Book excerpt: Electron Microscopy of Plant Cells serves as manual or reference of major modern techniques used to prepare plant material for transmission and scanning electron microscopy. There have been other books that generally discuss electron microscope methodology. This book focuses on problem areas encountered through the presence of tough cell walls and large central vacuole. It details preparative techniques for botanical specimens. Each of the nine chapters of this book covers the basic principles, useful applications, and reliable procedures used on the method of electron microscopy. Other topics discussed in each chapter include the general preparation and straining of thin sections, quantitative morphological analysis, and enzyme cytochemistry. This book also explains the immunogold labelling, rapid-freezing methods, and ambient- and low-temperature scanning electron microscopy among others. This book will be invaluable to general scientists, biologists, botanists, and students specializing in plant anatomy.

Book excerpt: The modem microbiologist is often a real specialist who has difficulty under standing and applying many of the techniques beyond those in his or her own immediate field. On the other hand, most benefits to modem microbiology are obtained when a broad spectrum of scientific approaches can be focused on a problem. In early studies, electron microscopy was pivotal in understanding bacterial and viral morphology, and we still feel that we will understand a disease better if we have seen an electron micrograph of the causative agent. Today, because there is an increased awareness of the need to understand the rela tionships between microbial structure and function, the electron microscope is still one of the most important tools microbiologists can use for detailed analysis of microorganisms. Often, however, the aforementioned modem microbiologist still thinks of ultrastructure as involving negative staining or ultrathin sectioning in order to get a look at the shape of a "bug. " Many of the newer ultrastructure techniques, such as gold-labeled antibody localization, freeze-fracture, X-ray microanalysis, enzyme localization, and even scanning electron microscopy, are poorly under stood by, and therefore forbidding to, the average microbiologist. Even many cell biologists admit to having difficulty staying in touch with current develop ments in the fast-moving field of electron microscopy techniques.

Book excerpt: The concept for a treatise covering selected natural polymer systems was initiated during a national meeting in cell biology in 1978. The challenge to the editor was to organize a book dealing principally but not exclusively with cellulose. A brief background may help to provide the reader with information to understand the reasons for the specific selections within this volume. better During the past decade, we have witnessed significant changes in the sciences as well as the day-to-day life styles of our citizens. It will not be forgotten that during the early seventies, a significant change was to take many Americans by surprise. The oil embargo on The United States caused unexpected shortages of fuels. The long gasoline lines impressed in the minds of Americans that our energy-rich future with non-renewable resources is limited. The modelling of ecosystems, population growth, urban development, etc., have continued to raise our awareness that life on earth, including renewable resources, is indeed fragile. Contrary to popular belief, even wood and wood products are not limitless.

Available in PDF, EPUB and Kindle. Book excerpt: Modern materials include a vast array of polymers and plastics which are found in applications such as housing, appliances, clothing and household textiles and automotive and aerospace industries. Thus research scientists, engineers and materials science graduate students need to be aware of the methods and techniques required to understand the structure-property relations of polymer materials. This book will review the field of the microscopy of polymers. There is a vast literature which describes the research results obtained by study of polymer materials using microscopy and other complementary analytical techniques and such studies are best left to journals on specific topics. The major objective of this text is to provide the basic microscopy techniques and specimen preparation methods applicable to polymers. The book will attempt to provide enough detail so that the methods described can be applied, and also to reference appropriate publications for the investigator interested in more detail. The selection of two authors for this text came from the desire for a comprehen sive review of polymer microscopy with emphasis on methods and techniques, rather than on research results. The synergism provided by two authors with widely varied backgrounds was thought to be important: one author (LCS) has an industrial focus and a background in chemistry whilst the other (DTG) has an academic environment and offers a background in polymer physics.

The recent awards of Nobel Prizes awarded for super-resolution optical microscopy and cryo-electron microscopy have demonstrated the rich scientific opportunities for research in novel microscopies. Earlier Nobel Prizes for electron microscopy (the instrument itself and applications to biology), scanning probe microscopy and holography are a reminder of the central role of microscopy in modern science, from the study of nanostructures in materials science, physics and chemistry to structural biology. Separate chapters are devoted to confocal, fluorescent and related novel optical microscopies, coherent diffractive imaging, scanning probe microscopy, transmission electron microscopy in all its modes from aberration corrected and analytical to in-situ and time-resolved, low energy electron microscopy, photoelectron microscopy, cryo-electron microscopy in biology, and also ion microscopy. In addition to serving as an essential reference for researchers and teachers in the fields such as materials science, condensed matter physics, solid-state chemistry, structural biology and the molecular sciences generally, the Springer Handbook of Microscopy is a unified, coherent and pedagogically attractive text for advanced students who need an authoritative yet accessible guide to the science and practice of microscopy.

Book excerpt: Although the art of making cheese can be traced to prehistoric times, it has continued to evolve as modern civilization progressed. The advent of new technologies and instrumentation has brought exponential growth in the understanding of cheese components and their function. Even more recently, the evolution of cheesemaking has accelerated, driven by economic factors such as the establishment of the European Economic Community, the changing diet of developed countries, and the environmental and economic concerns associated with whey disposal. Molecular biology has revolutionized the development of starter and adjunct cultures as well as rennets, and genetics will make it possible to maintain ideal milk components for cheesemaking. The ability to accelerate traditional ripening procedures has altered the production of certain cheeses, and the emphasis on decreasing the intake of dietary fat, especially in the United States, has prompted the development of technology for producing low-fat cheeses with traditional texture and flavor. In assembling a distinguished group of participants for the symposium, "Chemistry of the Structure/Function Relationships in Cheese," we hoped to review the interplay of these trends and forecast the direction of future research. Contributors evaluated the current status of cheesemaking and highlighted the information that will be essential for new developments. They also focused the attention of agricultural and food chemists on the opportunities in cheese research and the potential contributions they might make to the future of cheese, a most valuable food product. Book excerpt: The oil crisis during the 1970s turned interest towards the utilization of renewable resources and towards lignocellulosics in particular. The 1970s were also the cradle period of biotechnology, and the years when biotechnical utilization of lignocellulosic waste from agriculture and forestry gained priori ty. This was a logical conclusion since one of nature's most important biologi cal reactions is the conversion of wood and other lignocellulosic materials to carbon dioxide, water and humic substances. However, while biotechnology in other areas like medicine and pharmacology concerned production of expen sive products on a small scale, biotechnical utilization and conversion of ligno cellulosics meant production of inexpensive products on a large scale. Biotechnical utilization of lignocellulosic materials is therefore a very difficult task, and the commercial utilization of this technology has not progressed as rapidly as one would have desired. One reason for this was the lack of basic knowledge of enzyme mechanisms involved in the degradation and conversion of wood, other lignocellulosics and their individual components. There are also risks associated with initiating a technical development before a stable platform of knowledge is available. Several of the projects started with en thusiasm have therefore suffered some loss of interest. Also contributing to this failing interest is the fact that the oil crisis at the time was not a real one. At present, nobody predicts a rapid exhaustion of the oil resources and fuel production from lignocellulosics is no longer a high priority.
