"Turnover" is defined in different ways, and the term is used in different contexts. It is used rather broadly in the present volume, and intentionally so. The turnover of macromolecules is only one aspect; here "turnover" in dicates the simultaneous and coordinated formation and breakdown of macromolecular species. The complexities of cerebral protein turnover are shown in aseparate chapter dealing with the synthesis ofproteins, in another on breakdown, and in still another on the relationship ofthese two (showing how the two halves of turnover are controlled). The fact that most likely the two halves of protein turnover, synthesis and breakdown, are separated spatially and the mechanisms involved are different further emphasizes the complexity of macromolecular turnover. "Turnover" is used in a different context when the turnover of a cycle is discussed; but he re again a number of complex metabolic reactions have to be interrelated and controlled; some such cycles are discussed briefly in this volume, additional cycles have been discussed with metabolism, and some cycles still await elucidation or discovery.

Available in PDF, EPUB and Kindle. Book excerpt: This volume is concerned with metabolic reactions occurring in the nervous system. Some time ago, it was thought that since most of the intermediary metabolism that can be observed in the brain is not specific to this organ, there is little justification in studying neural metabolism as such. Later it was realized that for an understanding of neural functions, the understanding of metabolism in the brain and its alterations is essential. All aspects of the metabolism of a substrate in brain, or all metabolic reactions of the nervous system, could not be included in this volume; some will be dealt with in other volumes (such as the ones covering metabolic turn over, alterations of metabolism, or pathology). Review of the aspects covered here clearly shows that the study of metabolic reactions in the nervous system is a very active field, producing important results. As in so many areas of research, as we learn more, new aspects become known, new questions emerge, and we see that in solving some problems we open areas with many additional problems to solve. But the accomplishments to date are impressive and indicate further important advances in the future. Brain metabolism is more active, more plastic, and more comprehensive than previously estimated. It is an essential part of brain function, and with its alteration, brain function will be altered. This shows the importance of more knowledge in this area. It is hoped that this volume will be of assistance in such further studies.

The book discusses the molecular structure and morphology of the adult nervous tissue; the chemical composition and cytochemical localization of adult nervous tissue; and the permeability and blood-brain barrier. The text also describes topics on electrolytes and nervous conduction; the metabolism of isolated nerve and ganglion; and the metabolism of the brain in vivo. The metabolism of brain tissue preparations in vitro; energy metabolism and coenzymes in relation to the nervous system; and lipid and fatty acid metabolism are also considered. The book further tackles nucleic acid metabolism; protein and amino acid metabolism; and cholinergic and non-cholinergic transmission. The text also discusses other pharmacologically active compounds related to the adult nervous tissue.

Book excerpt: This book is based on the Symposium "Metabolic Regulation and Functional Activity in the Central Nervous System" which was held on September 16 and 17, 1972, at Saint Vincent (Aosta)/Italy, and was sponsored by the Accademia di Medicina di Torino with the scientific cooperation of the Istituto di Farmacologia, Universita di Torino, and the Pharmakologisches Institut der Freien Universitat Berlin. Its purpose was to give a greater number of scientists from different countries an opportunity to report their latest results under a heading concerning general relationships between metabolism and function. We quite deliberately refrained from pursuing the partly heterogenous subjects into details. Thus, the organizers hoped to be able to interest a greater circle of readers for the manifold subjects from various fields dealing with the investigation of metabolic processes in the central nervous system. The discussion remarks to the lectures could not be considered for printing. The Accademia di Medicina di Torino and all others concerned do not only thank Dr. R. Di Carlo and other members of the Istituto di Farmacologia, Universita di Torino, for the excellent work they performed in preparing this Symposium, but also the Amministrazione Regionale della Valle di Aosta and SIT A V di Saint-Vincent for their generous support and their kind hospitality which made the stay at Saint-Vincent most agreeable for all participants.

Available in PDF, EPUB and Kindle. Book excerpt: The concept of chemical transmission in the central nervous system has taken some time to be generally accepted, but an increasing number of compounds are now being recognized as hav ing a transmitter role in the brain. The acetylcholine system was the first to be discovered in the periphery and its charac teristic features of storage of transmitter in vesicles in the nerve terminal, its electrically-evoked release and rapid extra neuronal breakdown were considered to be necessary criteria for any neurotransmitter candidate. The subsequent elucidation of the noradrenergic system made it apparent that rapid enzymatic breakdown was not essential for a released transmitter, and the possibility of high-affinity re-uptake processes became establ ished as an alternative means of terminating the synaptic actions of a transmitter. With the eventual acceptance of the amino acids as excitat ory or inhibitory transmitters, the requirement for a transmit ter to be present in a low concentration overall (although locally concentrated in specific terminals) also had to be discarded. This necessitated the additional concept of specif ic metabolic pools with different functions being located in different cells or within different regions of the same cell. Some localization of glutamate and aspartate remote from excit able membranes is clearly essential since their overall brain concentrations would be sufficient to maximally depolarize the majority of neurones in the brain. The concept of separate metabolic pools has been supported by stUdies on turnover rate (see Chapter 5).

