Available in PDF, EPUB and Kindle. Book excerpt: Fragile X Syndrome (FXS) is the most common form of inherited mental retardation, affecting approximately 1/4000 males and 1/6000 females worldwide. FXS is caused by loss of FMR1 gene expression, resulting in the lack of the protein product, Fragile X protein (FMRP). FMRP is an RNA-binding protein thought to regulate synaptic plasticity by controlling the localization and translation of specific mRNAs in neurons. To determine whether FMRP is also required in early brain development we examined the distribution of cell cycle markers in Drosophila FMR1 (dFmr1) mutant brains compared to wild-type brains. Our results indicate that the loss of dFmr1 leads to a significant increase in the number of mitotic neuroblasts and BrdU incorporation in the brain, consistent with the notion that FMRP controls proliferation in neural stem cells. To determine the role of FMRP in neuroblast division and differentiation, we used Mosaic Analysis with a Repressible Marker (MARCM) approaches in the developing larval brain and found that single dFmr1 neuroblasts generate significantly more neurons than controls. Developmental studies suggest that FMRP also inhibits neuroblast exit from quiescence, or reactivation, in early larval brains, as indicated by misexpression of the G1 to S phase transition marker Cyclin E. We have also identified a novel role for FMRP in the glia surrounding the neuroblasts, indicating that FMRP in these cells contributes to the regulation of neuroblast reactivation via signaling from the supporting glial cells. Our results demonstrate that FMRP is required during brain development to control the exit from quiescence and proliferative capacity of neuroblasts as well as neuron production, which may provide insights into Fragile X Syndrome and other Autism-Spectrum disorders.

Book excerpt: Fragile X related protein 1 (FXR1P) is part of the fragile X family of RNA binding proteins (FXRs), which includes the fragile X mental retardation protein (FMRP) and fragile X related protein 2 (FXR2P). Both FMRP and FXR2P regulate adult neural stem cells (aNSCs) and neurogenesis, a process linked to neurological disorders, such as Fragile X syndrome. Although FXR1P has been implicated in diverse developmental processes and neurological diseases, its role in neurodevelopment is not well-understood. The goal of this study was to elucidate the function of FXR1P in aNSCs and hippocampal neurogenesis. We used a Tamoxifen-inducible triple transgenic mouse model to investigate how Fxr1 deletion affects aNSC proliferation and fate specification. Deletion of Fxr1 resulted in significantly decreased populations of radial glia-like cells, neuroblasts, immature neurons, neurons, and astrocytes. We hypothesized that this reduction in new cell numbers resulted from impaired proliferation, which was confirmed both in vivo and in vitro. We discovered that FXR1P-deficient aNSCs have altered expression of a select number of cell-cycle genes, and identified the mRNA of cyclin-dependent kinase inhibitor 1A (Cdkn1a, p21) as a direct target of FXR1P. Restoration of p21 mRNA to wildtype levels rescued the proliferation deficit in aNSCs lacking FXR1P, demonstrating that p21 is a mediator of FXR1P in aNSCs. Since Fxr1 deletion in aNSCs results in a reduction of new neurons, we also performed behavioral tests for locomotion, anxiety, and hippocampal learning and memory. Although these results are preliminary, it is likely that the loss of FXR1P in aNSCs results in impaired hippocampal-dependent learning and memory.

Book excerpt: Fragile X syndrome (FXS), the most prevalent inherited intellectual disability, is caused by a loss of the fragile X mental retardation protein (FMRP). FXS symptoms manifest early in life. FMRP plays a critical function in adult hippocampal neurogenesis. The development of the DG mainly occurs postnatally. However, the role of FMRP in hippocampal neurogenesis during postnatal development remains unknown. In this study, we used Fmr1-cTag::Nestin-Cre double transgenic mice to map FMRP expression patterns in neural stem progenitor cells (NSPCs) and neurons in the dentate gyrus (DG) during the juvenile and adolescent period. We found that the majority of Sox2 expressing NSPCs in the subgranular zone (SGZ) of the DG does not express GFP-tagged FMRP. On the other hand, post-mitotic NeuN expressing neurons in the DG showed a robust expression of GFP-tagged FMRP. Additionally, we used Fmr1 knockout (KO) mouse to investigate the function of FMRP in the proliferation and neuronal differentiation of hippocampal neural stem and progenitor cells (NSPCs) at different stages of postnatal development in mouse. We found that constitutive FMRP deficiency results in smaller hippocampal NSPC population size in early adolescence. This reduction may not be due to decreased proliferation of NSPCs in Fmr1 KO animals as cell division remained unchanged assessed using both Fmr1 KO and conditional KO (cKO). We showed that constitutive lack of FMRP has no effect on proliferation and neuronal differentiation of hippocampal NSPCs in the early postnatal week, early adolescence, and late adolescence. Therefore, our data FMRP regulates the size NSPCs pool in early adolesce. We have previously shown that impaired adult hippocampal neurogenesis and cognitive deficits in a FXS mouse model are rescued by Nutlin-3 treatment analyzed shortly after treatment. The second part of this study investigated whether the beneficial effect of Nutlin-3 is sustained after a transient administration. We treated 2-month-old young adult FMRP-deficient mice with Nutlin-3 for ten days and then assessed the persistent effect of Nutlin-3 on both cognitive functions and adult neurogenesis when mice were 6-month-old mature adults. We found that transient treatment with Nutlin-3 of 2-month-old young adult FMRP-deficient mice prevents the emergence of neurogenic and cognitive deficits in mature adult FXS mice at 6 months of age. In addition, we investigated the mechanisms underlying the persistent effects of Nutlin-3 by analyzing the proliferation and differentiation of NSPCs isolated from these mice and assessing the transcriptome of the hippocampal tissues of treated mice. We found no evidence of the long-lasting restoration of proliferation and neurogenesis in the isolated adult NSPCs. However, transcriptomic analysis of the hippocampal tissue revealed that transient Nutlin-3 treatment leads to significant expression changes in genes related to the extracellular matrix, secreted factors, and cell membrane proteins in the FMRP-deficient hippocampus. In summary, we report that brief Nutlin-3 treatment may prevent impairment of adult hippocampal neurogenesis-dependent cognitive functions induced by a lack of FMRP expression in FXS.
