Book excerpt: Nanomaterials have been a popular research field for decades and scientists are always trying to develop novel materials. This can be achieved with a hybrid of existing material to either significantly improve the performance for certain applications or generate new properties which can be exploited for new applications. Two perfect examples are gold-silver (Au/Ag) alloy nanoparticles (NPs) with strong and tunable extinction spectra in the visible range and superluminescent NPs with a metallic core and dye-doped silica shell. Both of them can be used as contrast agents for biomedical imaging applications. This research is based on the combination of these two types of NPs: the demonstration of a multiplexed bioimaging application with fluorescent NPs composed of a Au/Ag alloy core and a fluorophore-doped silica shell(Au/Ag SiO2). It has long been known that immunofluorescence, as the standard technique to label cells, has some drawbacks such as intensity limit, limited multiplexing and loss of signal due to photobleaching. Plasmonic NPs do not photobleach and do not lose their scattering signal. The Au/Ag alloy NPs with a core-shell structure not only exploit the merits of a tunable scattering signal from the core but also the metal enhanced fluorescence (MEF) effect from the shell near a metallic surface. In this research, we have fabricated the hybrid NPs and applied them in a multimodal platform to detect both scattering and fluorescence signals in order to improve the multiplexing capability. During the master study, Au/Ag SiO2 NPs were successfully fabricated with high reproducibility. Different fluorophores with different emission wavelengths were incorporated in the silica shell of Au/Ag SiO2 NPs resulting in a stronger fluorescence signal in the presence of a metallic core. An etching effect of the metallic core was noted during the silica shell growth, creating a spacer between the surface of the metallic core and silica shell resulting in a slight blueshift of the plasmonic peak. This effect is more severe for silver-rich cores. Four types of NPs were prepared, with two different Au/Ag composition ratios (pure Ag and 50/50 Au/Ag) and two different fluorophores (fluorescein and rhodamine B) were incorporated into the silica shell for a bimodal multiplexed imaging test in scattering and fluorescence. A proportion of 86% of NPs on a glass slide can be identified and a similar result was obtained for NPs on cells.

Book excerpt: A new method is developed referred to as Gold Nanorod Optical Modeling Equations (GNOME) for determining the average aspect ratio of gold nanorods in solution. In this method, the observed inhomogeneously broadened optical spectrum is fitted to a number of calculated homogeneously broadened spectra with different aspect ratios having different contributions. From this method, the average aspect ratio is determined. This is a more accurate than the presently used method of TEM. The surface plasmon enhanced fluorescence spectra of gold nanorods are calculated as a function of the aspect ratio and compared to experimental spectra. In this calculation, the inclusion of both the aspect ratio distribution calculated from the GNOME method as well as the incorporation of the intrinsic fluorescence of bulk gold are found necessary to model the enhanced fluorescence spectrum of gold nanorods using previously published equations. The enhanced spectrum decreases rapidly as the aspect ratio increases and the surface plasmon band shift away from the gold interband absorption. Photochemical methods are used to synthesize silver nanoparticles on silica surfaces and gold nanoparticles in solution. The formation silver nanoparticles utilizes benzophenone as a photosensitizing agent to initiate the reaction. The effects of the light source and irradiation time are investigated. The presence of different forms of silica are investigated in the formation of metal nanoparticles. This method produced silver nanoparticles on silica that can be in the form of film or powder that are useful in heterogeneous catalysis. Direct photochemical methods are applied to generate gold nanoparticles from chloroauoroic acid in ethylene glycol in the presence of polyvinylpyrrolidone as a capping material. A detailed mechanism of the formation of the gold nanoparticle is determined. This is done by following the kinetics of formation of the gold nanoparticles after irradiation under different conditions. The disproportionation of the gold ions as well as their reduction by ethylene glycol is found to be important in the formation of the nanoparticles. Photochemical synthesis provides room temperature techniques to generate metal nanoparticles in a variety of environments.

However, their unfavorable interaction with living systems and lack of stability in aqueous solvents has so far prevented their adoption in biology and medicine. In this paper, a simple synthetic pathway integrates gold quantum dots within a mesoporous silica shell, alongside larger gold nanoparticles within the shell's central cavity. This "quantum rattle" structure is stable in aqueous solutions, does not elicit cell toxicity, preserves the attractive near-infrared photonics and paramagnetism of gold quantum dots, and enhances the drug-carrier performance of the silica shell. In vivo, the quantum rattles reduced tumor burden in a single course of photothermal therapy while coupling three complementary imaging modalities: near-infrared fluorescence, photoacoustic, and magnetic resonance imaging. The incorporation of gold within the quantum rattles significantly enhanced the drug-carrier performance of the silica shell. Finally, this innovative material design based on the mutually beneficial interaction of gold and silica introduces the use of gold quantum dots for imaging and therapeutic applications.

This thesis investigates the influence of quantum dot positioning, relative to the gold nanoparticle, on the analytical performance of these assays. Stoichiometric assembly of the quantum dot-gold nanoparticle complexes required accurate knowledge of the quantum dot concentration. A technique was developed to quantify the molar concentration of nanoparticles in solution. This technique relied on the quantitative analysis of the statistical distribution of nanoparticle-ligand conjugates that form when nanoparticles are functionalized with small equivalents of ligand. The technique was successfully validated using gold nanoparticles, and was subsequently applied to determine the concentration of the alloy quantum dots. The precise positioning of the quantum dots relative to the gold nanoparticles required monovalent conjugates of the quantum dots. These were prepared using a magnetic bead based technique where positively charged magnetic beads were used to capture and selectively isolate the monovalent quantum dot-DNA conjugates. Successful isolation of monovalent conjugates was confirmed using single molecule fluorescence spectroscopy and the monovalent conjugates were found to retain their activity as determined using quantitative hybridization assays. Optimizing the quantum dot-gold nanoparticle assay required the investigation of two different configurations of nanoparticle arrangement. The first involved arranging quantum dots around a gold nanoparticle core, while the second involved arranging gold nanoparticles around a quantum dot core. The first configuration was found to have optimal analytical performance, as evaluated using quenching efficiencies and influence of inner filter effect. The ability to arrange multiple quantum dots around a single large gold nanoparticle minimized the negative influence of the inner filter effect, while the high quenching efficiency ensured high responses to target hybridization.
