Book excerpt: The pathways stimulated by the oncogenes HER2/Neu/ErbB2, Wnt-l, and H-ras are aberrantly activated in many human cancers, including breast cancer, and have been shown to play a role in carcinogenesis. In order to better investigate the role these genes play in breast cancer, we generated mouse models that inducibly express activated forms of these oncogenes in the mammary gland. This system allows us to activate these oncogenic pathways during specific windows of mammary gland development, as well as to remove the initiating oncogenic stimulus after the development of a tumor or during later stages of tumor progression. As such, we anticipate that the study of these models will provide valuable insights into the mechanisms by which activation of these pathways contributes to breast cancer development or progression, and will ultimately aid in the generation of therapeutic strategies that specifically target these pathways. We have found that expression of Neu in the mammary glands of transgenic mice results in the development of both invasive adenocarcinomas and pulmonary metastases that remain dependent upon Neu expression for maintenance of the transformed state. We have also determined that both Wnt-l and c-Myc synergize with Neu in mammary tumorigenesis.

Book excerpt: The purpose of the research funded by this award is to identify molecular interactions between the c-MYC proto-oncogene and normal mammary developmental events. Ultimately, our goal is to identify how these interactions affect the process of mammary carcinogenesis. To this end, we seek to identify mammary development stage specific effects of aberrant c-MYC activation, to identify molecular mechanisms accounting for these effects, and to determine what role these interactions plays in mediating the biological effects of aberrant c-MYC expression within the mammary epithelium. This work has led to the finding that aberrant c-MYC activation for discrete periods during early and late pregnancy has dramatically different outcomes. Specifically, c-MYC activation during late pregnancy results in premature Stat5 activation followed by precocious lactation, whereas c-MYC activation during early pregnancy results in neither of these dramatic phenotypes. We are now in the process of determining what role c-MYC-induced activation of the Stat5 mammary developmental signaling pathway plays in mediating the effects of aberrant activation of c-proliferation, differentiation, apoptosis, and tumorigenesis.

Book excerpt: C-Myc oncogene has been reported to be amplified in 25-30% and overexpressed in more than 70% of human breast cancers and has been demonstrated to be involved in signaling cell proliferation and apoptosis. The Bcl-xL protein, a member of the Bcl-2 apoptosis- modulatory family, is known to block apoptotic cell death under a wide variety of conditions and has been shown to be overexpressed in some human breast cancers and breast cancer cell lines. Evidence from a c-Myc/TGFalpha bitransgenic mouse model suggests that escape from c-Myc-induced apoptosis may be necessary for continued cell cycle progression and neoplastic development. The focus of this study is to determine if Bcl-xL expression cooperates with c-Myc overexpression in mammary tumorigenesis in vi and in vitro by blocking c-Myc-induced apoptosis but not c-Myc-mediated cell cycle progression and cell proliferation. Transgenic c-Myc mice will be mated with transgenic Bcl-xL mice to produce bitransgenic offspring which will be observed to determine whether Bcl-xL expression is sufficient to synergize with c-Myc and enhance mammary tumor incidence and accelerate tumor onset (latency). The utilization of this model system will aid in the dissection of the molecular mechanisms behind the development of breast cancer.

The term multidrug resistance is also apt – in that these patients die after having undergone multiple rounds of different and structurally unrelated cancer therapies. However, for some, the concept of m- tidrug resistance is a worn out idea, stemming from disappointment with the drug resistancereversalstrategiesthatwerecarriedoutinthe1990susingpumpinhibitors to block drug resistance mediated by P-glycoprotein, product of the MDR-1 gene. However, if one takes the larger de?nition – multidrug resistance as simultaneous resistance to multiple structurally unrelated anticancer therapies – its existence c- not be denied. The purpose of this book is to explore new concepts related to drug resistance in cancer, including resistance to the new molecularly targeted agents. Perhaps new terminology is needed for resistance that occurs following therapy with the targeted agents: Novel Targeted Agent Resistance (NTR). Alternatively, we can return to the original de?nition of multidrug resistance as simply the res- tance to multipleagents that occurs in the course of normalcancer progression.This resistance is likely to be mediated by many factors.
