Available in PDF, EPUB and Kindle. Book excerpt: It has been thirty years since one of the authors (EJD) began a collaboration with Professor Milton Kerker at Clarkson University in Potsdam, New York using light scattering methods to study aerosol processes. The development of a relatively short-lived commercial particle levitator based on a modification of the Millikan oil drop experiment attracted their attention and led the author to the study of single droplets and solid microparticles by levitation methods. The early work on measurements of droplet evaporation rates using light scattering techniques to determine the size slowly expanded and diversified as better instrumentation was developed, and faster computers made it possible to perform Mie theory light scattering calculations with ease. Several milestones can be identified in the progress of single microparticle studies. The first is the introduction of the electrodynamic balance, which provided more robust trapping of a particle. The electrodynamic levitator, which has played an important role in atomic and molecular ion spectroscopy, leading to the Nobel Prize in Physics in 1989 shared by Wolfgang Paul of Bonn University and Hans Dehmelt of the University of Washington, was easily adapted to trap microparticles. Simultaneously, improvements in detectors for acquiring and storing light scattering data and theoretical and experimental studies of the interesting optical properties of microspheres, especially the work on morphology dependent resonances by Arthur Ashkin at the Bell Laboratories, Richard Chang, from Yale University, and Tony Campillo from the Naval Research Laboratories in Washington D. C.

Available in PDF, EPUB and Kindle. Book excerpt: This book, written by expert scientists in the field, analyses how these diverse fields of research interact on a specific example - RNA polymerase. The book concentrates on RNA polymerases because they play a central role among all the other machines operating in the cell and are the target of a wide range of regulatory mechanisms. They have also been the subject of spectacular advances in their structural understanding in recent years, as testified by the attribution of the Nobel prize in chemistry in 2006 to Roger Kornberg. The book focuses on two aspects of the transcription cycle that have been more intensively studied thanks to this increased scientific cooperation - the recognition of the promoter by the enzyme, and the achievement of consecutive translocation steps during elongation of the RNA product. Each of these two topics is introduced by an overview, and is then presented by worldwide experts in the field, taking the viewpoint of their specialty. The overview chapters focus on the mechanism-structure interface and the structure-machine interface while the individual chapters within each section concentrate more specifically on particular processes-kinetic analysis, single-molecule spectroscopy, and termination of transcription, amongst others. Specific attention has been paid to the newcomers in the field, with careful descriptions of new emerging techniques and the constitution of an atlas of three-dimensional pictures of the enzymes involved.

Available in PDF, EPUB and Kindle. Book excerpt: This book summarizes the various microfluidic-based approaches for single-cell capture, isolation, manipulation, culture and observation, lysis, and analysis. Single-cell analysis reveals the heterogeneities in morphology, functions, composition, and genetic performance of seemingly identical cells, and advances in single-cell analysis can overcome the difficulties arising due to cell heterogeneity in the diagnostics for a targeted model of disease. This book provides a detailed review of the state-of-the-art techniques presenting the pros and cons of each of these methods. It also offers lessons learned and tips from front-line investigators to help researchers overcome bottlenecks in their own studies. Highlighting a number of techniques, such as microfluidic droplet techniques, combined microfluidics-mass-spectrometry systems, and nanochannel sampling, it describes in detail a new microfluidic chip-based live single-cell extractor (LSCE) developed in the editor’s laboratory, which opens up new avenues to use open microfluidics in single-cell extraction, single-cell mass spectrometric analysis, single-cell adhesion analysis and subcellular operations. Serving as both an elementary introduction and advanced guidebook, this book interests and inspires scholars and students who are currently studying or wish to study microfluidics-based cell analysis methods.

Book excerpt: This book offers an overview of state-of-the-art in non amplified DNA detection methods and provides chemists, biochemists, biotechnologists and material scientists with an introduction to these methods. In fact all these fields have dedicated resources to the problem of nucleic acid detection, each contributing with their own specific methods and concepts. This book will explain the basic principles of the different non amplified DNA detection methods available, highlighting their respective advantages and limitations. Non-amplified DNA detection can be achieved by adopting different techniques. Such techniques have allowed the commercialization of innovative platforms for DNA detection that are expected to break into the DNA diagnostics market. The enhanced sensitivity required for the detection of non amplified genomic DNA has prompted new strategies that can achieve ultrasensitivity by combining specific materials with specific detection tools. Advanced materials play multiple roles in ultrasensitive detection. Optical and electrochemical detection tools are among the most widely investigated to analyze non amplified nucleic acids. Biosensors based on piezoelectric crystal have been also used to detect unamplified genomic DNA. The main scientific topics related to DNA diagnostics are discussed by an outstanding set of authors with proven experience in this field.
