Book excerpt: The book provides a reference for years to come, written by world-renowned expert investigators studying sex differences, the role of sex hormones, the systems biology of sex, and the genetic contribution of sex chromosomes to metabolic homeostasis and diseases. In this volume, leaders of the pharmaceutical industry present their views on sex-specific drug discovery. Many of the authors presented at the Keystone Symposium on “Sex and gender factors affecting metabolic homeostasis, diabetes and obesity” to be held in March 2017 in Lake Tahoe, CA. This book will generate new knowledge and ideas on the importance of gender biology and medicine from a molecular standpoint to the population level and to provide the methods to study them. It is intended to be a catalyst leading to gender-specific treatments of metabolic diseases. There are fundamental aspects of metabolic homeostasis that are regulated differently in males and females, and influence both the development of diabetes and obesity and the response to pharmacological intervention. Still, most preclinical researchers avoid studying female rodents due to the added complexity of research plans. The consequence is a generation of data that risks being relevant to only half of the population. This is a timely moment to publish a book on sex differences in diseases as NIH leadership has asked scientists to consider sex as a biological variable in preclinical research, to ensure that women get the same benefit of medical research as men.

Book excerpt: Brain function is under metabolic control, which in turn determines the equilibrium of homeostatic systems that affect neuronal and glial networks on the molecular, cellular, and systems levels. The collection of articles ranges from molecules and mechanisms involved in regulating homeostasis and neuronal excitability to therapeutic mechanisms tailored to restore homeostatic function. It also features neurological diseases and novel treatment approaches that are based on metabolic and homeostatic interventions. Together, the collection of articles outlines novel strategies to restore brain function in neurology and highlights limitations of conventional pharmacological approaches. We suggest that restoration of molecular and biochemical networks could lead to a new era of therapeutic opportunities.

A short account of the history of the effort led to the Phosphate Workshops is appro priate and can be of interest to the reader. The idea for Phosphate Workshops was born in the early days of November, 1974. One of us (S. G. M. ) suggested the thought to a group of scientists gathered for a luncheon in one of the attrac tive small restaurants in Weisbaden, Germany. The purpose of the workshop was to bring together interested scientists to discuss the newer developments and the recent advances in the field of phosphate metabolism and the other related minerals. An Organizing Committee made of Shaul G. Massry (USA), Louis V. Avioli (USA), Philippe Bordier (France), Herbert Fleisch (Switzerland), and Eduardo Slatopolsky (USA) was formed. The First Workshop was held in Paris during June 5-6, 1975 and was hosted by Dr. Philippe Bordier. Its proceeding was already published. The Second Workshop took place in Heidelberg during June 28-30, 1976 and was hosted by Dr. Eberhard Ritz. Both of these workshops were extremely successful scientific endeavors, and the need for them was demonstrated by the great interest they generated among the scientific community. The Or ganizing Committee, therefore, decided to continue with the tradi tion to hold additional Workshops annually or every other year.

Available in PDF, EPUB and Kindle. Book excerpt: The obesity epidemic has generated immense interest in recent years due to the wide-ranging and significant adverse health and economic consequences that surround the problem. Much attention has been focused on behaviors that lead to obesity, in particular to over consumption of energy-dense food and to sedentary lifestyle. However, obesity is an extremely complex condition with poorly defined pathogenesis. Thanks to greatly enhanced research in the area, the discovery of pathways in the brain and peripheral organs that mediate energy homeostasis has provided a framework for understanding the biological basis of obesity. Metabolic Basis of Obesity adds an important new dimension to the growing literature on obesity by offering a comprehensive review of specifically how metabolic imbalance culminates in obesity. Developed by a team of expert authors, this important title discusses the principles of energy balance, genetics of body weight regulation, hormones and adipokines, and metabolic pathways in the brain, liver, muscle and fat, to name just several of the areas covered. The book also examines the connection between obesity and diabetes, cardiovascular disease and other complications. Current and future diagnostic and treatment strategies are also reviewed. Comprehensive and timely, Metabolic Basis of Obesity is an essential reference for understanding the burgeoning problem of obesity.