Book excerpt: Few can deny the paramount importance of the neurosciences, undoubtedly one of the most challenging fields in contemporary science. Recent years have witnessed the awakening of interest in brain research by many dis tinguished investigators from other branches of science, which has made possible the multidisciplinary approach needed for the complex problems of this field. The present book, which deals with one aspect of this research, is the result of the symposium held under the auspices of the New York State Research Institute for Neurochemistry and Drug Addiction in April 1968. It has become clear that brain proteins are involved in all aspects of mental function and dysfunction, and the present volume documents the latest advances in our knowledge (advances made to a large extent by con tributors to this volume). The chapters not only convey some of the enthu siasm and wonderful, cooperative spirit of the many excellent scientists ex ploring the brain, and their wealth of ideas; they also illustrate the many approaches from which cerebral proteins can be studied in a meaningful manner. In some areas even preliminary evidence is worth discussing: e.g., it is an exciting achievement that we can begin to apply the disciplines of bio chemistry to phenomena of learned behavior and information handling.

Book excerpt: The present volume contains all the contributions and general discussion presented at the International Satellite Meeting on Func tion and Metabolism of Phospholipids in Central and Peripheral Nervous Systems held at Cortona, Tuscany, Italy in August 1975. The Satellite Meeting was organized on the frame of the 5th Inter national Congress of the International Society for Neurochemistry (Barcelona, 2-7 september 1975) and was just run before it. The publication of the scientific content of this volume has been made possible by the collaboration of the speakers, the discussants, the Meeting Chairman, the section chairmen and of all the scientists who have taken part at the Symposium and who deeply and actively discussed the lectures and the contributions to the General Discus sion which were delivered. In order to obtain rapid publication of the volume, however, the single discussions for each delivered contribution will not be reported here. The general subject of membrane structure, of the turnover of its lipid components in CNS and PNS, their functional implica tions and pharmacological actions, was explored in details from the stand-points of the various contributors in biophysics, biochem istry, physiology, cytology, pharmacology and pathology. The whole Symposium was effiCiently introduced and closed by Dr. W. Stoffel. The meeting has been thought to have been very successful.

Book excerpt: This book systemically describes the mechanisms underlying the neural regulation of metabolism. Metabolic diseases, including obesity and its associated conditions, currently affect more than 500 million people worldwide. Recent research has shown that the neural regulation of metabolism is a central mechanism that controls metabolic status physiologically and pathophysiologically. The book first introduces the latest studies on the neural and cellular mechanisms of hypothalamic neurons, hypothalamic glial cells, neural circuitries, cellular signaling pathways, and synaptic plasticity in the control of appetite, body weight, feeding-related behaviors and metabolic disorders. It then summarizes the humoral mechanisms by which critical adipocyte-derived hormones and lipoprotein lipase regulate lipid and glucose metabolism, and examines the role of the hypothalamus-sympathetic nerve, a critical nerve pathway from CNS to peripheral nervous system (PNS), in the regulation of metabolism in multiple tissues/organs. Furthermore, the book discusses the functions of adipose tissue in energy metabolism. Lastly, it explores dietary interventions to treat neural diseases and some of the emerging technologies used to study the neural regulation of metabolism. Presenting cutting-edge developments in the neural regulation of metabolism, the book is a valuable reference resource for graduate students and researchers in the field of neuroscience and metabolism.

Book excerpt: The first symposium on metabolic compartmentation in brain was held at the Rockefeller Foundation, Bellagio, Italy, July 11-16, 1971; the proceedings CR. Balazs and J. E. Cremer, editors, MacMillan) appeared in 1973. At the conclusion of the first symposium it was decided to assess in 2-3 years the progress in this rapidly developing area. This volume represents the proceedings of an Advanced Study Institute, made possible by a grant from the North Atlantic Treaty Organization, Scientific Affairs Division. Additional generous support by the Wellcome Trust allowed attendance by participants from non-NATO countries. S. Ber! D. D. Clarke D. Schneider xi Introduction The term metabolic compartmentation, as related to the chemistry of the brain, appeared in print for the first time about 15 years ago. The concept was developed in the laboratory of Dr. Heinrich Waelsch as a result of studies related to the metabolism of glutamic acid and glutamine. It was welcomed by a number of neurochemists who felt that the concept had validity and would help explain metabolic phenomena that were otherwise quite puzzling. The concept gradually achieved general acceptance, and by 1971 the amount of information that could profit by being examined from the point of view of metabolic compartmentation had increased sufficiently to warrant a symposium on the subject ("Metabolic Compartmentation in the Brain," Balazs and Cremer, eds. , MacMillan, 1973). Almost all the participants at that initial symposium were present at the second, on which this volume is based.
