Epidemiological studies have highlighted the emergence of distinct potato PVY variants able to induce necroses on potato tubers. Due to the lack of efficient resistance to PVY isolates inducing necrotic symptoms in cultivated varieties and the plant-to-plant transmission of isolates through the daughter tubers, PVY has become the most economically important virus for the potato industry. The review offers an overview of several decades of research on PVY but also focuses on the latest data obtained by expert on PVY worldwide on the biological characteristics of PVY, interactions between aphids-hosts, its evolution and management. Identified knowledge gaps to understand further PVY biology will be discussed.

Plant roots, bacteria, archaea, fungi, nematodes, and other macroscopic organisms interact here with each other. Plants represent the main influencing force as they produce a mixture of chemical molecules and extrude them in the form of root exudates. Those exudates determine not only the nutrient availability of the plant but also influence the outcome of the interactions in the vicinity of the roots. In response to the plant-derived signals, a subset of the bulk soil microbes can proliferate in the proximity of the root and some of them can eventually overcome the plant defense system to colonize the plants. The root-associated microbiota is assumed to be crucial for plant health, and belowground plant-microbe and microbe-microbe interactions can result in altered growth and nutritional quality of the aboveground plant parts. Such interactions affect community composition and productivity in natural ecosystems or crop yield in agricultural fields. Root associations with plant growth-promoting rhizobacteria (PGPR) producing phytohormones that alter root architecture, or produce antibiotics to suppress antagonistic pathogens, affecting nutrient availability and competition between plants are just a few among many examples of such belowground interactions in the rhizosphere. Moreover, the role of rhizosphere microorganisms in stimulating the plant immune system leading to Induced Systemic Resistance (ISR) has also been a focus of an active investigation. However, the molecular mechanisms are still largely unknown. On one hand, it has been proposed that plants actively recruit beneficial microorganisms under certain pathological conditions or influence the soil microbial communities and create a so-called “soil memory”, which is conveyed to future plant generations. This is crucial to evolve appropriate control practices and to prevent the virus infection. Researchers have achieved considerable progress in characterization, detection and management of virus on different crop species in the last decade. The etiologic agents for AHPND are virulent strains of bacteria belonging to the genus Vibrio parahaemolyticus and related species, which harbor specific toxin genes. While these bacterial species are part of the normal microflora of the marine environment, they may cause substantial mortalities in whiteleg shrimp (Penaeus vannamei) and giant tiger prawn (Penaeus monodon) cultured in countries in Asia and the Americas. These strains of these Vibrio bacteria secrete a PirABvp binary toxin resulting in sloughing of tubule epithelial cells and dysfunctions of the hepatopancreas in the acute form; mortality can reach 100 percent in affected ponds. Chronic presentation of this disease involves secondary bacterial infection of hepatopancreas and running mortality over the culture cycle. Acute or chronic presentation would greatly depend on the culture conditions. This disease can be considered a toxicosis rather than an infection. Economic losses due to this disease have amounted to over USD 7 billion annually. Further outbreaks of AHPND, particularly in areas that are currently free of the disease, would be expected to experience similar devastating effects on local shrimp producers and the surrounding communities; and thus, there is an urgent need to develop a contingency plan to control and eradicate this disease. This manual includes information on: 1) the nature of AHPND: a brief review of current knowledge in disease etiology, susceptible species and global distribution; 2) diagnosis of disease: a description of gross clinical signs and laboratory methods; 3) prevention and treatment: farm management, the use and development of antibiotics, bacteriophages, probiotics, disease-tolerant shrimp, shrimp immunity and vaccination; 4) epidemiology: AHPND’s geographic distribution, genotype, persistence in the environment, reservoir hosts, modes of transmission, risk factors, and economic impacts; 5) principles of control and eradication: methods for containment, mitigation and eradication of AHPND, and trade and industry considerations; and 6) policy development and implementation: AHPND-specific objectives, options and strategies for eradication and control, education, capacity building, funding, and compensation.
