Book excerpt: Biological Techniques is a series of volumes aimed at introducing to a wide audience the latest advances in methodology. The pitfalls and problems of new techniques are given due consideration, as are those small but vital details not always explicit in the methods sections of journal papers. In recent years, most biological laboratories have been invaded by computers and a wealth of new DNA technology and this will be reflected in many of the titles appearing in the series. The books will be of value to advanced researchers and graduate students seeking to learn and apply new techniques, and will be useful to teachers of advanced undergraduate courses involving practical or project work. Methods of constructing artificial membranes (planar lipid bilayers) from the main components of cell membranes (lipids) date from the early 1960s.Planar bilayers offer direct, quantitative experimental approaches to the study of membranes of precisely determined composition which can be manipulated by the experimenter. Pore-forming molecules, transporter molecules, ATP-dependent enzymes and other entities can be incorporated into the bilayers to simulate biological functions.Reconstitution of such functions in this way remains a key final step in attributing a functional role to purified cell membrane proteins. This book aims to demystify these techniques and begins with a broad overview of the development of the subject before dealing with the protocols involved.Key references are provided at the end of the book together with a list of suppliers. Full practical details include: How to set up conventional painted and folded bilayer experiments Patch-dipping methods and use of giant liposomes Lipid characterization, preparation and purification Discussion of construction of essential apparatus, flux measurements, electrical recording, data acquisition and computer support Biochemical methods for use in planar bilayer experiments Techniques for incorporation of native proteins and other molecules

Book excerpt: The Elsevier book-series "Advances in Planar Lipid Bilayers and Liposomes' (APLBL) provides a global platform for a broad community of experimental and theoretical researchers studying cell membranes, lipid model membranes and lipid self-assemblies from the micro- to the nanoscale. Planar lipid bilayers are widely studied due to their ubiquity in nature and find their application in the formulation of biomimetic model membranes and in the design of artificial dispersion of liposomes. Moreover, lipids self-assemble into a wide range of other structures including micelles and the liquid crystalline hexagonal and cubic phases. Consensus has been reached that curved membrane phases do play an important role in nature as well, especially in dynamic processes such as vesicles fusion and cell communication. Self-assembled lipid structures have enormous potential as dynamic materials ranging from artificial lipid membranes to cell membranes, from biosensing to controlled drug delivery, from pharmaceutical formulations to novel food products to mention a few. An assortment of chapters in APLBL represents both an original research as well as comprehensives reviews written by world leading experts and young researchers.

This book was released on 2012-01-02 with total page 303 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book addresses the possibilities and challenges in mimicking biological membranes and creating membrane-based sensor and separation devices. Recent advances in developing biomimetic membranes for technological applications will be presented with focus on the use of integral membrane protein mediated transport for sensing and separation. It describes the fundamentals of biosensing as well as separation and shows how the two processes are working in a cooperative manner in biological systems. Biomimetics is a truly cross-disciplinary approach and this is exemplified using the process of forward osmosis will be presented as an illustration of how advances in membrane technology may be directly stimulated by an increased understanding of biological membrane transport. In the development of a biomimetic sensor/separation technology, both channels (ion and water channels) and carriers (transporters) are important. An ideal sensor/separation device requires the supporting biomimetic matrix to be virtually impermeable to anything but the solute in question. In practice, however, a biomimetic support matrix will generally have finite permeabilities to water, electrolytes, and non-electrolytes. These non-protein mediated membrane transport contributions will be presented and the implications for biomimetic device construction will be discussed. New developments in our understanding of the reciprocal coupling between the material properties of the biomimetic matrix and the embedded proteins will be presented and strategies for inducing biomimetic matrix stability will be discussed. Once reconstituted in its final host biomimetic matrix the protein stability also needs to be maintained and controlled. Beta-barrel proteins exemplified by the E. Coli outer membrane channels or small peptides are inherently more stable than alpha-helical bundle proteins which may require additional stabilizing modifications. The challenges associated with insertion and stabilization of alpha-helical bundle proteins including many carriers and ligand and voltage gated ion (and water) channels will be discussed and exemplified using the aquaporin protein. Many biomimetic membrane applications require that the final device can be used in the macroscopic realm. Thus a biomimetic separation device must have the ability to process hundred of liters of permeate in hours – effectively demanding square-meter size membranes. Scalability is a general issue for all nano-inspired technology developments and will be addressed here in the context biomimetic membrane array fabrication. Finally a robust working biomimetic device based on membrane transport must be encapsulated and protected yet allowing massive transport though the encapsulation material. This challenge will be discussed using microfluidic design strategies as examples of how to use microfluidic systems to create and encapsulate biomimetic membranes. The book provides an overview of what is known in the field, where additional research is needed, and where the field is heading.

Book excerpt: This book summarizes the current status of research on bilayer lipid membranes (planar lipid bilayers and spherical liposomes). In addition to describing the properties of lipid bilayers and examining biomembrane phenomena, the book has two other objectives. The first is to present practical methods for the formation and study of lipid bilayers with either aqueous or metal-lipid bilayer interfaces. The second aim is to treat planar lipid bilayers as a new type of interfacial adsorption phenomena. The first nine chapters cover properties of biomembranes, basic principles of membrane biophysics, transport, electrochemistry, physiology, bioenergetics, and photobiology. Chapter 10 presents the following topics: lipid bilayers in medicine, supported lipid bilayers as sensors, a short discussion of liposomes, and solar energy transduction via semiconductor septum photovoltaic cells based on natural photosynthesis.
