a workshop convened by the IOM Forum on Neuroscience and Nervous System Disorders to examine opportunities to accelerate early phases of drug development for nervous system drug discovery. Workshop participants discussed challenges in neuroscience research for enabling faster entry of potential treatments into first-in-human trials, explored how new and emerging tools and technologies may improve the efficiency of research, and considered mechanisms to facilitate a more effective and efficient development pipeline. There are several challenges to the current drug development pipeline for nervous system disorders. The fundamental etiology and pathophysiology of many nervous system disorders are unknown and the brain is inaccessible to study, making it difficult to develop accurate models. Patient heterogeneity is high, disease pathology can occur years to decades before becoming clinically apparent, and diagnostic and treatment biomarkers are lacking. In addition, the lack of validated targets, limitations related to the predictive validity of animal models - the extent to which the model predicts clinical efficacy - and regulatory barriers can also impede translation and drug development for nervous system disorders. Improving and Accelerating Therapeutic Development for Nervous System Disorders identifies avenues for moving directly from cellular models to human trials, minimizing the need for animal models to test efficacy, and discusses the potential benefits and risks of such an approach. This report is a timely discussion of opportunities to improve early drug development with a focus toward preclinical trials.

Book excerpt: Salt and Water Balance, Volume 8 presents the proceedings of the 3rd International Pharmacological Meeting, held in Sao Paulo, Brazil, on July 24–30, 1966. This book features the modulation of renal control of electrolyte and water balance, which is the most effective approach of handling most clinical problems wherein fluid retention needs to be reduced. Comprised of 10 chapters, this compilation of papers starts with an overview of the diuretic agents that are effective diuretic therapy. This text then describes the active transport, which proceeds at the expense of energy against the combined chemical and electrical gradients in the case of electrolytes. Other chapters consider the historical importance in therapy and the mechanisms of action of mercurial diuretics. This book discusses as well the complications and unwanted side effects of diuretic therapy. The final chapter deals with the hemodynamic mechanisms and their possible influence on the tubular function. This book is a valuable resource for physicians, health care professionals, and patients.

Book excerpt: The Control of Growth Processes by Chemical Agents covers the proceedings of the 1966 Third International Pharmacological Meeting on Control of Growth Processes by Chemical Agents, held in Sao Paolo, Brazil. This book is composed of seven chapters, and begins with a survey of the comparative biology of dihydrofolate reductases as a basis for chemotherapy. The succeeding chapters deal with the mechanism of pyrimidine metabolism as a chemotherapeutic target and the biochemical and biological research works with arabinoforanosyl cytosine, known as cytarabine. These chapters specifically describe the cytotoxicity and antitumor activity of cytarabine. The remaining chapters discuss the advances in viral chemotherapy and the regulatory mechanisms of enzyme synthesis and their alteration in malignancy. This book will be of value to biologists, biochemists, and oncologists.

Available in PDF, EPUB and Kindle. Book excerpt: Section on Pharmacology of the International Union of Physiological Sciences (SEPHAR), Proceedings of the Second International Pharmacological Meeting, August 20-23, 1963, Volume 3: Pharmacology of Cholinergic and Adrenergic Transmission focuses on the effects of drugs on muscles, nerve fibers, and the central nervous system. The selection first offers information on the role of sodium ions in the release of acetylcholine and the distribution and release of acetylcholine in muscles. Discussions focus on the effects of sodium deficiency on ACh release in perfused ganglia; effects of sodium pump inhibitors on ganglionic and myoneural transmission; distribution of ACh and choline acetylase in muscle; and ACh release after denervation. The text then ponders on the roles of acetylcholine and acetylcholinesterase in junctional transmission and correlated studies of monoamines and acetylcholinesterase in sympathetic ganglia, manifesting the distribution of adrenergic and cholinergic neurons. The publication examines the action of acetylcholine and related drugs on mammalian nonmyelinated nerve fibers; possible mechanisms of acetylcholine action in muscles; and electrophysiological analysis of cholinergic transmission in sympathetic ganglia. The text then reviews the interactions of cholinomimetic and cholinergic blocking drugs at sympathetic ganglia; evolution of cholinoreceptive sites of locomotor muscle; and pharmacological blocking of central cholinoreactive systems and the possibilities of its therapeutic application. The selection is a dependable source of data for readers interested in the pharmacology of cholinergic and adrenergic transmission.

Available in PDF, EPUB and Kindle. Book excerpt: Physico-Chemical Aspects of Drug Action, Volume 7 covers topics on drug kinetics and the overall physicochemical properties of the drug in relation therewith, and the physicochemical aspects of the drug-receptor interaction, putting emphasis on receptor mechanisms and specific properties required for certain types of drugs in this respect. The book starts with some contributions dealing with various general aspects of drug kinetics followed by some contributions dealing with the relationship between certain physicochemical properties of drug molecules and their action. The text describes the pharmacokinetics and dose-concentration relationships; the time course of the biological response to drugs; and the empirical equations for correlating biological efficiency of organic compounds. The text also describes molecular basis for the action of chemotherapeutic drugs; the structure-activity studies on sulphonamides; and the water extrusion hypothesis. The mathematical treatment of two-point attachment between drug and receptor; the molecular properties and biological activity of catecholamines and certain related compounds; and the structure-activity relationships of diarylcarbinolethers are also considered. The book further tackles quantum mechanically-derived electronic distributions in the conformers of 2-pam; and the molecular basis for the action of certain drugs in the central nervous system. Pharmacologists and chemists will find the book invaluable.
