Book excerpt: Cytokines are regulatory polypeptides synthesized by a variety of cells in response to injury, inflammation or infection and which act on different tissues by changing gene expression and cellular metabolism. This book provides the results from recent in vitro studies of the structure and function of various cytokines. It offers an up-to-date overview of the latest advances in cytokine research, including research developments dealing with colony stimulating factors, interferons and growth and differentiation factors - as well as classical hormones such as insulin, neurokinin, somatomedin and parathormones. Several newly identified cytokine receptor structures are also discussed, as are certain activities of the recently discovered 8,000-10,000 m.w. cytokine family.

Book excerpt: The immune system is a complex network in which different cell types and soluble factors interact to efficiently eliminate various kinds of microorganisms as well as aberrant cell clones. The roots of immunologic investigations reach far into the past. In 430 BC, Thucydides reported that survivors of the plague did not present a second time with similar symptoms. The first report of a successful immu- therapy was made by Edward Jenner in 1798 who found a protective effect of cowpox vaccination against human pox. Since then, much knowledge has been accumulated; today, investigations of the molecular mechanisms of immune regulation are of central research interest. The novel insights into gene polymorphisms and gene regulation gathered from this work has improved our knowledge of individual immune reactions and risk factors in overcoming infections. Strategies to use the immune system for cancer treatment have been propelled by the discovery of divergent immunoregulatory cytokines and the introduction of new gene therapy strategies to modify immune responses. Recently, the discovery of various dendritic cells has focused attention on these cell types as central elements of the immune response and to the possibility of dendritic cell expansion, maturation, and consecutive stimulation with immuno- active tumor-specific peptides. Similarly, methods for ex vivo expansion of various stem cell-derived cell types have led to an improved therapeutic management of various benign and malignant diseases.

Book excerpt: Our most complete understanding of hormone receptor signaling may be exemplified in a family of hormones and receptors known as the cytokine superfamily. This volume is devoted to describing members of this hormone/receptor family from their molecular structures to their physiological effects. These hormones are not only biologically fascinating, but are also tremendously useful medicines. The first three chapters review the basic events in binding and signaling. The remaining papers present the cellular biology and physiological processes controlled by these hormones and receptors.

Book excerpt: Based on the assumption that invertebrates as well as vertebrates possess factors regulating hematopoiesis, response to infection or wounding, studies dealing with the evolution of immunity have focused on the isolation and characterization of putative cytokine-related molecules from invertebrates. Until recently, most of our knowledge of cytokine- and cytokine receptor-like molecules in invertebrates has relied on functional assays and similarities at the physicochemical level. As such, a phylogenetic relationship between invertebrate cytokine-like molecules and invertebrate counterparts could not be convincingly demonstrated. In the present book, recent studies demonstrating cytokine-like activities and related signaling pathways in invertebrates are critically reviewed, focusing on findings from molecular biology and taking advantage of the completion of the genome from the fly Drosophila and the worm Caenorhabditis elegans.

It is an up-to-date collection of articles that addresses the specific issue of how the cytokines are able to condition tissue specific homeostasis. The book helps the reader understand how cytokines network inside the tissues and highlights whether tissue-protection or exacerbation will be finally controlled. It describes the cytokines detected and regulated in different tissues, such as the brain, lungs, spleen, liver, pancreas and intestine, also addressing the issue of timing in specific cell types. Categorizes the cytokines based primarily on tissue and target cells Emphasizes different roles and outcomes observed during innate and adaptive response Represents a rapid guide to cytokines in health and disease in tissue and organ context Presents a different view on how known mediators may work if analyzed in a different perspective, determining the final outcome on tissue-specific target cells

Available in PDF, EPUB and Kindle. Book excerpt: Cytokines are soluble mediators of intercellular communication. They contribute to a chemical signalling language that regulates development, tissue repair, haemopoiesis, inflammation and the immune response. Potent cytokine polypepides have pleiotropic activities and functional redundancy. They act in a complex network where one cytokine can influence the production of, and response to, many other cytokines. In the past five years, this bewildering array of more than 100 effector molecules and associated cell surface receptors has been simplified by study of cytokine and cytokinereceptor structure; elucidation of convergent intracellular signalling pathways; and molecular genetics, and targeted gene disruption to 'knock-out' production of individual cytokines in mice. It is also now clear that the pathophysiology of infectious, autoimmune and malignant disease can bepartially explained by the induction of cytokines and the subsequent cellular response. Viral homologues exist for many cytokines and receptors and genetic variations in cytokine production may influence response to pathogenic stimuli. Cytokine and cytokine antagonists have shown therapeuticpotential in a number of chronic and acute diseases. The Cytokine Network: Frontiers in Molecular Biology is not a survey of individual cytokines, but guides the reader through the latest research on the cytokine network as a whole covering genomics, signalling pathways, control of the immuneresponse, and therapeutics.

Book excerpt: In the last few years molecular biologists have discovered a great deal of information about cytokine structures and genes, along with a clarification of the roles of individual cytokines. The possibility that cytokines are involved in the immunopathology of a number of diseases and could therefore be used in new therapeutic techniques has provided the incentive for extensive investigation of these molecules. This book provides an introduction to the cell and molecular biology of cytokines. The text is designed for students, research workers and clinicians who are entering this field for the first time or for those wishing to update their knowledge.

This book was released on 1999-05-13 with total page 722 pages. Available in PDF, EPUB and Kindle. Book excerpt: Every aspect of immune function and host defense is dependent upon a proper supply and balance of nutrients. Severe malnutrition can cause significant alteration in immune response, but even subclinical deficits may be associated with an impaired immune response, and an increased risk of infection. Infectious diseases have accounted for more off-duty days during major wars than combat wounds or nonbattle injuries. Combined stressors may reduce the normal ability of soldiers to resist pathogens, increase their susceptibility to biological warfare agents, and reduce the effectiveness of vaccines intended to protect them. There is also a concern with the inappropriate use of dietary supplements. This book, one of a series, examines the impact of various types of stressors and the role of specific dietary nutrients in maintaining immune function of military personnel in the field. It reviews the impact of compromised nutrition status on immune function; the interaction of health, exercise, and stress (both physical and psychological) in immune function; and the role of nutritional supplements and newer biotechnology methods reported to enhance immune function. The first part of the book contains the committee's workshop summary and evaluation of ongoing research by Army scientists on immune status in special forces troops, responses to the Army's questions, conclusions, and recommendations. The rest of the book contains papers contributed by workshop speakers, grouped under such broad topics as an introduction to what is known about immune function, the assessment of immune function, the effect of nutrition, and the relation between the many and varied stresses encountered by military personnel and their effect on health.