Book excerpt: This book has a dual purpose, to review in depth the control of fuel homeostasis in the brain and the role of the nervous system in the control of fuel deposition in the body. From the methodological point of view the emphasis is on the application of advanced technologies to assess fuel transport and brain metabolism, the role of peptides in the neuroendocrine system and the response of the brain to hypoglycemia. These technologies include positron emmission tomography, nuclear magnetic resonance, immunocytochemistry, molecular biology, autoradiography. To study fuel homeostasis in the body advanced tracer methods that include modelling are set out. From the pathophysiological point of view the emphasis is on abnormalities in stress, brain metabolism in diabetes, eating and degenerative disorders. This book contains contributions from endocrinologists, physiologists, neurologists, psychoneuroendocrinOlogists, biophysicists, biochemists and experts in nutrition. This authorship represents a unique diversity of researchers who, for the first time, cover comprehensively the interaction between the nervous system and fuel homeostasis, both in health and disease. We hope this book will be an important source of information for both researchers and practicing clinicians. Mladen Vranic Suad Efendic Charles Hollenberg v ACKNOWLEDGEMENTS The Symposium from which this volume arose (University of Toronto, June 27-28, 1990) was the first Toronto-Stockholm symposium on Perspectives in Diabetes Research. Book excerpt: Functional Metabolism of Cells is the first comprehensive survey of metabolism, offering an in-depth examination of metabolism and regulation of carbohydrates, lipids, and amino acids. It provides a basic background on metabolic regulation and adaptation as well as the chemical logic of metabolism, and covers the interrelationship of metabolism to life processes of the whole organism. The book lays out a structured approach to the metabolic basis of disease, including discussion of the normal pathways of metabolism, altered pathways leading to disease, and use of molecular genetics in diagnosis and treatment of disease. It also takes a unique comparative approach in which human metabolism is a reference for metabolism in microorganisms and plant design, and presents novel coverage of development and aging, and human health and animal adaptation. The final chapter reviews the past and future promise of new genetic approaches to treatment and bioinformatics. This, the most exhaustive treatment of metabolism currently available, is a useful text for advanced undergraduates and graduates in biochemistry, cell/molecular biology, and biomedicine, as well as biochemistry instructors and investigators in related fields.

At the cellular level such changes are distinct and multidimensional: DNA replication ceases, cells stop dividing, they become senescent and eventually die. DNA metabolism and chromosomal maintenance, together with protein metabolism are critical in the aging process. The focus of this book is on the role of protein metabolism and homeostasis in aging. An overview is provided of the current knowledge in the area, including protein synthesis, accuracy and repair, post-translational modifications, degradation and turnover, and how they define and influence aging. The chapters mainly focus on well-characterised factors and pathways, but new areas are also presented, where associations with aging are just being elucidated by current experimental data. Protein turnover, the balance between protein synthesis and protein degradation are carefully maintained in healthy cells. Chapters 1 and 2 illustrate that aging cells are characterised by alterations in the rate, level and accuracy of protein synthesis compared to young ones, and that mRNA translation, essential for cell growth and survival, is controlled at multiple levels. The theory that growth and somatic maintenance are believed to be antagonistic processes is described in Chapter 3: inhibition of protein synthesis results in decreased rates of growth and development, but also confers an extension of lifespan, as shown for example by the effects of dietary restriction in various models organisms.

Book excerpt: This book systemically describes the mechanisms underlying the neural regulation of metabolism. Metabolic diseases, including obesity and its associated conditions, currently affect more than 500 million people worldwide. Recent research has shown that the neural regulation of metabolism is a central mechanism that controls metabolic status physiologically and pathophysiologically. The book first introduces the latest studies on the neural and cellular mechanisms of hypothalamic neurons, hypothalamic glial cells, neural circuitries, cellular signaling pathways, and synaptic plasticity in the control of appetite, body weight, feeding-related behaviors and metabolic disorders. It then summarizes the humoral mechanisms by which critical adipocyte-derived hormones and lipoprotein lipase regulate lipid and glucose metabolism, and examines the role of the hypothalamus-sympathetic nerve, a critical nerve pathway from CNS to peripheral nervous system (PNS), in the regulation of metabolism in multiple tissues/organs. Furthermore, the book discusses the functions of adipose tissue in energy metabolism. Lastly, it explores dietary interventions to treat neural diseases and some of the emerging technologies used to study the neural regulation of metabolism. Presenting cutting-edge developments in the neural regulation of metabolism, the book is a valuable reference resource for graduate students and researchers in the field of neuroscience and metabolism.

Available in PDF, EPUB and Kindle. Book excerpt: This book offers a broad overview of the concepts and research findings in immunometabolism. The immune system is made up of numerous different cell types, pathways, and components that must be able to respond rapidly to a pathogen or cancer, but must also remain quiescent in the absence of challenges. Immune cells rely on metabolic pathways to adapt to changing environments and stimuli. Additionally, these cells can be modified in function or fate by fluctuations in available nutrients. The chapters in this book describe ways in which immune cells utilize and are regulated by metabolic pathways. Topics include how immune-cell metabolism shapes immune homeostasis, and how dysregulation of these pathways can lead to immune disorders. In different contexts, such as a tumor microenvironment, immune-cell function and identity may be modified not only by cytokines and checkpoint molecules, but also by nutrient availability and other metabolic stimuli. Transcriptional reprogramming confers many of the changes in immune cell metabolism that are seen when a T-cell, for example, undergoes activation or functional adaptation to different environments. Lastly, immune cells can destructively or protectively participate in human metabolic homeostasis or disorders. This book summarizes immune-metabolism from a variety of different perspectives, including the ways in which metabolic cues, pathways, and requirements of immune cells change in conditions of homeostasis and activation. The exploration of the significance of metabolic checkpoints and other cues, particularly in the context of cancer and immune disorders, may form the foundation for the development of therapeutics.
