Available in PDF, EPUB and Kindle. Book excerpt: The Peptides: Analysis, Synthesis, Biology, Volume 6: Opioid Peptides: Biology, Chemistry, and Genetics presents a biological topic of peptide research. This book is divided into nine chapters. Chapter 1 reviews the opioid peptide precursors and their genes. The proenkephalin and products of its processing are discussed in Chapter 2. In Chapter 3, the role of pro-opiomelanocortin (POMC) as a protein at the interface of the endocrine and nervous systems is examined. Chapter 4 provides a comprehensive account of the biology and chemistry of the dynorphin peptides. The opioid receptors are described in Chapter 5. Chapter 6 evaluates the structure-activity relationships of ?-endorphin, while Chapter 7 considers the conformational analysis of enkephalins and conformation-activity relationships. The structure-activity relationships among enkephalin peptides are elaborated in Chapter 8. The last chapter is devoted to the clinical significance of opioid peptides in humans. This publication is a good reference for biologists, specialists, and researchers concerned with peptides and proteins.

Book excerpt: This two-volume book provides the first comprehensive survey of opioid research, a field which has accumulated a tremendous amount of literature since the identification of opioid receptors and their endogenous ligands. In more than 60 chapters experts present state-of-the-art reviews of this fascinating field, the topics ranging from molecular biology to clinical applications. Part I covers the multiplicity of opioid receptors, the chemistry of opiates and opioid peptides as well as the neurophysiology of opioids. Part II reviews a broad spectrum of physiological and behavioral functions and pharmacological actions of opioids, together with their neuroendocrinology, opioid tolerance and dependence, concluding with pathophysiological aspects and clinical use.

Book excerpt: Opioid Receptors: Methods and Protocols serves as a comprehensive guide to both key new techniques and established methods for the investigation of genetics, structural biology, transcription, and post-transcriptional events of opioid receptors. Other methods cover the cellular detection and trafficking of opioid receptors in vitro and in vivo. Procedures aimed to investigate signaling pathways modulated by opioid receptors and model systems to study opioid receptor-mediated functions are also included. Finally, methods to assay behavioral effects mediated by opioid receptors are described. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective introductions, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Wide-ranging and authoritative, Opioid Receptors: Methods and Protocols will help both experienced and new entrants in this field to carry out their experiments successfully and with new inspiration.

Book excerpt: Drug overdose, driven largely by overdose related to the use of opioids, is now the leading cause of unintentional injury death in the United States. The ongoing opioid crisis lies at the intersection of two public health challenges: reducing the burden of suffering from pain and containing the rising toll of the harms that can arise from the use of opioid medications. Chronic pain and opioid use disorder both represent complex human conditions affecting millions of Americans and causing untold disability and loss of function. In the context of the growing opioid problem, the U.S. Food and Drug Administration (FDA) launched an Opioids Action Plan in early 2016. As part of this plan, the FDA asked the National Academies of Sciences, Engineering, and Medicine to convene a committee to update the state of the science on pain research, care, and education and to identify actions the FDA and others can take to respond to the opioid epidemic, with a particular focus on informing FDA's development of a formal method for incorporating individual and societal considerations into its risk-benefit framework for opioid approval and monitoring.
